    General relationships of mammalian orders and evolutionary development of primates inferred from best-fit α-globin phylogenies

    A best-fit α-globin phylogeny was identified by means of a global minimization approach from among the topologies generated by a parsimony strategy. Zip parsimony method was used to derive a set of near-parsimonious trees. For each of these topologies, a difference matrix was computed; and the topology with the best goodness of fit with the original matrix was retained as the best tree. Based on this phylogenetic scheme inter-relationships among eutherian orders and the evolutionary development of primates has been discussed

    Type Iax SNe as a few-parameter family

    We present direct spectroscopic modeling of five Type Iax supernovae (SNe) with the one dimensional Monte Carlo radiative transfer code TARDIS. The abundance tomography technique is used to map the chemical structure and physical properties of the SN atmosphere. Through via fitting of multiple spectral epochs with self-consistent ejecta models, we can then constrain the location of some elements within the ejecta. The synthetic spectra of the best-fit models are able to reproduce the flux continuum and the main absorption features in the whole sample. We find that the mass fractions of IGEs and IMEs show a decreasing trend toward the outer regions of the atmospheres using density profiles similar to those of deflagration models in the literature. Oxygen is the only element, which could be dominant at higher velocities. The stratified abundance structure contradicts the well-mixed chemical profiles predicted by pure deflagration models. Based on the derived densities and abundances, a template model atmosphere is created for the SN Iax class and compared to the observed spectra. Free parameters are the scaling of the density profile, the velocity shift of the abundance template, and the peak luminosity. The results of this test support the idea that all SNe Iax can be described by a similar internal structure, which argues for a common origin of this class of explosions.Comment: 21 pages, 7 tables, 16 figures, accepted by MNRA

    Dalam upaya meningkatkan  derajat  kesehatan  di  sekolah, penerapan perilaku hidup bersih dan sehat di sekolah sangatlah penting. Selain  manfaat  penerapan perilaku hidup bersih dan sehat dalam kebiasaan sehari-hari dapat mencegah kerentanan timbulnya penyakit, tentunya juga bermanfaat dalam meningkatkan kualitas pembelajaran di kelas karena fisik yang sehat pada pelajar. Tujuan penelitian ini Untuk mengetahui gambaran penyuluhan kesehatan terhadap pengetahuan tentang Perilaku Hidup Bersih dan Sehat pada pelajar kelas X di SMA Negeri 1 Sinonsayang. Penelitian ini merupakan penelitian deskriptif yang dilaksanakan di SMA Negeri 1 Sinonsayang minahasa selatan pada Oktober 2021 - Januari 2022 dengan total populasi 51 siswa kelas X. Hasil penelitian menunjukan angka pengetahuan yang awalnya (88,24%) meningkat menjadi (100%) setelah dilakukannya penyuluhan.  Kata Kunci : PHBS, Penyuluhan Kesehatan, SMA Negeri 1 Sinonsayang ABSTRACTIn an effort to improve the health status in schools, the application of clean and healthy living behavior in schools is very important. The purpose of this study was to determine the description of health education on knowledge about Clean and Healthy Life Behavior in class X students at SMA Negeri 1 Sinonsayang. This research is a descriptive study that was conducted at SMA Negeri 1 Sinonsayang, South Minahasa in October 2021 - January 2022 with a total population of 51 students in class X.     Adding a quadrivalent human papillomavirus vaccine to the UK cervical cancer screening programme: A cost-effectiveness analysis

    <p>Abstract</p> <p>Background</p> <p>We assessed the cost-effectiveness of adding a quadrivalent (6/11/16/18) human papillomavirus (HPV) vaccine to the current screening programme in the UK compared to screening alone.</p> <p>Methods</p> <p>A Markov model of the natural history of HPV infection incorporating screening and vaccination was developed. A vaccine that prevents 98% of HPV 6, 11, 16 and 18-associated disease, with a lifetime duration and 85% coverage, in conjunction with current screening was considered.</p> <p>Results</p> <p>Vaccination with screening, compared to screening alone, was associated with an incremental cost-effectiveness ratio of ÂŁ21,059 per quality adjusted life year (QALY) and ÂŁ34,687 per life year saved (LYS). More than 400 cases of cervical cancer, 6700 cases of cervical intraepithelial neoplasia and 4750 cases of genital warts could be avoided per 100,000 vaccinated girls. Results were sensitive to assumptions about the need for a booster, the duration of vaccine efficacy and discount rate.</p> <p>Conclusion</p> <p>These analyses suggest that adding a quadrivalent HPV vaccine to current screening in the UK could be a cost-effective method for further reducing the burden of cervical cancer.</p

    Strategies for the introduction of human papillomavirus vaccination: modelling the optimum age- and sex-specific pattern of vaccination in Finland

