Caracterização funcional de genes codificadores de proteínas hipotéticas diferencialmente expressos durante a metaciclogenese de Trypanosoma Cruzi

identificação de um conjunto de aproximadamente 1.000 genes diferencialmente expressos,
em sua maioria proteínas hipotéticas. Visando aumentar o conhecimento sobre a função
desses genes como a compreensão da expressão, função e regulação dos mesmos, com ênfase
nas proteínas hipotéticas, iniciou-se o presente trabalho. Inicialmente, sete proteínas
hipotéticas conservadas foram selecionadas, com base em sua confiabilidade de expressão
diferencial. Posteriormente, foi realizada uma clusterização hierárquica com diferentes dados
de metaciclogênese, para aumentar o grau de informação com relação aos padrões de
expressão. Dentro de cada cluster dos genes previamente selecionados, foram identificados
mais três genes distintos, codificando outra proteína hipotética conservada (gene secundário),
uma proteína hipotética (gene acessório) e uma proteína com função conhecida (gene guia).
Esses critérios visam aumentar a quantidade de proteínas hipotéticas sendo caracterizadas,
bem como ter elementos de referência para futuras análises. Estes 28 genes foram clonados
em um vetor de entrada (pDONR; Invitrogen), a partir do qual é possível inserir o gene de
interesse em outros vetores, os quais apresentam propriedades diferentes. Inicialmente,
realizamos a expressão em E. coli utilizando vetores de destinação (pDEST17, Invitrogen)
para obtenção de proteínas recombinantes. Dos 28 genes selecionados foram obtidos 27
clones de entrada, os quais foram totalmente transferidos para o pDEST17. Na etapa de
expressão, foram obtidas 19 proteínas, todas insolúveis, procedendo-se à obtenção de
anticorpos policlonais, através da inoculação em camundongos, sendo que obteve-se soros
para o reconhecimento de 18 proteínas recombinantes. Ao se realizar western blot para a
comprovação da expressão diferencial, a proteína nativa foi identificada adequadamente por
onze anticorpos, e o padrão de expressão diferencial, juntamente com outros 12 soros
existente no instituto, em sua grande maioria corrobora os resultados do microarranjo. Foram
verificados padrões de imunolocalização para 9 proteínas sendo 3 hipotéticas conservadas e 6
de função conhecida. Outros resultados obtidos por este trabalho foi o incremento nas
informações sobre os genes, com relação a aspectos bioinformáticos, possibilitando uma
melhor anotação e compreensão dos mesmos.Microarray analysis of T. cruzi metacyclogenesis resulted in the identification of
approximately 1,000 differentially expressed genes and several of them encode hypothetical
proteins. Aiming to increase the knowledge about the function of these genes, a better
understanding of the expression, function and regulation, with an emphasis on hypothetical
proteins, we have devised this work. Initially, seven conserved hypothetical proteins were
selected on basis of differential expression confiability. Subsequently, hierarchical clustering
was applied to the metacyclogenesis data and, within each cluster of genes previously
selected, were identified other three distinct genes, encoding another conserved hypothetical
protein (secondary gene), a hypothetical protein (accessory gene) and a protein with known
function (guide gene). This approach increases the expectation of having at least one
hypothetical protein better characterized, and also provides reference elements for future
analysis. These 28 genes were cloned in entry vector of the Gateway platform (pDONR;
Invitrogen), from which it is possible to insert the selected gene in other vectors, with
different functional characterization capabilities. Initially, the 28 selected genes were
expressed as proteins in E. Coli using appropriate destination vectors (pDEST17, Invitrogen),
obtaining a recombinant protein. Of the 28 selected genes, 27 entry clones were obtained,
which have been transferred to pDEST17. In the expression stage, we have obtained 19
proteins, all insoluble, which were used for polyclonal antibody production through
inoculation in mice, and we have obtained sera recognizing 18 recombinant proteins. Western
blot assay was performed aiming the differential expression analysis. The native protein was
identified properly by 11 antibodies, and the differential expression pattern, including other
12 sera available in the Institute, mostly corroborates the microarray results. Nine protein
were immunolocalized, 3 hypothetical protein and 6 of known function. Other results of this
work were a better annotation, using bioinformatics tools, producing an increase in the
functional knowledge of these proteins
A high-throughput cloning system for reverse genetics in Trypanosoma cruzi
<p>Abstract</p> <p>Background</p> <p>The three trypanosomatids pathogenic to men, <it>Trypanosoma cruzi</it>, <it>Trypanosoma brucei </it>and <it>Leishmania major</it>, are etiological agents of Chagas disease, African sleeping sickness and cutaneous leishmaniasis, respectively. The complete sequencing of these trypanosomatid genomes represented a breakthrough in the understanding of these organisms. Genome sequencing is a step towards solving the parasite biology puzzle, as there are a high percentage of genes encoding proteins without functional annotation. Also, technical limitations in protein expression in heterologous systems reinforce the evident need for the development of a high-throughput reverse genetics platform. Ideally, such platform would lead to efficient cloning and compatibility with various approaches. Thus, we aimed to construct a highly efficient cloning platform compatible with plasmid vectors that are suitable for various approaches.</p> <p>Results</p> <p>We constructed a platform with a flexible structure allowing the exchange of various elements, such as promoters, fusion tags, intergenic regions or resistance markers. This platform is based on Gateway<sup>® </sup>technology, to ensure a fast and efficient cloning system. We obtained plasmid vectors carrying genes for fluorescent proteins (green, cyan or yellow), and sequences for the <it>c-myc </it>epitope, and tandem affinity purification or polyhistidine tags. The vectors were verified by successful subcellular localization of two previously characterized proteins (<it>Tc</it>Rab7 and PAR 2) and a putative centrin. For the tandem affinity purification tag, the purification of two protein complexes (ribosome and proteasome) was performed.</p> <p>Conclusions</p> <p>We constructed plasmids with an efficient cloning system and suitable for use across various applications, such as protein localization and co-localization, protein partner identification and protein expression. This platform also allows vector customization, as the vectors were constructed to enable easy exchange of its elements. The development of this high-throughput platform is a step closer towards large-scale trypanosome applications and initiatives.</p
