    Comparative analysis of long noncoding RNAs in angiosperms and characterization of long noncoding RNAs in response to heat stress in Chinese cabbage

    Long noncoding RNAs (lncRNAs) are widely present in different species and play critical roles in response to abiotic stresses. However, the functions of lncRNAs in Chinese cabbage under heat stress remain unknown. Here, we first conducted a global comparative analysis of 247,242 lncRNAs among 37 species. The results indicated that lncRNAs were poorly conserved among different species, and only 960 lncRNAs were homologous to 524 miRNA precursors. We then carried out lncRNA sequencing for a genome-wide analysis of lncRNAs and their target genes in Chinese cabbage at different stages of heat treatment. In total, 18,253 lncRNAs were identified, of which 1229 differentially expressed (DE) lncRNAs were characterized as being heat-responsive. The ceRNA network revealed that 38 lncRNAs, 16 miRNAs, and 167 mRNAs were involved in the heat response in Chinese cabbage. Combined analysis of the cis- and trans-regulated genes indicated that the targets of DE lncRNAs were significantly enriched in the “protein processing in endoplasmic reticulum” and “plant hormone signal transduction” pathways. Furthermore, the majority of HSP and PYL genes involved in these two pathways exhibited similar expression patterns and responded to heat stress rapidly. Based on the networks of DE lncRNA-mRNAs, 29 and 22 lncRNAs were found to interact with HSP and PYL genes, respectively. Finally, the expression of several critical lncRNAs and their targets was verified by qRT-PCR. Overall, we conducted a comparative analysis of lncRNAs among 37 species and performed a comprehensive analysis of lncRNAs in Chinese cabbage. Our findings expand the knowledge of lncRNAs involved in the heat stress response in Chinese cabbage, and the identified lncRNAs provide an abundance of resources for future comparative and functional studies

    Autogenous deformation of high performance concrete exposed to semi-adiabatic and isothermal conditions

    587-596This paper presents an experimental investigation on early-age autogenous deformation (AD) of high performance concrete (HPC) exposed to isothermal and semi-adiabatic conditions. A new testing apparatus equipped with eddy-current displacement sensors is used for monitoring deformation from final setting to 28 days. Deformation measurements are conducted on a series of HPC mixtures with different water-binder ratios (w/b) and cementitious materials. In order to accurately separate AD from the measured deformation, time-dependent coefficients of thermal expansion (CTEs) of HPC, other than a constant value, are employed. The results indicated that AD under semi-adiabatic condition is fundamentally different from the one under isothermal condition. The former undergoes three stages successively (i.e., initial rapid shrinkage, abrupt expansion and stable shrinkage stages), while the latter shows continuous shrinkage only. The considerable expansion under semi-adiabatic condition can be explained from delayed ettringite formation and thermal shock effect of early-age AD

    Abstract Background The aim of this study is to evaluate the prognostic value of grading MRI–detected skull-base invasion in nasopharyngeal carcinoma (NPC) with skull-base invasion after intensity-modulated radiotherapy (IMRT). Methods This study is a retrospective chart review of 469 non-metastatic NPC patients with skull-base invasion. Patients were classified as extensive skull-base invasion (ESBI) group and limited skull-base invasion (LSBI) group. Results Multivariate analysis showed that the skull-base invasion (LSBI vs. ESBI) was an independent prognostic predictor of progression free survival (PFS). The estimated 5-year local failure free survival (LFFS), distant metastasis free survival (DMFS), PFS, and overall survival (OS) rates for patients in the T3-LSBI and T3-ESBI group were 92.9% versus 93.5, 89.8% versus 86.1, 81.6% versus 76.4, and 93.5% versus 86.3%, respectively (P > 0.05). Conclusion Grading of MRI-detected skull-base invasion is an independent prognostic factor of NPC with skull-base invasion. It is scientific and reasonable for skull-base invasion as a single entity to be classified as T3 classification

    Composition of dissolved organic matter controls interactions with La and Al ions : Implications for phosphorus immobilization in eutrophic lakes

