    Adsorption of mono- and multivalent cat- and anions on DNA molecules

    Adsorption of monovalent and multivalent cat- and anions on a deoxyribose nucleic acid (DNA) molecule from a salt solution is investigated by computer simulation. The ions are modelled as charged hard spheres, the DNA molecule as a point charge pattern following the double-helical phosphate strands. The geometrical shape of the DNA molecules is modelled on different levels ranging from a simple cylindrical shape to structured models which include the major and minor grooves between the phosphate strands. The densities of the ions adsorbed on the phosphate strands, in the major and in the minor grooves are calculated. First, we find that the adsorption pattern on the DNA surface depends strongly on its geometrical shape: counterions adsorb preferentially along the phosphate strands for a cylindrical model shape, but in the minor groove for a geometrically structured model. Second, we find that an addition of monovalent salt ions results in an increase of the charge density in the minor groove while the total charge density of ions adsorbed in the major groove stays unchanged. The adsorbed ion densities are highly structured along the minor groove while they are almost smeared along the major groove. Furthermore, for a fixed amount of added salt, the major groove cationic charge is independent on the counterion valency. For increasing salt concentration the major groove is neutralized while the total charge adsorbed in the minor groove is constant. DNA overcharging is detected for multivalent salt. Simulations for a larger ion radii, which mimic the effect of the ion hydration, indicate an increased adsorbtion of cations in the major groove.Comment: 34 pages with 14 figure

    Availability of Evidence for Predictive Machine Learning Algorithms in Primary Care: A Systematic Review

    Importance: The aging and multimorbid population and health personnel shortages pose a substantial burden on primary health care. While predictive machine learning (ML) algorithms have the potential to address these challenges, concerns include transparency and insufficient reporting of model validation and effectiveness of the implementation in the clinical workflow. Objectives: To systematically identify predictive ML algorithms implemented in primary care from peer-reviewed literature and US Food and Drug Administration (FDA) and Conformité Européene (CE) registration databases and to ascertain the public availability of evidence, including peer-reviewed literature, gray literature, and technical reports across the artificial intelligence (AI) life cycle. Evidence Review: PubMed, Embase, Web of Science, Cochrane Library, Emcare, Academic Search Premier, IEEE Xplore, ACM Digital Library, MathSciNet, AAAI.org (Association for the Advancement of Artificial Intelligence), arXiv, Epistemonikos, PsycINFO, and Google Scholar were searched for studies published between January 2000 and July 2023, with search terms that were related to AI, primary care, and implementation. The search extended to CE-marked or FDA-approved predictive ML algorithms obtained from relevant registration databases. Three reviewers gathered subsequent evidence involving strategies such as product searches, exploration of references, manufacturer website visits, and direct inquiries to authors and product owners. The extent to which the evidence for each predictive ML algorithm aligned with the Dutch AI predictive algorithm (AIPA) guideline requirements was assessed per AI life cycle phase, producing evidence availability scores. Findings: The systematic search identified 43 predictive ML algorithms, of which 25 were commercially available and CE-marked or FDA-approved. The predictive ML algorithms spanned multiple clinical domains, but most (27 [63%]) focused on cardiovascular diseases and diabetes. Most (35 [81%]) were published within the past 5 years. The availability of evidence varied across different phases of the predictive ML algorithm life cycle, with evidence being reported the least for phase 1 (preparation) and phase 5 (impact assessment) (19% and 30%, respectively). Twelve (28%) predictive ML algorithms achieved approximately half of their maximum individual evidence availability score. Overall, predictive ML algorithms from peer-reviewed literature showed higher evidence availability compared with those from FDA-approved or CE-marked databases (45% vs 29%). Conclusions and Relevance: The findings indicate an urgent need to improve the availability of evidence regarding the predictive ML algorithms' quality criteria. Adopting the Dutch AIPA guideline could facilitate transparent and consistent reporting of the quality criteria that could foster trust among end users and facilitating large-scale implementation

