C1ql4 regulates breast cancer cell stemness and epithelial-mesenchymal transition through PI3K/AKT/NF-κB signaling pathway
BackgroundThe stemness characteristic of breast cancer (BC) is a crucial factor underlying cancer recurrence and metastasis after operative therapy and chemoradiotherapy. Understanding the potential mechanism of breast cancer stem cells (BCSCs) may ameliorate the prognosis of patients.MethodsWe collected clinical specimens of BC patients for staining and statistical analysis to verify the expression status and clinical significance of complement C1q-like 4 (C1ql4). Western blot and qRT-PCR were employed to detect the expression of molecules. Flow cytometry was used to examine cell cycle, cell apoptosis and the portion of BCSCs. Wound healing and Transwell assays were used to detect cell metastasis. The effect of C1ql4 on breast cancer progression in vivo was examined in a nude mouse tumor bearing model.ResultsOur clinical analysis showed that C1ql4 was highly expressed in BC tissues and cell lines, and the high expression of C1ql4 was significantly corelated with the malignancy of BC patients. Moreover, we also found that C1ql4 was overexpressed in BCSCs. C1ql4 knockdown suppressed the BCSC and EMT properties, promoted cell cycle progression, enhanced BC cell apoptosis, and inhibited cell migration and invasion, whereas the C1ql4 overexpression exhibited the opposite effects. Mechanistically, C1ql4 promoted the activation and nuclear location of NF-κB and the expression of downstream factors TNF-α and IL-1β. Moreover, inhibition of PI3K/AKT signaling suppressed the C1ql4-induced stemness and EMT.ConclusionsOur findings suggest that C1ql4 promotes the BC cell stemness and EMT via modulating the PI3K/AKT/NF-κB signaling, and provides a promising target for BC treatment
Structure and Luminescence Properties of Eu3+-Doped Cubic Mesoporous Silica Thin Films
Eu3+ ions-doped cubic mesoporous silica thin films with a thickness of about 205 nm were prepared on silicon and glass substrates using triblock copolymer as a structure-directing agent using sol–gel spin-coating and calcination processes. X-ray diffraction and transmission electron microscopy analysis show that the mesoporous silica thin films have a highly ordered body-centered cubic mesoporous structure. High Eu3+ ion loading and high temperature calcination do not destroy the ordered cubic mesoporous structure of the mesoporous silica thin films. Photoluminescence spectra show two characteristic emission peaks corresponding to the transitions of5D0-7F1 and 5D0-7F2 of Eu3+ ions located in low symmetry sites in mesoporous silica thin films. With the Eu/Si molar ratio increasing to 3.41%, the luminescence intensity of the Eu3+ ions-doped mesoporous silica thin films increases linearly with increasing Eu3+ concentration
Study on elastoplastic coupling mechanics model of the milled mixture of sugar cane
Abstract At present, the phenomenon of elastoplastic coupling isn’t considered in establishing the mechanics model of the milled mixture of sugar cane. In order to describe elastoplastic coupling and yield characteristics, based on uniaxial confined compression tests and direct shear tests, the evolution of the elastic parameters of the solid fiber with void ratio and the plastic deformation, and the plastic mechanics behavior of the solid fiber is described by Modified Drucker-Prager Cap model, the expressions of the parameters of the plastic mechanics model are derived and the corresponding parameters are calculated; and the elastoplastic coupling mechanics model is established. The evolution of the parameters of the model is achieved by the custom subroutine written in Fortran, the numerical simulation of the elastoplastic coupling model is realized by ABAQUS. The results of uniaxial confined compression tests and finite element simulation show that the elastoplastic coupling mechanics model is more accurate than the non-elastoplastic coupling mechanics model to predict the axial pressure. The results provide an important reference for the analysis and understanding of the milling process of sugar cane and the establishment of an accurate mathematical model of the milled mixture of sugar cane
A multimodal scheme for program segmentation and representation in broadcast video streams
With the advance of digital video recording and playback systems, the request for efficiently managing recorded TV video programs is evident so that users can readily locate and browse their favorite programs. In this paper, we propose a multimodal scheme to segment and represent TV video streams. The scheme aims to recover the temporal and structural characteristics of TV programs with visual, auditory, and textual information. In terms of visual cues, we develop a novel concept named program-oriented informative images (POIM) to identify the candidate points correlated with the boundaries of individual programs. For audio cues, a multiscale Kullback-Leibler (K-L) distance is proposed to locate audio scene changes (ASC), and accordingly ASC is aligned with video scene changes to represent candidate boundaries of programs. In addition, latent semantic analysis (LSA) is adopted to calculate the textual content similarity (TCS) between shots to model the inter-program similarity and intra-program dissimilarity in terms of speech content. Finally, we fuse the multimodal features of POIM, ASC, and TCS to detect the boundaries of programs including individual commercials (spots). Towards effective program guide and attracting content browsing, we propose a multimodal representation of individual programs by using POIM images, key frames, and textual keywords in a summarization manner. Extensive experiments are carried out over an open benchmarking dataset TRECVID 2005 corpus and promising results have been achieved. Compared with the electronic program guide (EPG), our solution provides a more generic approach to determine the exact boundaries of diverse TV programs even including dramatic spots

