    Expression of flagellin FLjB derived from Salmonella enterica serovar typhimurium in Escherichia coli BL21

    Flagellin FljB composes flagellar antigen (H:1,2) of S. Typhimurium. This kind of antigen increases immunogenicity of any conjugated antigen upon administration. Thus, it is supposed to have an enormous potentiality for vaccine development against bacterial infections and cancer diseases. fljB gene (1515 nucleotides) coding for mature FljB was amplified by PCR from genomic DNA of S. Typhimurium and inserted into pET32a(+) for expression in E. coli BL21. The protein FljB was well expressed under the fusion form with Trx, S-tag at N terminal and hexa-histidine at C terminal, thus the recombinant protein was abbreviated to TrxFljB. Study on the impact of temperature on the gene expression showed that TrxFljB was synthesized at lower level at 37oC comparing to the levels at 22oC and 25oC. 13% of the protein synthesized at 37oC was inclusion body. Lower temperatures used during induction phase increased the solubility of the recombinant protein. About 97% of TrxFljB synthesized at 25oC was soluble. IPTG concentration had a strong effect on the growth of freshly transformed cells but did not affect on the growth of stored and re-cultivated cells. The increase of IPTG concentration resulted in the decrease of the growth of freshly transformed cells and the TrxFljB productivity. However, 0.05 mM IPTG concentration was found to gain the full TrxFljB expression. TrxFljB productivity declined during storage of cells at 4oC and re-cultivation. At optimal condition, volumetric productivity of TrxFljB was about 300 mg/ l broth.

    Cloning and expression of pigC gene in Escherichia coli

    Prodigiosin (Pg), which is particularly of interest because of anticancer and antimicrobial activities, can be produced through the PigC-catalyzed condensation reaction of 4-methoxy-2, 2’-bipyrrole-5-carboxyaldehyde (MBC) and 2-methyl-3-amylpyrrole (MAP). Therefore, the PigC protein plays an important role in prodigiosin biosynthetic pathway. However, studies related to PigC protein have not been carried out in Vietnam yet. In this work, the pigC gene was cloned and expressed in Escherichia coli DH10B and BL21 (DE3), respectively. Using PCR and universal primers, we amplified a fragment of 3 kb covering entire coding region of the pigC gene from Serratia sp. strain M5. The pigC gene was inserted into pJET1.2 vector, and then transformed into E. coli DH10B. The sequence of a recombinant vector pJET1.2/pigC was evaluated by using whole colony PCR amplification. Sequence alignment results revealed that the obtained pigC gene possesses 71.5% and 75.4% of nucleotide identity in comparison with two strains, Serratia 39006 and Serratia sp. AS9 published in GenBank with their respective accession numbers of AJ833001 and CP002773. The recombinant vector pJET1.2/pigC was used to reamplify pigC, and the acquired amplicon was inserted into pET22b vector at the site of HindIII and XhoI. The clone E. coli BL21 (DE3) containing recombinant vector pET22b/pigC was expressed in the auto-induced medium. The presence of PigC protein in the lysate was identified as a 100 kDa band through Western Blot analysis using anti his-tag antibody. Afterward, the PigC protein was purified by Ni-NTA column, and its expression level was quantified through SDS-PAGE analysis. The results of our study provide a potential material for producing prodigiosin from recombinant protein in Vietnam

    Unravelling the economic impact of climate change in Vietnam's Mekong River Delta and Southeast region

    Vietnam faces a heightened susceptibility to natural disasters stemming from climate change, yet the impact of these disasters varies markedly across its regions. Consequently, it is imperative to scrutinize the climatic factors affecting the regional economy, with a particular dearth of research addressing the nexus between natural threats and the economies of the Mekong River Delta and the Southeast region of Vietnam. The primary objective is to investigate how temperature fluctuations, rising sea levels, storms, and forest fires can detrimentally affect agriculture, forestry, and infrastructure in these regions. The present research uses the secondary data from Vietnam's General Statistics Office and the German Agency for International Cooperation. Employing quantitative analysis, the study endeavours to assess and compare the influence of climate change elements on the economic trajectories of the Mekong River Delta and the Southeast region. The findings reveal that the Mekong River Delta is disproportionately impacted by economic losses attributed to rising sea levels when juxtaposed with other regions in Vietnam. Firstly, the escalating sea levels significantly jeopardize residential and agricultural land in the Mekong River Delta (1). Secondly, while wildfires inflict damage on forestry in both the Mekong River Delta and the Southeast area, their overall impact is deemed negligible (2). Thirdly, the study ascertains that houses in the Mekong River Delta and the Southeast region remain relatively unscathed by storms (3). However, it is noted that specific storms have inflicted substantial damage on both regions (4). In light of these findings, future research should delve deeper into the analysis of storm risks in the Mekong River Delta and the Southeast region. Such insights are crucial for enhancing our understanding of the vulnerabilities and devising effective strategies to mitigate the economic repercussions of climate change in these pivotal areas