    Applications of aluminium (Al) salt or lanthanum (La) modified bentonite (LMB) have become popular methodologies for immobilizing phosphorus (P) in eutrophic lakes. The presence of humic substances, has been shown to inhibit this form of treatment due to the complexation with La/Al. However, the effects of other dissolved organic matter (DOM), especially that derived from phytoplankton (the dominant source in eutrophic lakes) are unknown. In this study, the interaction with La/Al of Suwannee River Standard Humic Acid Standard II (SRHA) and algae-derived DOM (ADOM) were investigated and compared. Differed to SRHA which was dominated by polyphenol-like component (76.8%, C1-SRHA), majority in ADOM were protein-like substance, including 41.9% tryptophan-like component (C2-ADOM) and 21.0% tyrosine-like component (C3-ADOM). Two reactions of complexation and coprecipitation were observed between SRHA/ADOM and La/Al. Complexation dominated at low metal inputs less than 10 μM and coprecipitation was the main reaction at higher metal inputs. For ADOM, the tryptophan-like component (C2-ADOM) was the important component to react with metal. The reaction rate for C2-ADOM with La were about two-third of that for C1-SRHA, indicating that the influence of C2-ADOM was significant during the P immobilization by La/Al-based treatment in eutrophic lakes. The P removal data in the presence of ADOM confirmed the significant inhibition of ADOM. In addition, based on the composition of coprecipitates and relatively biodegradable character of tryptophan-like substances (C2-ADOM), the coprecipitation of ADOM was assumed to reduce the stability of precipitated P in eutrophic lakes. The release of P from the potential biodegradation of the coprecipitates and thus the possible decline of the performance of P immobilization by La/Al-based treatments is an important work in the future.</p

    Simultaneous Screening and Quantification of 479 Pesticides in Green Tea by LC-QTOF-MS

    A high-throughput screening and quantification method for 479 pesticides in green tea was established based on solid-phase extraction combined with liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-QTOF-MS). Pesticides were extracted from samples using an optimized SPE (TPT cartridges) procedure. LC-QTOF-MS in All Ions MS/MS scan mode acquired full MS data for quantification and product ion spectra for identification. LC-QTOF-MS quantification was achieved using matrix-matched standard calibration curves to achieve the optimal method accuracy. The method performance characteristics included the linearity, overall recovery, precision, and measurement uncertainty being evaluated. The validation results exhibited a good sensitivity with the LOQs of 5–55 µg/kg, which was satisfactory for their MRLs in China or the EU. The recoveries of more than 92.7% of the 479 pesticides in green tea were 70–120% at the three spiked levels with a precision of ≤20%. Finally, this method was employed to analyze 479 pesticides in 95 tea samples from markets in China. The test results of the tea samples showed that tolfenpyrad, buprofezin, and pyridaben were found with lower concentrations.     Effect of Pyrolysis Temperature on Removal Efficiency and Mechanisms of Hg(II), Cd(II), and Pb (II) by Maize Straw Biochar

    Pyrolysis temperature significantly affects the properties of biochar, which in turn can affect the removal of heavy metal ions and the underlying mechanism. In this work, biochars from the pyrolysis of maize straw at 300, 400, and 500 °C (BC300, BC400, and BC500, respectively) and wheat straw at 400 °C (WBC400) were investigated. The influence of production temperature on the adsorption of Hg2+, Cd2+, and Pb2+ by maize straw biochar was investigated by the characterization of the biochars and by adsorption tests. The adsorption capacities of maize and wheat straw biochar were compared in an adsorption experiment. Biochar BC400 showed the best physical and chemical properties and had the largest number of surface functional groups. The pseudo-second-order kinetic model was more suitable for describing the adsorption behavior of metal ions to biochar. The Langmuir model better fit the experimental data. Biochar BC400 had a higher adsorption speed and a stronger adsorption capacity than WBC400. The sorption of Pb2+ and Hg2+ to maize straw biochar followed the mechanisms of surface precipitation of carbonates and phosphates and complexation with oxygenated functional groups and delocalized π electrons. The adsorption mechanism for Cd2+ was similar to those of Hg2+ and Pb2+, but precipitation mainly occurred through the formation of phosphate. In the multi-heavy-metal system, the adsorption of Cd2+ by BC400 was inhibited by Pb2+ and Hg2+.
