    Musik fĂĽr eine humanistischere Gesellschaft: Leben und Werk des Komponisten GĂĽnter Kochan

    Günter Kochan (1930-2009) war einer der erfolgreichsten und bekanntesten Komponisten der DDR. Er hinterließ über 200 Werke, darunter zahlreiche bedeutende sinfonische Arbeiten, die jedoch nach der Wende größtenteils in Vergessenheit gerieten. Kochan galt als staatsnaher Komponist und wurde dementsprechend nach 1990 an den Rand gedrängt, seine Musik wurde nun vor allem nach ihrem politischen Inhalt hinterfragt und geriet aufgrund ausbleibender Aufführungen und Neuveröffentlichungen alsbald in Vergessenheit. Die Arbeit behandelt das Spannungsfeld zwischen auferlegten kulturpolitischen Regeln und individuellem künstlerischen Entfaltungsstreben vor dem Hintergrund der offiziellen Leitlinie des "sozialistischen Realismus" in der DDR. Sie beleuchtet dieses exemplarisch anhand der Biographie und des Schaffens von Günter Kochan. Nach einer allgemeinen Darstellung der DDR-Kulturpolitik von 1949-1990, einer Kurzbiographie und einem Überblick über Kochans Gesamtschaffen konzentriert sich die Arbeit auf die Analyse und den Vergleich der 2. und 6. Sinfonie (entstanden 1969 und 2006), die als Beispiele für Kochans musikalische Hauptdomäne herangezogen werden und an denen typische Merkmale seiner Musik im mittleren und im greiften Stil vergleichend herausgearbeitet werden. Die Arbeit baut auf mehreren Interviews mit Vertrauten Kochans auf und nutzt zudem bisher wenig bis gar nicht beachtete Quellen sowie bisher unveröffentlichte Werke Kochans. Sie ist die erste Gesamtdarstellung zu Leben und Werk des Komponisten und enthält zudem das erste vollständige, systematisch und chronologisch geordnete Werkverzeichnis der Kompositionen Günter Kochans sowie eine Übersicht der derzeit verfügbaren Tondokumente

    Age- and sex-specific standard scores for the reading the mind in the eyes test

    The reliable, valid and economic assessment of social cognition is more relevant than ever in the field of clinical psychology. Theory of Mind is one of the most important socio-cognitive abilities but standardized assessment instruments for adults are rare. The Reading the Mind in the Eyes Test (RMET) is well-established and captures the ability to identify mental states from gaze. Here, we computed standard scores for the German version of the RMET derived from a large, community-dwelling sample of healthy adults (20–79 years). The standardization sample contains 966 healthy adult individuals of the population-based Leipzig Research Center for Civilization Diseases (LIFE) study. Before standardization, weighting factors were applied to match the current sample with distribution characteristics of the German population regarding age, sex, and education. RMET scores were translated into percentage ranks for men and women of five age groups (20–29, 30–39, 40–49, 50–59, 60+ years). Age-specific percentage ranks are provided for men and women. Independent of age, men present a larger variance in test scores compared to women. Within the specific age groups, women score higher and their scoring range is less variable. With increasing age, the scoring variance increases in both men and women. This is the first study providing age- and sex-specific RMET standard scores. Data was weighted to match German population characteristics, enabling the application of standard scores across German-speaking areas. Our results contribute to the standardized assessment of socio-cognitive abilities in clinical diagnostics

    Resveratrol differentially regulates NAMPT and SIRT1 in hepatocarcinoma cells and primary human hepatocytes