    Prevalence of arterial hypertension in Vietnamese seafarers aboard merchant vessels: a cross-sectional study

    Background: Hypertension is one of the leading causes of morbidity and mortality globally. It is a major riskfactor for major cardiovascular events such as stroke, myocardial infarction, heart failure, kidney failure,and blindness. The aim of this research is to assess the prevalence and some factors related to arterialhypertension on Vietnamese seafarers aboard merchant vessels.Materials and methods: Seven hundred eight Vietnamese seafarers working aboard merchant ships wereexamined at the Institute of Marine Medicine before going to sea during the period from January 2022to December 2022. It was a cross-sectional descriptive epidemiological study. The following parameterswere measured: blood pressure, height, weight, waist circumference, buttock circumference to assess theprevalence of hypertension, overweight, and obesity. Seafarers we directly interviewed about workplaceon ships and physical exercise, smoking tobacco, alcohol abuse, and anxiety symptoms to identify severalfactors associated with hypertension.Results: The prevalence of hypertension in seafarers was 32.9%, prehypertension 26.4%, overweight32.4%, obesity 13.3%, abdominal obesity 47.7%. Factors related to hypertension of seafarers included: jobduration at sea > 10 years, odds ratio (OR) = 8.23 (95% confidence interval [CI] 4.34–17.27); non-officers,OR = 2.11 (95% CI 1.45–2.82); engine room crew, OR = 2.11 (95% CI 1.45–3.58); obesity, OR = 3.34(95% CI 2.15–5.63); abdominal obesity, OR = 9.12 (95% CI 4.23–18.45); current smoking, OR = 1.32(95% CI 1.02–1.99); irregular exercise, OR =1.43 (95% CI 1.03–2.18); anxiety symptoms, OR = 1.56 (95%CI 1.08–2.27).Conclusions: Hypertension is a health problem for Vietnamese seafarers. To minimise hypertension, seafarersneed to adjust their lifestyle, increase regular exercise and improve psychological issues on board

    Nanoparticles as a control for cyanobacterial bloom

    This study aims to investigate the toxicity of copper material synthesized by chemical reduction method and effects of environmental variables on growth of phytoplankton community (dominated by Microcystis genus) in the Tien eutrophic lake, Hanoi, Vietnam. The variables analyzed include: physical (pH and Turbidity), chemical (content of NH4+, PO43- and copper metal), biological (content of Chlorophyll-a, cell density). The characteristic of nanomaterial was confirmed by using UVvisible spectrophotometer, XRD, SEM and TEM methods. The CuNPs showed they spherical form and uniform size about 20-40 nm. The experimental results showed that the treated with CuNPs inhibition on growth against phytoplankton after 8 days. The cell density of phytoplankton community and Microcystis genus in samples exposure with CuNPs declined after 8 days from 647.037 and 467.037 down to 381.111 and 202.592, respectively.Mục đích của nghiên cứu này là khảo sát độc tính của vật liệu nano đồng được tổng hợp bằng phương pháp khử hóa học và ảnh hưởng của các yếu tố môi trường đến sinh trưởng và phát triển của quần xã thực vật nổi (chủ yếu là chi Microcystis) trong nước hồ Tiền phú dưỡng, tại Hà Nội, Việt Nam. Các thông số phân tích bao gồm: thủy lý (pH và độ đục), hóa học (hàm lượng amoni, photphat và hàm lượng đồng kim loại), sinh học (hàm lượng chất diệp lục, mật độ tế bào). Đặc trưng của vật liệu được xác định bằng các phương pháp quang phổ UV-VIS, XRD, SEM và TEM. Vật liệu nano đồng có dạng hình cầu, kích thước đồng nhất từ 20 đến 40 nm. Kết quả thử nghiệm sau 8 ngày cho thấy các mẫu có bổ sung vật liệu nano đồng ức chế sinh trưởng quần xã thực vật nổi ở nồng độ 1mg/l. Mật độ quần xã thực vật nổi và chi Microcystis trong mẫu xử lý với CuNPs đã giảm tương ứng sau 8 ngày từ 647.037 và 467.037 xuống còn 381.111 và 202.592

    Smoke-free environment policy in Vietnam: What did people see and how did they react when they visited various public places?

