    Overexpression and Small Molecule-Triggered Downregulation of CIP2A in Lung Cancer

    Lung cancer is the leading cause of cancer deaths worldwide, with a five-year overall survival rate of only 15%. Cancerous inhibitor of PP2A (CIP2A) is a human oncoprotein inhibiting PP2A in many human malignancies. However, whether CIP2A can be a new drug target for lung cancer is largely unclear.Normal and malignant lung tissues were derived from 60 lung cancer patients from southern China. RT-PCR, Western blotting and immunohistochemistry were used to evaluate the expression of CIP2A. We found that among the 60 patients, CIP2A was undetectable or very low in paratumor normal tissues, but was dramatically elevated in tumor samples in 38 (63.3%) patients. CIP2A overexpression was associated with cigarette smoking. Silencing CIP2A by siRNA inhibited the proliferation and clonogenic activity of lung cancer cells. Intriguingly, we found a natural compound, rabdocoetsin B which is extracted from a Traditional Chinese Medicinal herb Rabdosia coetsa, could induce down-regulation of CIP2A and inactivation of Akt pathway, and inhibit proliferation and induce apoptosis in a variety of lung cancer cells.Our findings strongly indicate that CIP2A could be an effective target for lung cancer drug development, and the therapeutic potentials of CIP2A-targeting agents warrant further investigation

    Asthma susceptible genes in Chinese population: A meta-analysis

    <p>Abstract</p> <p>Background</p> <p>Published data regarding the associations between genetic variants and asthma risk in Chinese population were inconclusive. The aim of this study was to investigate asthma susceptible genes in Chinese population.</p> <p>Methods</p> <p>The authors conducted 18 meta-analyzes for 18 polymorphisms in 13 genes from eighty-two publications.</p> <p>Results</p> <p>Seven polymorphisms were found being associated with risk of asthma, namely: <it>A Disintegrin and Metalloprotease 33 </it>(<it>ADAM33</it>) T1-C/T (odds ratio [OR] = 6.07, 95% confidence interval [CI]: 2.69-13.73), <it>Angiotensin-Converting Enzyme </it>(<it>ACE</it>) D/I (OR = 3.85, 95%CI: 2.49-5.94), <it>High-affinity IgE receptor β chain </it>(<it>FcεRIβ</it>) -6843G/A (OR = 1.49, 95%CI: 1.01-2.22), <it>Interleukin 13</it>(<it>IL-13</it>) -1923C/T (OR = 2.99, 95%CI: 2.12-4.24), <it>IL-13 </it>-2044A/G (OR = 1.49, 95%CI: 1.07-2.08), <it>Regulated upon Activation, Normal T cell Expressed and Secreted </it>(<it>RANTES</it>) -28C/G (OR = 1.64, 95%CI: 1.09-2.46), <it>Tumor Necrosis Factor-α </it>(<it>TNF-α</it>) -308G/A(OR = 1.42, 95%CI: 1.09, 1.85). After subgroup analysis by age, the <it>ACE </it>D/I, <it>β2-Adrenergic Receptor </it>(<it>β2-AR</it>) -79G/C, <it>TNF-α </it>-308G/A, <it>Interleukin 4 receptor</it>(<it>IL-4R</it>) -1902G/A and <it>IL-13 </it>-1923C/T polymorphisms were found significantly associated with asthma risk in Chinese children. In addition, the <it>ACE </it>D/I, <it>FcεRIβ </it>-6843G/A, <it>TNF-α </it>-308G/A, <it>IL-13 </it>-1923C/T and <it>IL-13 </it>-2044A/G polymorphisms were associated with asthma risk in Chinese adults.</p> <p>Conclusion</p> <p><it>ADAM33, FcεRIβ, RANTES, TNF-α, ACE, β2-AR, IL-4R </it>and <it>IL-13 </it>genes could be proposed as asthma susceptible genes in Chinese population. Given the limited number of studies, more data are required to validate these associations.</p

    Collaborative location recommendation by integrating multi-dimensional contextual information