    Phase III trials have demonstrated the efficacy of human papillomavirus (HPV) vaccines in preventing transient and persistent high-risk (hr) HPV infection and precancerous lesions. A mathematical model of HPV type 16 infection and progression to cervical cancer, parameterised to represent the infection in Finland, was used to explore the optimal age at vaccination and pattern of vaccine introduction. In the long term, the annual proportion of cervical cancer cases prevented is much higher when early adolescents are targeted. Vaccinating against hr HPV generates greater long-term benefits if vaccine is delivered before the age at first sexual intercourse. However, vaccinating 12 year olds delays the predicted decrease in cervical cancer, compared to vaccinating older adolescents or young adults. Vaccinating males as well as females has more impact on the proportion of cases prevented when vaccinating at younger ages. Implementing catch-up vaccination at the start of a vaccination programme would increase the speed with which a decrease in HPV and cervical cancer incidence is observed

    Implementation of HIV Self-Testing to Reach Men in Rural uMkhanyakude, KwaZulu-Natal, South Africa. a DO-ART Trial Sub Study

    Background: KwaZulu–Natal, South Africa has one of the highest HIV prevalence rates globally. Persons <35 years and men have lower rates of HIV testing. HIV self-testing (HIVST) may overcome many barriers of facility-based HIV testing in order to identify HIV positive young persons and men and link them to care. We investigated whether HIVST distribution was a feasible approach to reach men and assessed the proportion of participants who reported their HIVST results, tested positive and linked to care. / Methods: Teams comprised of a nurse, clinic research assistant, and recruiters distributed HIVST kits in rural uMkhanyakude, KwaZulu-Natal from August—November 2018 with a focus on testing men. Workplaces (farms), social venues, taxi ranks, and homesteads were used as HIVST kit distribution points following community sensitisation through community advisory boards and community leaders. HIVST kits, demonstration of use, and small incentives to report testing outcomes were provided. The Department of Health provided confirmatory testing and HIV care at clinics. / Results: Over 11 weeks in late 2018, we distributed 2,634 HIVST kits of which 2,113 (80%) were distributed to persons aged <35 years, 2,591 (98%) to men and 356 (14%) to first time testers. Of the HIVST distributed, 2,107 (80%) reported their results to the study team, and 157 (7%) tested positive. Of persons who tested positive, 107/130 (82%) reported having a confirmatory test of which 102/107 (95%) were positive and initiated on ART. No emergencies or social harms were reported. / Conclusion: Large scale distribution of HIVST kits targeting men in rural KwaZulu-Natal is feasible and highly effective in reaching men, including those who had not previously tested for HIV. While two-thirds of persons who tested HIV positive initiated ART, additional linkage strategies are needed for those who do not link after HIVST. HIVST should be used as a tool to reach men in order to achieve 95% coverage in the UNAIDS testing and care cascade in KwaZulu-Natal

    Comparative secretome analysis of Colletotrichum falcatum identifies a cerato-platanin protein (EPL1) as a potential pathogen-associated molecular pattern (PAMP) inducing systemic resistance in sugarcane

    Colletotrichum falcatum, an intriguing hemibiotrophic fungal pathogen causes the devastating red rot disease of sugarcane. Repeated in vitro subculturing of C. falcatum under dark condition alters morphology and reduces virulence of the culture. Hitherto, no information is available on this phenomenon at molecular level. In this study, the in vitro secretome of C. falcatum cultured under light and dark conditions was analyzed using 2-DE coupled with MALDI TOF/TOF MS. Comparative analysis identified nine differentially abundant proteins. Among them, seven proteins were less abundant in the dark-cultured C. falcatum, wherein only two protein species of a cerato-platanin protein called EPL1 (eliciting plant response-like protein) were found to be highly abundant. Transcriptional expression of candidate high abundant proteins were profiled during host-pathogen interaction using qRT-PCR. Comprehensively, this comparative secretome analysis identified five putative effectors, two pathogenicity-related proteins and one pathogen-associated molecular pattern (PAMP) of C. falcatum. Functional characterization of three distinct domains of the PAMP (EPL1) showed that the major cerato-platanin domain (EPL1 Δ N1–92) is exclusively essential for inducing defense and hypersensitive response (HR) in sugarcane and tobacco, respectively. Further, priming with EPL1 Δ N1–92 protein induced systemic resistance and significantly suppressed the red rot disease severity in sugarcane. Biological significance Being the first secretomic investigation of C. falcatum, this study has identified five potential effectors, two pathogenicity-related proteins and a PAMP. Although many reports have highlighted the influence of light on pathogenicity, this study has established a direct link between light and expression of effectors, for the first time. This study has presented the influence of a novel N-terminal domain of EPL1 in physical and biological properties and established the functional role of major cerato-platanin domain of EPL1 as a potential elicitor inducing systemic resistance in sugarcane. Comprehensively, the study has identified proteins that putatively contribute to virulence of C. falcatum and for the first time, demonstrated the potential role of EPL1 in inducing PAMP-triggered immunity (PTI) in sugarcane
