Obtenció de materials estereocomplexes de PLA i estudi de propietats

L’àcid polilàctic o PLA és un polièster termoplàstic que s’ha estudiat durant més de quaranta anys, en l’àmbit científic i industrial, ja que presenta bones propietats d’enginyeria. La seva característica més interessant és la biodegradabilitat i que prové de recursos naturals . Tot i això, presenta una sèrie de deficiències com una temperatura de transició vítria baixa i una resistència térmica dolenta que ha limitat l’ús del biopolímer en algunes aplicacions. L’àcid polilàctic presenta dues estructures enantiomèriques degut a la presència del carboni quiral del monòmer, l’àcid làctic. La mescla dels dos homopolímers porten a una estereocomplexació, que obté una nova estructura cristal·lina capaç d’augmentar 50ºC el punt de fusió, aportant així un augment en la resistència tèrmica. Aquest treball té com a objectiu obtenir una gran quantitat d’estereocomplex de PLA (sc-PLA) d’alt pes molecular per precipitació en dissolució a partir d’una mescla equimolar de PDLA i PLLA amb dioxà a 25ºC i 80ºC. Les dues obtencions es van dur a terme a partir d’una mescla equimolar de PDLA i PLLA d’alt pes molecular dissoltes amb dioxà per una concentració aproximada del 10g/dL. Per fer un seguiment del procés, diàriament es va prendre mostres del sistema de reacció a cada temperatura i així obtener dues fases: la fase precipitada (o insoluble) i una fase líquida on el PLA restava en dissolució (fase soluble). Les fases es separaven per centrifugació i es caracteritzaven per espectroscòpia IR, quantitat de producte i, en alguns casos, calorimetria diferencial d’escombrat (DSC). S’ha obtingut una cinètica per a cada temperatura de la formació d’estereocomplex de PLA a partir del percentatges de fase insoluble obtinguts diàriament. L’estructura química s’ha estudiat per espectroscòpia d’infraroig (IR) i s’ha realitzat una calorimetria diferencial d’escombrat per veure la cristal·linitat dels productes. A més, s’ha realitzat una cromatografia GPC per observar el pes molecular de l’estereocomplex de PLA del producte final. També s’ha utilitzat la microscòpia electrònica de rastreig (SEM) per estudiar la morfologia de les partícules finals de l’estereocomplex de PLA en pols. La caracterització ha mostrat que, a ambdues temperatures s’ha format una quantitat significativa d’estereocomplex d’alta cristal·linitat , concretament 60 % i 73 %, a 25ºC i 80ºC respectivament. L’estereocomplex obtingut a 80ºCpresenta descomposició química, tan com s’ha comprovat per GPC i per espectroscòpia IR. S’han obtingut imatges de SEM que mostren morfologies diferents en les partícules obtingudes a cada temperatur

Photosynthetic and growth response of freshwater picocyanobacteria are strain-specific and sensitive to photoacclimation

We investigated the effect of different light conditions on primary production and growth rates of three closely related freshwater picocyanobacterial strains from three different ribotypes in laboratory cultures. The primary goal was to test whether not only different pigment types (PC-rich versus PE-rich) but also other physiological characteristics suggested by different phylogenetic positions could affect growth and photosynthetic rates of picocyanobacteria. Secondly, we tested whether photacclimation is strain specific. Experiments were conducted over light intensities ranging from 6 to 1500 μmol photons m−2 s−1 with cultures that were acclimated to low (10 μmol photons m−2 s−1) and moderate (100 μmol photons m−2 s−1) irradiance. The PE-rich strain was sensitive to high light conditions and reached highest photosynthesis and growth rates at low light intensities. The relative effect of photoacclimation was different between the two PC-rich strains, with one strain showing only moderate changes in growth rates in response to the light level used during the acclimation period. Overall, growth rates differed widely in response to light intensity and photoacclimation. Photoacclimation significantly affected both primary production and growth rates of all three strains investigated. We conclude that strain-specific photoacclimation adds to the niche partitioning among closely related freshwater picocyanobacteria

Vertical Distribution of Epibenthic Freshwater Cyanobacterial Synechococcus spp. Strains Depends on Their Ability for Photoprotection

Epibenthic cyanobacteria often grow in environments where the fluctuation of light intensity and quality is extreme and frequent. Different strategies have been developed to cope with this problem depending on the distribution of cyanobacteria in the water column. and either constant or enhanced levels of carotenoids were assayed in phycocyanin-rich strains collected from 1.0 and 0.5 m water depths. Protein analysis revealed that while the amount of biliproteins remained constant in all strains during light stress and recovery, the amount of D1 protein from photosystem II reaction centre was strongly reduced under light stress conditions in strains from 7.0 m and 1.0 m water depth, but not in strains collected from 0.5 m depth. spp. strains, depending on their genetically fixed mechanisms for photoprotection