    No Brasil, pouca atenção se tem dado às formas e caminhos com que os fluxos energéticos se distribuem nos sistemas produtivos. Na agropecuária, a atenção tem sido voltada a novas fontes de energia (biomassa) ou em tecnologia alternativa, visando a racionalização do uso de energia fóssil ou elétrica. Entretanto, a agricultura tem se desenvolvido baseada fortemente na utilização intensiva de máquinas agrícolas, com conseqüente uso de combustíveis fósseis. Um fator de estrangulamento muito forte no consumo energético geral tem sido a utilização massiva de fertilizantes derivados do petróleo nos agroecossistemas. Estudos de Balanços Energéticos visam determinar os pontos de estrangulamento energético fundamentando a busca por tecnologias poupadoras de energia, especialmente aquelas de origem fóssil (combustível, fertilizante, agrotóxicos, energia despendida na fabricação das máquinas e implementos, etc.). No Brasil, a Região Sul, é onde se encontram vários trabalhos buscando uma agricultura mais auto-sustentável, do ponto de vista da utilização da energia. Em vista da possibilidade de eventuais futuras crises energéticas, o presente trabalho procura analisar o estado-da-arte dos estudos em Balanço Energético, no Brasil e no Mundo, como uma ferramenta de indicação da sustentabilidade dos sistemas agropecuários

    Comparación de distintas estrategias para la predicción de muerte a corto plazo en el paciente anciano infectado

    Objective. The aim of this study was to determine the utility of a post hoc lactate added to SIRS and qSOFA score to predict 30-day mortality in older non-severely dependent patients attended for infection in the Emergency Department (ED). Methods. We performed an analytical, observational, prospective cohort study including patients of 75 years of age or older, without severe functional dependence, attended for an infectious disease in 69 Spanish ED for 2-day three seasonal periods. Demographic, clinical and analytical data were collected. The primary outcome was 30-day mortality after the index event. Results. We included 739 patients with a mean age of 84.9 (SD 6.0) years; 375 (50.7%) were women. Ninety-one (12.3%) died within 30 days. The AUC was 0.637 (IC 95% 0.587-0.688; p= 2 and 0.698 (IC 95% 0.635- 0.761; p= 2. Comparing receiver operating characteristic (ROC) there was a better accuracy of qSOFA vs SIRS (p=0.041). Both scales improve the prognosis accuracy with lactate inclusion. The AUC was 0.705 (IC95% 0.652-0.758; p<0.001) for SIRS plus lactate and 0.755 (IC95% 0.696-0.814; p<0.001) for qSOFA plus lactate, showing a trend to statistical significance for the second strategy (p=0.0727). Charlson index not added prognosis accuracy to SIRS (p=0.2269) or qSOFA (p=0.2573). Conclusions. Lactate added to SIRS and qSOFA score improve the accuracy of SIRS and qSOFA to predict short-term mortality in older non-severely dependent patients attended for infection. There is not effect in adding Charlson index

