In Vitro Evaluation of the Toxicological Profile and Oxidative Stress of Relevant Diet-Related Advanced Glycation End Products and Related 1,2-Dicarbonyls
During food processing and storage, and in tissues and fluids under physiological conditions, the Maillard reaction occurs. During this reaction, reactive 1,2-dicarbonyl compounds arise as intermediates that undergo further reactions to form advanced glycation end products (AGEs). Diet is the primary source of exogenous AGEs. Endogenously formed AGEs have been proposed as a risk factor in the pathogenesis of diet-related diseases such as diabetes, insulin resistance, cardiovascular diseases, or chronic disease. AGEs may differently contribute to the diet-related exacerbation of oxidative stress, inflammation, and protein modifications. Here, to understand the contribution of each compound, we tested individually, for the first time, the effect of five 1,2-dicarbonyl compounds 3-deoxyglucosone (3-DG), 3-deoxygalactosone (3-DGal), 3,4-dideoxyglucosone-3-ene (3,4-DGE), glyoxal (GO), and methylglyoxal (MGO) and four different glycated amino acids N-ε-(carboxyethyl)lysine (CEL), N-ε-(carboxymethyl)lysine (CML), methylglyoxal-derived hydroimidazolone-1 (MG-H1), and pyrraline (Pyrr) in a cell line of human keratinocytes (HaCaT). We found that most of the glycated amino acids, i.e., CEL, CML, and MG-H1, did not show any cytotoxicity. At the same time, 1,2-dicarbonyl compounds 3-DGal, 3,4-DGE, GO, and MGO increased the production of reactive oxygen species and induced cell death. MGO induced cell death by apoptosis, whereas 3-DGal and 3,4-DGE induced nuclear translocation of the proinflammatory NF-κB transcription pathway, and the activation of the pyroptosis-related NLRP3 inflammasome cascade. Overall, these results demonstrate the higher toxic impact of 1,2-dicarbonyl compounds on mucosal epithelial cells when compared to glycated amino acids and the selective activation of intracellular signaling pathways involved in the crosstalk mechanisms linking oxidative stress to excessive inflammation
Rapid Plant Identification Using Species- and Group-Specific Primers Targeting Chloroplast DNA
Plant identification is challenging when no morphologically assignable parts are available. There is a lack of broadly applicable methods for identifying plants in this situation, for example when roots grow in mixture and for decayed or semi-digested plant material. These difficulties have also impeded the progress made in ecological disciplines such as soil- and trophic ecology. Here, a PCR-based approach is presented which allows identifying a variety of plant taxa commonly occurring in Central European agricultural land. Based on the trnT-F cpDNA region, PCR assays were developed to identify two plant families (Poaceae and Apiaceae), the genera Trifolium and Plantago, and nine plant species: Achillea millefolium, Fagopyrum esculentum, Lolium perenne, Lupinus angustifolius, Phaseolus coccineus, Sinapis alba, Taraxacum officinale, Triticum aestivum, and Zea mays. These assays allowed identification of plants based on size-specific amplicons ranging from 116 bp to 381 bp. Their specificity and sensitivity was consistently high, enabling the detection of small amounts of plant DNA, for example, in decaying plant material and in the intestine or faeces of herbivores. To increase the efficacy of identifying plant species from large number of samples, specific primers were combined in multiplex PCRs, allowing screening for multiple species within a single reaction. The molecular assays outlined here will be applicable manifold, such as for root- and leaf litter identification, botanical trace evidence, and the analysis of herbivory

Las consultas de CirugÃa Plástica se están llenando de pacientes con expectativas crecientes acerca de los resultados de los tratamientos con distintas fuentes de energÃa. El objetivo es conseguir un elevado Ãndice de beneficio cosmético en el mÃnimo número de sesiones y con un tiempo breve de recuperación. Los tres elementos sobre los que podemos trabajar son el soporte, la textura y las alteraciones pigmentarias de la piel. Los sistemas de luz y de radiofrecuencia permiten combinar distintos niveles de tratamiento, como son la dermis profunda, la dermis papilar y la epidermis y constituyen un complemento a los procedimientos quirúrgicos, aportando un concepto "biológico" al estimular la regeneración y la neocolagenogénesis. En este artÃculo se describen los fundamentos de estos tratamientos complementarios que son de obligado conocimiento para los cirujanos plásticos.Plastic Surgery offices are currently full of patients who have growing expectations about the outcome of the treatments made with different energy sources. The aim is to get a high cosmetic benefit index with the lowest number of sessions and recovery time. The three elements we can work with are tone, texture and pigmentary skin disorders. The light and radiofrequency systems allow the combination of different treatment levels, such as deep dermis, papillary dermis and epidermis. These systems represent a complement for the surgical procedures, providing a "biological" concept as they stimulate the tissue regeneration and the neocollagenosis. Estudio de los tumores sólidos desarrollados por los ratones durante la producción de Anticuerpos Monoclonales
Los ratones constituyen un elemento fundamental para la producción de lÃquido ascÃtico rico en anticuerpos monoclonales (AcM). Durante este proceso suelen aparecer tumores sólidos que afectan directamente el volumen de producción, por lo que el objetivo de este trabajo consistió en estudiar estas neoformaciones para establecer sus caracterÃsticas y de esta manera poder predecir las afectaciones en la producción a escala industrial de lÃquido ascÃtico. Se utilizó la información de 12 250 animales de ambos sexos agrupados en 12 lotes de producción de los hibridomas CB-HEP.1,IOR r3 e IOR t1, determinando el momento de aparición, la incidencia de tumores por sexos e
hibridomas, asà como sus caracterÃsticas anatomopatológicas. Se demostró que los tumores comienzan a aparecer a partir del dÃa 14 postinoculación, llegando a afectar al 3,4% de los animales el dÃa 20 postinoculación, existiendo diferencias significativas (p<0,05) en la frecuencia de aparición en los machos respecto a las hembras (4,4% vs 2,3%). Además, se observó mayor incidencia del hibridoma CB-HEP.1, seguido de ior t1 e ior r3 (3,9%, 1% y 0,3%, respectivamente). El análisis histopatológico reveló que se trata de una discrasia de células plasmáticas, es decir, mielomas que van creciendo pudiendo llegar a ocupar toda la cavidad peritoneal y limitar la recolección y producción del lÃquido ascÃtico
