A gonadotropin-releasing hormon (GnRH) és az FSH szekréció szabályozásának vizsgálata = Investigation on the regulation of gonadotropin-releasing hormone (GnRH) and FSH secretion
A lamprey gonadotropin-releasing hormon (GnRH)-III különböző analógjait hoztuk létre, és vizsgáltuk azok FSH/LH- felszabadító, valamint tumorgátló hatásukat humán emlő carcinoma sejteken. Az Arg8-GnRH-III specifikusan kötődik az emlős GnRH receptorhz, és 3-11-szer nagyobb gonadotropin-releasing hatáserősséget mutat mint a GnRH-III, az emlős GnRH-hoz viszonyított erőssége azonban 1% alatt van. A tumorgátló aktivitás fokozása céljából létrehozott dimer analógok antitumor aktivitása és enzimekkel szembeni stabilitása fokozódott, míg endokrín hatáserőssége lényegesen csökkent. A GnRH-III copolymer konjugátumának endokrín aktivitása közel zéró, ugyanakkor tumorgátló hatáserőssége humán emlőcarcinoma sejteken 2-3-szoros. Adataink arra utalnak, hogy a GnRH analógok tumorgátló szelektivitása fokozható. A klinikumban is használatos GnRH antagonista Cetrorelix alakalmazásával igazoltuk, hogy az analóg alkalmas a hypophysis-gonád tengely részleges gátlására, és ezáltal az endometriosis és a prostata hyperplasia kezelésére is. Kimutattuk, hogy a GnRH-gonadotropin rendszer részt vesz a hypophyseális inhibin/aktivin és follisztatin génexpresszió szabályozásában. Fő szabályozó a GnRH, amely stimulálja a follisztatin génexpressziót, az inhibin-specifikus - lánc és a 'B alegység génexpressziót pedig gátolja. A 'A lánc mRNS expresszója jelentősen kisebb, mint a 'B láncé, ími arra utal, hogy a hypophysisben főként az inhibin/aktivin B forma fordul elő. | We developed various analogs of lamprey gonadotropin-releasing hormon (GnRH)-III and investigated their LH/FSH-releasing potency and antiproliferative activity on human breast cancer cells. Arg8-GnRH-III specifically binds to the mammalian (m) GnRH receptor and has 3-11 fold gonadotropin-releasing potency compared to GnRH-III but less than 1% potency compared to mGnRH. Dimer analogs, which were developed to improve the anticancer potency, showed higher stability against degrading enzymes, stronger anticancer effects, and weaker endocrine potency. A copolymer conjugate of GnRH-III had 2-3 fold anticancer activity while loosing endocrine potency. Our data show that the anticancer selectivity of the GnRH analogs can be improved. Investigating the GnRH antagonist Cetrorelix, which is used in the clinical practice, we confirmed that Cetrorelix at low doses only a partially inhibit the pituitary-gonad axis and might be indicated for treatment of endometriosis and benign prostatic hyperplasia. We demonstrated that the GnRH-gonadotropin system plays a role in the regulation of gene expression of the pituitary inhibin/activin and follistatin. GnRH is the main regulator, which stimulates follistatin gene expression but inhibits the inhibin-specific - subunit and the ?B gene expression. The expression of the ?A subunit is significantly lower than that of the 'B, indicating that the B forms of inhibin/activin are more significant in the pituitary than the A forms
Az óragének és a környezeti fényviszonyok szerepe az N-acetiltranszferáz génexpressziójának szabályozásában csirke retinában = The role of clock-genes and environmental lighting conditions in the regulation of N-acetyltransferase gene expression in the chicken retina
Csirke retina és tobozmirigy sejtekben a melatonin (MT) szintézis kulcsenzimének, az arilalkilamin N-acetiltranszferáznak génje (cAanat) co-expresszálódik az óragénekkel. Emellett a dopamin (DA) a fő katekolamin a retinában, mely neuroregulatorként hat. Tanulmányoztuk: 1. a cBmal1 és cAanat mRNS expressziók dinamikáját retinális/pineális sejtekben in vivo és in vitro RT-PCR-ral. A cBmal1 mRNS csúcsa a fényes fázisban volt Zeitgeber idő (ZT) 8 és 10 között, megelőzve az a cAanat expresszió éjszakai emelkedését ZT 16-17-nál. A retinális/pineális cBmal1 és cAanat mRNS ritmusainak fázisai arra utalnak, hogy kapcsolat van az expressziók között; 2. locked nucleinsav tartalmú cBmal1 antisense oligonukleotidok (AON) hatását a cAanat transzkriptumokra és az MT termelésre pinealocita kultúrában, melyet szuperfúziós rendszerben transzfektáltunk. Az AON, mely vagy az RNáz H-t aktiválta, vagy a transzlációs masinériához való kötődést blokkolta szignifikánsan csökkentette a cAanat transzkripcióját és az MT szekrécióját jelezvén a cBmal1 kulcsszerepét az indol metabolizmus szabályozásában; 3. két fő DA-receptor család (D1 és D2) szerepét a circadián retinális MT ritmus szabályozásában eye-cup modellen. A D2 agonista Quinpirol szignifikánsan csökkentette az MT szekrécióját sötét fázisban, míg a D1 agonista SKF hatástalan volt, amely arra utal, hogy a DA gátló hatást fejt ki a retinális MT szekrécióra (reciprok negativ feed-back), melyet elsősorban a D2 receptorok mediálnak. | In chicken retinal and pineal cells gene of arylalkylamine N-acetyltransferase (cAanat), the key enzyme of melatonin (MT) synthesis is co-expressed