    Point-of-Interest (POI) recommendation is a new type of recommendation task that comes along with the prevalence of location-based social networks and services in recent years. Compared with traditional recommendation tasks, POI recommendation focuses more on making personalized and context-aware recommendations to improve user experience. Traditionally, the most commonly used contextual information includes geographical and social context information. However, the increasing availability of check-in data makes it possible to design more effective location recommendation applications by modeling and integrating comprehensive types of contextual information, especially the temporal information. In this article, we propose a collaborative filtering method based on Tensor Factorization, a generalization of the Matrix Factorization approach, tomodel themulti-dimensional contextual information. Tensor Factorization naturally extendsMatrix Factorization by increasing the dimensionality of concerns, within which the three-dimensional model is the one most popularly used. Our method exploits a high-order tensor to fuse heterogeneous contextual information about users' check-ins instead of the traditional two-dimensional user-location matrix. The factorization of this tensor leads to a more compact model of the data that is naturally suitable for integrating contextual information to make POI recommendations. Based on the model, we further improve the recommendation accuracy by utilizing the internal relations within users and locations to regularize the latent factors. Experimental results on a large real-world dataset demonstrate the effectiveness of our approach

    In situ spectroelectrochemical behaviour of nanocrystalline TiO2 thin films electrode fabricated by pulsed laser deposition

    Nanocrystalline titanium oxide thin films have been successfully deposited on ITO coated glass by pulsed laser ablation of metallic Ti target in O-3/O-2 ambient gases. The intercalation of Li ions in the anatase TiO2 film electrode is examined by cyclic voltammetry. The electrochromic behaviour of TiO2 electrode is investigated by in-situ visible transmittance measurement, and two absorption bands at 420 and 650 nm are observed. The absorption falling and rising in color changing with excellent revisibility is relative to the insertion and deintercalation processes of Li ion. These results suggest that nanocrystalline titanium oxide films fabricated by pulsed laser deposition exhibit excellent spectroelectrochemical property

    When things matter: A survey on data-centric internet of things

    With the recent advances in radio-frequency identification (RFID), low-cost wireless sensor devices, and Web technologies, the Internet of Things (IoT) approach has gained momentum in connecting everyday objects to the Internet and facilitating machine-to-human and machine-to-machine communication with the physical world. IoT offers the capability to connect and integrate both digital and physical entities, enabling a whole new class of applications and services, but several significant challenges need to be addressed before these applications and services can be fully realized. A fundamental challenge centers around managing IoT data, typically produced in dynamic and volatile environments, which is not only extremely large in scale and volume, but also noisy and continuous. This paper reviews the main techniques and state-of-the-art research efforts in IoT from data-centric perspectives, including data stream processing, data storage models, complex event processing, and searching in IoT. Open research issues for IoT data management are also discussed

    The C-H-O isotope systematics of the Yangshan gold deposit, Gansu and its implication for the ore-fluid origin

    The Yangshan gold deposit, Wenxian county, Gansu province, containing 308 t Au with average grade of 4.74g/t, is now ranked as the China' s largest gold deposit. Locating in western Qinling Orogen, central China, it is a syn-collisionally formed Carlin-like gold deposit. Its orebodies are controlled by an east-trending shear-zone and hosted in the Devonian carbonaceous carbonate-phyllite-slate sequence or the granite-porphyry dikes intruding into the Devonian strata. The δ13 CCO2(PDB) and δ13 CCH4(PDB) ratios of fluid inclusions within quartz separates range -2.5‰ ∼ -5.6‰ and -23.1‰ ∼ -32.6‰, respectively, suggesting that the ore-fluids have been mainly sourced, through metamorphism and/or reworking, from the Devonian strata or/and similar lithologies which comprise carbonaceous phyllite, slate, chert and carbonate. This conclusion is strongly supported by the δ18 O values of the early- and main-stage ore fluids, which range from 9.5‰ to 15.3‰, with corresponding δD values between -86‰ ∼ -73‰. Two δ18 O values of late ore-forming stage ore-fluids are 2.7‰ and 6.8‰, implying a significant input of meteoric water. In general, the ore-forming fluid-system varies from early, deep, metamorphic fluid to late, shallow, meteoric water. It is worthy of stating that the above results and understandings are consistent with conclusions drawn from our studies of fluid inclusions and ore geology.link_to_subscribed_fulltex