    Introduction: Since Vietnam has signed WHO framework on tobacco control (FCTC) in 2003 and has issued tobacco control law in 2013, there has been little research concerning about what impacts smoke-free regulations have had on public compliance. The objective of this study was to assess public exposure to secondhand smoke and reaction toward smoke-free policy regulations in Vietnam and the associated factor. Methods: Using the design of GATS (Global Adult Tobacco Survey), a nationally representative sample of 8,996 adults were approached for data collection. Logistic regression was used to examine the associated factor.Results: The study revealed that the prevalence of respondents exposed to secondhand smoke was much higher in bars/café/tea shops (90.07%) and restaurants (81.81%) than in any other public places, universities (36.70%), government buildings (31.12%), public transport (20.04%), healthcare facilities (17.85%) and schools (15.84%). 13.23% of respondents saw smokers violate smoke-free regulations. Among those who saw them violate smoke-free regulations, just one-third cautioned them to stop smoking. Strikingly, a higher rate of cautioning smokers to stop smoking was observed among the older, married, and better educated respondents. Respondents who were married, better educated and in lower economic status were more likely to remind smokers to stop smoking.Conclusions: The study has called for strengthening two of the six MPOWER (Monitor, Protect, Offer, Warn, Enforce and Raise) components of the tobacco free initiative introduced by WHO, Monitoring tobacco use and prevention policies and Protecting people from tobacco smoke

    Host Transcription Profile in Nasal Epithelium and Whole Blood of Hospitalized Children Under 2 Years of Age With Respiratory Syncytial Virus Infection.

    BACKGROUND: Most insights into the cascade of immune events after acute respiratory syncytial virus (RSV) infection have been obtained from animal experiments or in vitro models. METHODS: In this study, we investigated host gene expression profiles in nasopharyngeal (NP) swabs and whole blood samples during natural RSV and rhinovirus (hRV) infection (acute versus early recovery phase) in 83 hospitalized patients <2 years old with lower respiratory tract infections. RESULTS: Respiratory syncytial virus infection induced strong and persistent innate immune responses including interferon signaling and pathways related to chemokine/cytokine signaling in both compartments. Interferon-α/β, NOTCH1 signaling pathways and potential biomarkers HIST1H4E, IL7R, ISG15 in NP samples, or BCL6, HIST2H2AC, CCNA1 in blood are leading pathways and hub genes that were associated with both RSV load and severity. The observed RSV-induced gene expression patterns did not differ significantly in NP swab and blood specimens. In contrast, hRV infection did not as strongly induce expression of innate immunity pathways, and significant differences were observed between NP swab and blood specimens. CONCLUSIONS: We conclude that RSV induced strong and persistent innate immune responses and that RSV severity may be related to development of T follicular helper cells and antiviral inflammatory sequelae derived from high activation of BCL6

    TextANIMAR: Text-based 3D Animal Fine-Grained Retrieval

    3D object retrieval is an important yet challenging task, which has drawn more and more attention in recent years. While existing approaches have made strides in addressing this issue, they are often limited to restricted settings such as image and sketch queries, which are often unfriendly interactions for common users. In order to overcome these limitations, this paper presents a novel SHREC challenge track focusing on text-based fine-grained retrieval of 3D animal models. Unlike previous SHREC challenge tracks, the proposed task is considerably more challenging, requiring participants to develop innovative approaches to tackle the problem of text-based retrieval. Despite the increased difficulty, we believe that this task has the potential to drive useful applications in practice and facilitate more intuitive interactions with 3D objects. Five groups participated in our competition, submitting a total of 114 runs. While the results obtained in our competition are satisfactory, we note that the challenges presented by this task are far from being fully solved. As such, we provide insights into potential areas for future research and improvements. We believe that we can help push the boundaries of 3D object retrieval and facilitate more user-friendly interactions via vision-language technologies.Comment: arXiv admin note: text overlap with arXiv:2304.0573

    Effectiveness of perindopril/amlodipine fixed-dose combination in the treatment of hypertension: a systematic review

    Background: Uncontrolled blood pressure is a major risk factor for cardiovascular diseases. Fixed-dose combination (FDC) therapy offers a promising approach to addressing this challenge by providing a convenient single-tablet solution that enhances the effectiveness of blood pressure control. In our systematic review, we assess the effectiveness of perindopril/amlodipine FDC in managing blood pressure.Methods: We conducted a comprehensive search across four primary electronic databases, namely, PubMed, Virtual Health Library (VHL), Global Health Library (GHL), and Google Scholar, as of 8 February 2022. Additionally, we performed a manual search to find relevant articles. The quality of the selected articles was evaluated using the Study Quality Assessment Tools (SQAT) checklist from the National Institute of Health and the ROB2 tool from Cochrane.Results: Our systematic review included 17 eligible articles. The findings show that the use of perindopril/amlodipine FDC significantly lowers blood pressure and enhances the quality of blood pressure control. Compared to the comparison group, the perindopril/amlodipine combination tablet resulted in a higher rate of blood pressure response and normalization. Importantly, perindopril/amlodipine FDC contributes to improved patient adherence with minimal side effects. However, studies conducted to date have not provided assessments of the cost-effectiveness of perindopril/amlodipine FDC.Conclusion: In summary, our analysis confirms the effectiveness of perindopril/amlodipine FDC in lowering blood pressure, with combination therapy outperforming monotherapy and placebo. Although mild adverse reactions were observed in a small subset of participants, cost-effectiveness assessments for this treatment remain lacking in the literature