with clock genes. In addition, dopamine (DA) is the major catecholamine in the retina serving as a neuroregulator. We studied: 1. the temporal profile of cBmal1 and cAanat mRNA expressions in retinal/pineal cells in vivo and in vitro using RT-PCR. cBmal1 mRNA had a peak in the light phase between Zeitgeber time (ZT) 8 and 10, preceding the nocturnal increase in cAanat expression at ZT 16 to 17. The phase of the rhythms of retinal/pineal cBmal1 and cAanat mRNAs suggests a link between their expressions; 2. the effect of cBmal1 antisense oligonucleotides (AONs) containing locked nucleic acid on cAanat transcripts and MT production in cultured pinealocytes transfected in superfusion system. These AONs synthesized for activating RNase H or blocking the binding of the translation machinery were able to reduce significantly cAanat transcription and MT secretion indicating the key role of cBmal1 in the regulation of indole metabolism; 3. the role of the two major DA-receptor families (D1 and D2) in the regulation of circadian retinal MT rhythm on an eye-cup model. Quinpirole (D2 agonist) reduced significantly MT secretion in dark phase, whereas SKF (D1 agonist) was ineffective suggesting that DA exerts an inhibitory effect on retinal MT secretion (reciprocal negative feed-back), which is mediated primarily via D2 receptors
Perifériás ideg epineuralis metilénkékfestése kadáverkézen
Bevezetés: A parciális aponeurectomia a Dupuytren-kontraktúra kezelésére leggyakrabban végzett műtéti eljárás.
A betegségben kialakuló patológiás szövet megváltoztathatja a digitális ideg anatómiai elhelyezkedését, ami megne-
hezíti az ideg műtét közbeni lokalizációját és dissectióját, és növeli a iatrogén idegsérülés kockázatát. Intraoperatív
idegfestési eljárással az ideg lokalizációja megkönnyíthető lenne, ezáltal a iatrogén idegsérülés kockázata is csökkenne. Állatkísérleteinkben korábban igazoltuk, hogy metilénkékoldattal a perifériás ideg in vivo megfesthető az ideg struktúrájának és funkciójának károsítása nélkül.
Célkitűzés: A patkány nervus ischiadicus modellen már sikeresen alkalmazott metilénkékoldattal végzett idegfestési
eljárás hatékonyságának vizsgálata humán kadáver digitális idegen.
Módszer: Vizsgálatunk első fázisában formalinnal fixált kézen négy digitális ideg epineuralis festését végeztük el 40 μl 1 : 80-as hígítású metilénkékoldattal. A második vizsgálatban fixáción át nem esett kadáverkézen hat digitális idegfestését végeztük. A megfesthető idegszakasz hosszának növelésére két ideg festéséhez 200 μl metilénkékoldatot használtunk.
Eredmények: Az epineuralis idegjelölés formalinfixált idegeken nem működött ideálisan. Friss, formalinos fixáláson át nem esett humán kadáver digitális idegen az idegfestési eljárást sikeresen alkalmaztuk, a megfestett idegszakasz tekintetében állatkísérletes eredményeinket reprodukálni tudtuk. 40 μl 1 : 80-as hígítású metilénkékoldattal átlagosan 13 mm-es, míg 200 μl oldat használatával 18 mm-es idegszakaszt sikerült megfesteni.
Következtetés : Formalinnal fixált digitális ideg festése a fixáció következtében fellépő szöveti zsugorodás miatt korlátozottan lehetséges. Formalinos fixáláson át nem esett digitális idegek esetén a megtartott anatómiai viszonyok mellett 18 mm-es idegszakasz megfesthető. További vizsgálatokat tervezünk Dupuytren-szövettel és hegszövettel körülvett digitális idegen, a technika kézsebészeti műtétek során történő alkalmazhatóságának megítélésére

Antagonists of growth hormone-releasing hormone arrest the growth of MDA-MB-468 estrogen-independent human breast cancers in nude mice
Since antagonists of growth hormone-releasing hormone (GH-RH) inhibit proliferation of various tumors, in this study we investigated the effects of GH-RH antagonists MZ-5-156 or JV-1-36 on growth of estrogen-independent MDA-MB-468 human breast cancers xenografted into nude mice. Both GH-RH antagonists administered at a dose of 20 μg/day induced regression of some and growth-arrest of other tumors, while control tumors continued to grow. After 5 weeks of therapy with MZ-5-156 or JV-1-36, final volume and weight of MDA-MB-468 tumors were significantly decreased (all p values <0.001) and serum IGF-I levels as well as tumor IGF-I mRNA expression were reduced as compared with controls. High affinity binding sites for IGF-I were detected by the ligand binding method. Gene expression of human IGF-I receptors, as measured by the RT-PCR, was not significantly different in control and treated MDA-MB-468 tumors. In cell culture, IGF-I did not stimulate, GH-RH slightly stimulated, while MZ-5-156 and JV-1-36 inhibited proliferation of MDA-MB-468 cells known to possess defective insulin and IGF-I receptor signaling. The expression of mRNA for human GH-RH was found in five of 8 tumors treated with GH-RH antagonists, and in one of the five control tumors. These results suggest that GH-RH antagonists inhibit MDA-MB-468 breast cancers possibly through mechanisms involving interference with locally produced GH-RH