    Na agricultura moderna, interessam sistemas de produção eficientes no uso da energia. Objetivou-se avaliar a conversão e o balanço energético de cinco sistemas de rotação de culturas envolvendo o triticale. Os sistemas avaliados, no período de 1987 a 1991, foram: I (triticale/soja), II (triticale/soja e aveia preta/soja), III (triticale/soja e ervilhaca/milho), IV (triticale/ soja, ervilhaca/milho e aveia preta/soja) e V (triticale/soja, triticale/soja, ervilhaca/milho e aveia preta/soja). Em 1990, nos sistemas II, IV e V, a aveia preta foi substituída por aveia branca. O experimento foi estabelecido em plantio direto, em delineamento de blocos ao acaso, com três repetições e parcelas com área útil de 24 m². Na média do período de 1987 a 1989, o sistema III apresentou conversão (9,30) e balanço energético (23.860 Mcal/ha) maiores do que os demais sistemas estudados (I: 5,38, II: 5,02, IV: 8,12 e V: 7,37; I: 18.067 Mcal/ha, II: 13.790 Mcal/ha, IV: 19.875 Mcal/ha e V: 19.264 Mcal/ha, respectivamente). Nesse período, as condições climáticas transcorreram normalmente. Na média do período de 1990 a 1991, não houve diferenças significativas entre as médias para conversão e para balanço energético. Nesse período, as condições climáticas foram adversas às culturas em estudo.<br>Efficient energy conversion production systems are important for modern agriculture. The objetive was to evaluate the energy conversion and balance of five rotation systems that included triticale. The evaluated systems, from 1987 to 1991, were: I (triticale/soybean), II (triticale/soybean and black oats/soybean), III (triticale/soybean and common vetch/corn), IV (triticale/soybean, common vetch/corn, and black oats/soybean), and V (triticale/soybean, triticale/soybean, common vetch/corn, and black oats/ soybean). In 1990, black oats was replaced by white oats in systems II, IV, and V. The experiment was set up under no-tillage, using a randomized block design with three replications and plots totalizing 24 m² . On average, for the period 1987 to 1989, system III showed higher energy conversou (9.30) and balance (23.860 Mcal/ha), as compared to the othen studied systems (I: 5.38, II: 5.02, IV: 8.12 e V: 7.37; I: 18.067 Mcal/ha, II: 13.790 Mcal/ha, IV: 19.875 Mcal/ha e V: 19.264 Mcal/ha, respectivily). In this period, the climatic conditions were normal. On average, the period 1990 to 1991, there were no significant differences between energy conversion and balance means.     Desenvolveu-se de 1997 a 2003, em Passo Fundo (RS), um experimento constituído de quatro sistemas de manejo de solo (plantio direto, cultivo mínimo, preparo convencional de solo com arado de discos e preparo convencional de solo com arado de aivecas) e de três sistemas de rotação de culturas: sistema I (trigo/soja); sistema II (trigo/soja e ervilhaca/milho ou sorgo) e sistema III (trigo/soja, ervilhaca/milho ou sorgo e aveia branca/soja). O delineamento experimental foi o de blocos ao acaso, com parcelas subdivididas e três repetições. A parcela principal foi constituída pelos sistemas de manejo de solo, e as subparcelas, pelos sistemas de rotação de culturas. Neste trabalho, apresentam-se a conversão energética (energia disponível/energia consumida) e o balanço energético (energia disponível - energia consumida) no período de 7 anos. No sistema plantio direto houve conversão e balanço energéticos (72,44 e 190.766 MJ ha-1) superiores ao cultivo mínimo (64,06 e 167.349 MJ ha-1), aos preparos convencionais de solo com arado de discos (54,35 e 134.982 MJ ha-1) e com arado de aivecas (52,02 e 128.159 MJ ha-1) respectivamente. O trigo em rotação de culturas foi mais eficiente energeticamente do que a monocultura desse cereal. Na cultura de milho foi observada maior eficiência energética.<br>The effects of soil management systems and crop rotations were assessed from 1997 to 2003, in Passo Fundo, Rio Grande do Sul State, Brazil. Four soil management systems (no-tillage, minimum tillage, conventional tillage using disk plow, and conventional tillage using moldboard plow) and three crop rotation systems [system I (wheat/soybean), system II (wheat/soybean and common vetch/corn or sorghum), and system III (wheat/soybean, common vetch/corn or sorghum and white oats/soybean)] were compared. An experimental design of blocks at random with split-plots and three replications was used. The main plot consisted of soil management systems, while the split-plots consisted of crop rotation systems. Energy conversion (energy available/energy consumed) and balance (energy available - energy consumed) during a seven-year period is presented in this paper. No-tillage showed higher energy conversion and balance (72.44 and 190,766 MJ ha-1) than minimum tillage (64.06 and 167,349 MJ ha-1), conventional tillage using disk plow (54.35 and 134,982 MJ ha-1), and conventional tillage using moldboard (52.02 and 128,159 MJ ha-1), respectively. Wheat within crop rotations presented higher energy efficiency in energy than the monoculture of this cereal. The corn crop was observed the highest energy efficiency of this study
