    Lyn kinase in Basal breast cancers

    Breast cancer is extremely heterogeneous, to the extent that some consider it to be composed of distinct diseases. Over the last 10 years, research has driven a new classification of Breast cancers, based on the expression of different molecular markers. This newer system is able to divide Breast cancers into at least 5 different subgroups. One subgroup, the ‘Basal breast cancers’ is of particular concern due to its younger age at diagnosis, increased aggressiveness, shorter survival and lack of targeted therapies. Studies have found that cell lines representing this subtype are sensitive to the multikinase inhibitor ‘Dasatinib’. This suggests that kinase/s targeted by Dasatinib may be overexpressed or deregulated in Basal breast cancers, and may thus represent novel therapeutic targets. To investigate this hypothesis, mRNA and protein expression of some of the known Dasatinib kinase targets was compared between cell lines representing Basal breast cancer and the less aggressive Luminal breast cancer. One candidate kinase, Lyn, was significantly overexpressed at the mRNA and protein level in Basal versus Luminal breast cancer lines, and exhibited increased activity in the Basal subgroup. To investigate the mechanisms regulating Lyn activation, cell lines were stimulated with candidate growth factors. HGF stimulated Lyn activity in one cell line suggesting that Lyn forms part of the Met signalling pathway. However, Lyn knock down did not affect the activity or total levels of signalling proteins comprising the Met pathway, nor did it affect HGF-induced cell scattering. Interestingly, Lyn depletion did affect the morphology of cells upon starvation, with cells becoming more dissociated and fibroblastic. Lastly, the translational relevance of these findings was tested by investigating Lyn expression in a Breast cancer patient cohort by immunohistochemical staining. Lyn was found to be significantly overexpressed in the Basal subgroup of patients and strongly associated with it. In addition, Lyn expression was associated with poor prognosis. Thus, while the functional role of Lyn in Basal breast cancer requires further characterisation, it represents a novel biomarker of the Basal breast cancer subgroup and may represent a therapeutic target in Basal breast cancer cells

    Waste-to-energy processes: What is the impact on air pollutants and health?

    Key points • Only half of the waste that Australia generates each year is recycled. Australian governments have committed to better waste management. While there is an increased demand for energy, greenhouse gas emissions must be curbed to mitigate climate change. • The Clean Energy Financing Corporation waste hierarchy highlights that the first step in tackling Australia’s waste problem should be to reduce the amount of waste produced before considering methods for recovery (re-purpose, recycle). • Waste-to-Energy (WtE) processes, near the bottom of the waste hierarchy, convert sorted municipal and industrial solid waste into electricity and/or heat. • WtE involves more complete combustion, therefore lower emissions (and higher amounts of electricity per weight) of pollutants, than incinerating unsorted municipal solid waste. • A recent assessment of the effects of inhaling combustion emissions showed WtE processes had a 64% lower health risk than incinerating unsorted municipal waste. However, increased concentration of some items being incinerated may lead to greater emissions of carcinogens if appropriate management of emissions is not performed. • A full assessment of the environmental impacts of WtE processes should be considered on a case-by-case basis to evaluate the risks and benefits of pollutant emissions and less obvious potential impacts on the local and wider community, within the life cycle of WtE and alternative/traditional processes

    Waste-to-Energy processes: what is the impact on air pollutants and health? A critical review of the literature