The work is intended to summarize the recent progress in the work of fractal theory in packaging material to provide important insights into applied research on fractal in packaging materials. The fractal analysis methods employed for inorganic materials such as metal alloys and ceramics, polymers, and their composites are reviewed from the aspects of fractal feature extraction and fractal dimension calculation methods. Through the fractal dimension of packaging materials and the fractal in their preparation process, the relationship between the fractal characteristic parameters and the properties of packaging materials is discussed. The fractal analysis method can qualitatively and quantitatively characterize the fractal characteristics, microstructure, and properties of a large number of various types of packaging materials. The method of using fractal theory to probe the preparation and properties of packaging materials is universal; the relationship between the properties of packaging materials and fractal dimension will be a critical trend of fractal theory in the research on properties of packaging materials

The enhanced treatment efficacy of invasive brain glioma by dual-targeted artemether plus paclitaxel micelles
AbstractHigh grade-gliomas are highly invasive and prone to metastasis, leading to poor survival and prognosis. Currently, we urgently need a new treatment strategy to effectively inhibit glioma. In this study, artemether and paclitaxel were used as two agents for tumour suppression. Two functional materials were synthesised and modified on the surface of the micelle as targeting molecules. The addition of two functional materials confers the ability of the micelles to effectively cross the blood-brain barrier (BBB) and then target the glioma cells. Thus, this dual-targeted delivery system allows the drug to play a better role in inhibiting tumour invasion and vasculogenic mimicry (VM) channels. In this paper, the anticancer effects of dual-targeted artemether plus paclitaxel micelles on glioma U87 cells were studied in three aspects: (I) In vitro and in vivo targeting assessment, including the role of penetrating BBB and targeting glioma; (II) In vitro regulation of invasion-associated proteins; (III) Inhibition of VM channels formation and invasion in vitro; (IV) The study of pharmacodynamics in tumour-bearing mice. These results suggest that dual-targeted artemether plus paclitaxel micelle may provide a new strategy to treat glioma via inhibiting invasive and VM channels
Taurocholic acid is an active promoting factor, not just a biomarker of progression of liver cirrhosis: evidence from a human metabolomic study and in vitro experiments
Abstract Background Previous studies have indicated that bile acid is associated with progression of liver cirrhosis. However, the particular role of specific bile acid in the development of liver cirrhosis is not definite. The present study aims to identify the specific bile acid and explore its possible mechanisms in promoting liver cirrhosis. Methods Thirty two cirrhotic patients and 27 healthy volunteers were enrolled. Age, gender, Child-Pugh classification and serum of patients and volunteers were collected. Liquid chromatography tandem mass spectrometry (LC-MS) was utilized to determine concentrations of 12 bile acids in serum. Principal component analysis, fold change analysis and heatmap analysis were used to identify the most changed bile acid. And pathway analysis was used to identify the most affected pathway in bile acid metabolism. Spearman rank correlation analysis was employed to assess correlation between concentrations of bile acids and Child-Pugh classification. Hepatic stellate cells (LX-2) were cultured in DMEM. LX-2 cells were also co-cultured with HepG2 cells in the transwell chambers. LX-2 cells were treated with Na+/taurocholate in different concentrations. Western blot was used to evaluate the expression of alpha smooth muscle actin (α-SMA), type I collagen, and Toll-like receptor 4 (TLR4) in LX-2 cells. Results Concentrations of 12 bile acids in serum of patients and healthy volunteers were determined with LC-MS successively. Principal component analysis, fold change analysis and heatmap analysis identified taurocholic acid (TCA) to be the most changed bile acid. Pathway analysis showed that TCA biosynthesis increased significantly. Spearman rank correlation analysis showed that concentration of TCA in serum of cirrhotic patients was positively associated with Child-Pugh classification. TCA increased the expression of α-SMA, type I collagen, and TLR4 in LX-2 cells. Moreover, the above effect was strengthened when LX-2 cells were co-cultured with HepG2 cells. Conclusions Increased TCA concentration in serum of liver cirrhotic patients is mainly due to increased bile acid biosynthesis. TCA is an active promoter of the progression of liver cirrhosis. TCA promoting liver cirrhosis is likely through activating hepatic stellate cells via upregulating TLR4 expression. TCA is a potential therapeutic target for the prevention and treatment of liver cirrhosis