    Resveratrol is reported to possess chemotherapeutic properties in several cancers. In this study, we wanted to investigate the molecular mechanisms of resveratrol-induced cell cycle arrest and apoptosis as well as the impact of resveratrol on NAMPT and SIRT1 protein function and asked whether there are differences in hepatocarcinoma cells (HepG2, Hep3B cells) and non-cancerous primary human hepatocytes. We found a lower basal NAMPT mRNA and protein expression in hepatocarcinoma cells compared to primary hepatocytes. In contrast, SIRT1 was significantly higher expressed in hepatocarcinoma cells than in primary hepatocytes. Resveratrol induced cell cycle arrest in the S- and G2/M- phase and apoptosis was mediated by activation of p53 and caspase-3 in HepG2 cells. In contrast to primary hepatocytes, resveratrol treated HepG2 cells showed a reduction of NAMPT enzymatic activity and increased p53 acetylation (K382). Resveratrol induced NAMPT release from HepG2 cells which was associated with increased NAMPT mRNA expression. This effect was absent in primary hepatocytes where resveratrol was shown to function as NAMPT and SIRT1 activator. SIRT1 inhibition by EX527 resembled resveratrol effects on HepG2 cells. Furthermore, a SIRT1 overexpression significantly decreased both p53 hyperacetylation and resveratrol-induced NAMPT release as well as S-phase arrest in HepG2 cells. We could show that NAMPT and SIRT1 are differentially regulated by resveratrol in hepatocarcinoma cells and primary hepatocytes and that resveratrol did not act as a SIRT1 activator in hepatocarcinoma cells

    Comparative analysis of the human saliva microbiome from different climate zones: Alaska, Germany, and Africa

    Background: Although the importance of the human oral microbiome for health and disease is increasingly recognized, variation in the composition of the oral microbiome across different climates and geographic regions is largely unexplored. Results: Here we analyze the saliva microbiome from native Alaskans (76 individuals from 4 populations), Germans (10 individuals from 1 population), and Africans (66 individuals from 3 populations) based on next-generation sequencing of partial 16S rRNA gene sequences. After quality filtering, a total of 67,916 analyzed sequences resulted in 5,592 OTUs (defined at ≥97% identity) and 123 genera. The three human groups differed significantly by the degree of diversity between and within individuals (e.g. beta diversity: Africans > Alaskans > Germans; alpha diversity: Germans > Alaskans > Africans). UniFrac, network, ANOSIM, and correlation analyses all indicated more similarities in the saliva microbiome of native Alaskans and Germans than between either group and Africans. The native Alaskans and Germans also had the highest number of shared bacterial interactions. At the level of shared OTUs, only limited support for a core microbiome shared across all three continental regions was provided, although partial correlation analysis did highlight interactions involving several pairs of genera as conserved across all human groups. Subsampling strategies for compensating for the unequal number of individuals per group or unequal sequence reads confirmed the above observations. Conclusion: Overall, this study illustrates the distinctiveness of the saliva microbiome of human groups living under very different climatic conditions

    OpenVirtualObjects: An open set of standardized and validated 3D household objects for virtual reality-based research, assessment, and therapy

    Virtual reality (VR) technology provides clinicians, therapists, and researchers with new opportunities to observe, assess, and train behavior in realistic yet well-controlled environments. However, VR also comes with a number of challenges. For example, compared to more abstract experiments and tests on 2D computer screens, VR-based tasks are more complex to create, which can make it more expensive and time-consuming. One way to overcome these challenges is to create, standardize, and validate VR content and to make it openly available for researchers and clinicians. Here we introduce the OpenVirtualObjects (OVO), a set of 124 realistic 3D household objects that people encounter and use in their everyday lives. The objects were rated by 34 younger and 25 older adults for recognizability, familiarity, details (i.e., visual complexity), contact, and usage (i.e., frequency of usage in daily life). All participants also named and categorized the objects. We provide the data and the experiment- and analysis code online. With OVO, we hope to facilitate VR-based research and clinical applications. Easy and free availability of standardized and validated 3D objects can support systematic VR-based studies and the development of VR-based diagnostics and therapeutic tools

    Mindreading from the eyes declines with aging: Evidence from 1,603 subjects

    Social cognition, in particular mindreading, enables the understanding of another individual’s feelings, intentions, desires, and mental states. The Reading the Mind in the Eyes Test (RMET) captures the ability to identify mental states from gaze. We investigated RMET accuracy in the context of age and cognition across the whole adult age-range (19–79 years) in a very large population-based sample (N = 1,603) with linear regression models accounting for cognitive abilities, neurological diseases, and psychiatric disorders. Higher age predicted lower RMET performance in women and men, suggesting difficulties to infer mental states from gaze at older age. Effects remained stable when taking other cognitive abilities and psychiatric disorders or neurological diseases into account. Our results show that RMET performance as a measure of social cognition declines with increasing age