    Background: Worldwide, governments are committing to better waste management. In Australia, only half of the waste generated each year is recycled. Meanwhile, there is increased demand for energy, while also reducing greenhouse gas emissions for climate change mitigation. While recovering energy (as electricity or heat) from waste is an end use in the waste hierarchy scheme, it has a number of economic and environmental benefits. The health benefits (or risks) of this recovery, however, are less clear.Methods: To summarise current understanding of the health benefits/risks with Waste-to-Energy (WtE) processes, a critical literature review was performed using Google Scholar. Search terms and Boolean operators used were "energy from waste" OR "waste to energy" AND "air pollution" OR "air quality" and health. Patents and citations were excluded. Further, inclusion criteria for papers were: (a) published in a peer-review, open-access journal; (b) published up to and including the year 2018, and; (c) published in English.Results: We found that WtE health impacts have been little studied. Several earlier? studies have assessed the health implications of waste incineration, however evidence suggests that WtE may have less health impact compared to waste incineration or fossil fuel combustion. The most comprehensive assessment to-date of the effects of combustion emissions showed WtE processes had a 64% lower health risk than incinerating unsorted municipal waste, but the increased concentration of some items in incinerated waste may lead to greater emissions of carcinogens. As well as better/more accurate methods to assign exposures, life-cycle analyses are needed to evaluate the impacts of WtE to human health, and how it compares to other methods of waste reduction and disposal in the waste hierarchy.Conclusions: A full assessment of environmental impacts of waste management activities associated with WtE should consider direct pollutant emissions and conversely, the positive effects of using this waste stream for energy generation, especially when benefits may outweigh the risks as is possible with modern, properly managed WtE facilities. As such, a life-cycle assessment of waste streams including reducing, recycling and reusing should be performed as a next step.<br/

    Policy implications for protecting health from the hazards of fire smoke. A panel discussion report from the workshop landscape fire smoke : Protecting health in an era of escalating fire risk

    Globally, and nationally in Australia, bushfires are expected to increase in frequency and intensity due to climate change. To date, protection of human health from fire smoke has largely relied on individual-level actions. Recent bushfires experienced during the Australian summer of 2019–2020 occurred over a prolonged period and encompassed far larger geographical areas than previously experienced, resulting in extreme levels of smoke for extended periods of time. This particular bushfire season resulted in highly challenging conditions, where many people were unable to protect themselves from smoke exposures. The Centre for Air pollution, energy and health Research (CAR), an Australian research centre, hosted a two-day symposium, Landscape Fire Smoke: Protecting health in an era of escalating fire risk, on 8 and 9 October 2020. One component of the symposium was a dedicated panel discussion where invited experts were asked to examine alternative policy settings for protecting health from fire smoke hazards with specific reference to interventions to minimise exposure, protection of outdoor workers, and current systems for communicating health risk.     Tyrosine phosphorylation profiling reveals the signaling network characteristics of basal breast cancer cells

    To identify therapeutic targets and prognostic markers for basal breast cancers, breast cancer cell lines were subjected to mass spectrometry-based profiling of protein tyrosine phosphorylation events. This revealed that luminal and basal breast cancer cells exhibit distinct tyrosine phosphorylation signatures that depend on pathway activation as well as protein expression. Basal breast cancer cells are characterized by elevated tyrosine phosphorylation of Met, Lyn, EphA2, epidermal growth factor receptor (EGFR), and FAK, and Src family kinase (SFK) substrates such as p130Cas. SFKs exert a prominent role in these cells, phosphorylating key regulators of adhesion and migration and promoting tyrosine phosphorylation of the receptor tyrosine kinases EGFR and Met. Consistent with these observations, SFK inhibition attenuated cellular proliferation, survival, and motility. Basal breast cancer cell lines exhibited differential responsiveness to small molecule inhibitors of EGFR and Met that correlated with the degree of target phosphorylation, and reflecting kinase coactivation, inhibiting two types of activated network kinase (e. g., EGFR and SFKs) was more effective than single agent approaches. FAK signaling enhanced both proliferation and invasion, and Lyn was identified as a proinvasive component of the network that is associated with a basal phenotype and poor prognosis in patients with breast cancer. These studies highlight multiple kinases and substrates for further evaluation as therapeutic targets and biomarkers. However, they also indicate that patient stratification based on expression/activation of drug targets, coupled with use of multi-kinase inhibitors or combination therapies, may be required for effective treatment of this breast cancer subgroup.
