Transport properties of dense deuterium-tritium plasmas
Consistent descriptions of the equation of states, and information about
transport coefficients of deuterium-tritium mixture are demonstrated through
quantum molecular dynamic (QMD) simulations (up to a density of 600 g/cm
and a temperature of eV). Diffusion coefficients and viscosity are
compared with one component plasma model in different regimes from the strong
coupled to the kinetic one. Electronic and radiative transport coefficients,
which are compared with models currently used in hydrodynamic simulations of
inertial confinement fusion, are evaluated up to 800 eV. The Lorentz number is
also discussed from the highly degenerate to the intermediate region.Comment: 4 pages, 3 figure
Highly efficient in vitro adventitious shoot regeneration of Adenosma glutinosum (Linn.) Druce using leaf explants
Adenosma glutinosum (Linn.) Druce is an important aromatic plant, but no information is available regarding its regeneration, callus induction and proliferation from leaf explants. In this study, an in vitro shoot regeneration procedure was developed for native A. glutinosum using leaf explants. Callus induction and shoots regeneration from leaf explants was evaluated on Murashige and Skoog (MS) media supplemented with combinations of 6-benzylaminopurine (6-BA) and α-naphthaleneacetic acid (NAA). Callus induction in all 16 treatments exceeded 95%, and the highest adventitious shoot number per callus (7.22 shoots per explant) was obtained when leaf explants were cultured on MS medium supplemented with 0.5 mg• L-1 6-BA, 0.1 mg• L-1 NAA, 3% sucrose and 0.72% agar. The highest shoots strengthening were obtained when adventitious buds were cultured on half-strength MS medium supplemented with 0.3 mg• L-1 NAA, 3% sucrose, 1.0 g• L-1 active carbon and 0.72% agar. The highest total root number (45.2) and root length (43.3 cm) were obtained when adventitious buds were cultured on half-strength MS medium supplemented with 0.0 mg• L-1 NAA, 3% sucrose, 1.0 g L−1 active carbon and 0.72% agar, while the highest total root surface area (4.1 cm2) and total root volume (114.1 mm3) were obtained when adventitious buds were cultured on half-strength MS medium supplemented with 0.5 mg• L-1 NAA, 3% sucrose, 1.0 g• L-1 active carbon and 0.72% agar. The efficient plant regeneration system developed here will be helpful for rapid micropropagation and further genetic improvement in A. glutinosum. Risk prediction models for postoperative delirium in elderly patients with hip fracture: a systematic review
ObjectivesTo systematically evaluate the risk prediction models for postoperative delirium in older adult hip fracture patients.MethodsRisk prediction models for postoperative delirium in older adult hip fracture patients were collected from the Cochrane Library, PubMed, Web of Science, and Ovid via the internet, covering studies from the establishment of the databases to March 15, 2023. Two researchers independently screened the literature, extracted data, and used Stata 13.0 for meta-analysis of predictive factors and the Prediction Model Risk of Bias Assessment Tool (PROBAST) to evaluate the risk prediction models for postoperative delirium in older adult hip fracture patients, evaluated the predictive performance.ResultsThis analysis included eight studies. Six studies used internal validation to assess the predictive models, while one combined both internal and external validation. The Area Under Curve (AUC) for the models ranged from 0.67 to 0.79. The most common predictors were preoperative dementia or dementia history (OR = 3.123, 95% CI 2.108–4.626, p < 0.001), American Society of Anesthesiologists (ASA) classification (OR = 2.343, 95% CI 1.146–4.789, p < 0.05), and age (OR = 1.615, 95% CI 1.387–1.880, p < 0.001). This meta-analysis shows that these were independent risk factors for postoperative delirium in older adult patients with hip fracture.ConclusionResearch on the risk prediction models for postoperative delirium in older adult hip fracture patients is still in the developmental stage. The predictive performance of some of the established models achieve expectation and the applicable risk of all models is low, but there are also problems such as high risk of bias and lack of external validation. Medical professionals should select existing models and validate and optimize them with large samples from multiple centers according to their actual situation. It is more recommended to carry out a large sample of prospective studies to build prediction models.Systematic review registrationThe protocol for this systematic review was published in the International Prospective Register of Systematic Reviews (PROSPERO) under the registered number CRD42022365258
Construction and validation of a machine learning model for the diagnosis of juvenile idiopathic arthritis based on fecal microbiota
PurposeHuman gut microbiota has been shown to be significantly associated with various inflammatory diseases. Therefore, this study aimed to develop an excellent auxiliary tool for the diagnosis of juvenile idiopathic arthritis (JIA) based on fecal microbial biomarkers.MethodThe fecal metagenomic sequencing data associated with JIA were extracted from NCBI, and the sequencing data were transformed into the relative abundance of microorganisms by professional data cleaning (KneadData, Trimmomatic and Bowtie2) and comparison software (Kraken2 and Bracken). After that, the fecal microbes with high abundance were extracted for subsequent analysis. The extracted fecal microbes were further screened by least absolute shrinkage and selection operator (LASSO) regression, and the selected fecal microbe biomarkers were used for model training. In this study, we constructed six different machine learning (ML) models, and then selected the best model for constructing a JIA diagnostic tool by comparing the performance of the models based on a combined consideration of area under receiver operating characteristic curve (AUC), accuracy, specificity, F1 score, calibration curves and clinical decision curves. In addition, to further explain the model, Permutation Importance analysis and Shapley Additive Explanations (SHAP) were performed to understand the contribution of each biomarker in the prediction process.ResultA total of 231 individuals were included in this study, including 203 JIA patients and Non-JIA individuals. In the analysis of diversity at the genus level, the alpha diversity represented by Shannon value was not significantly different between the two groups, while the belt diversity was slightly different. After selection by LASSO regression, 10 fecal microbe biomarkers were selected for model training. By comparing six different models, the XGB model showed the best performance, which average AUC, accuracy and F1 score were 0.976, 0.914 and 0.952, respectively, thus being used to construct the final JIA diagnosis model.ConclusionA JIA diagnosis model based on XGB algorithm was constructed with excellent performance, which may assist physicians in early detection of JIA patients and improve the prognosis of JIA patients
Molecular characterization of immunogenic cell death indicates prognosis and tumor microenvironment infiltration in osteosarcoma
IntroductionOsteosarcoma (OS) is a highly aggressive bone malignancy with a poor prognosis, mainly in children and adolescents. Immunogenic cell death (ICD) is classified as a type of programmed cell death associated with the tumor immune microenvironment, prognosis, and immunotherapy. However, the feature of the ICD molecular subtype and the related tumor microenvironment (TME) and immune cell infiltration has not been carefully investigated in OS.MethodsThe ICD-related genes were extracted from previous studies, and the RNA expression profiles and corresponding data of OS were downloaded from The Cancer Genome Atlas and Gene Expression Omnibus database. The ICD-related molecular subtypes were classed by the "ConsensusclusterPlus" package and the construction of ICD-related signatures through univariate regression analysis. ROC curves, independent analysis, and internal validation were used to evaluate signature performance. Moreover, a series of bioinformatic analyses were used for Immunotherapy efficacy, tumor immune microenvironments, and chemotherapeutic drug sensitivity between the high- and low-risk groups.ResultsHerein, we identified two ICD-related subtypes and found significant heterogeneity in clinical prognosis, TME, and immune response signaling among distinct ICD subtypes. Subsequently, a novel ICD-related prognostic signature was developed to determine its predictive performance in OS. Also, a highly accurate nomogram was then constructed to improve the clinical applicability of the novel ICD-related signature. Furthermore, we observed significant correlations between ICD risk score and TME, immunotherapy response, and chemotherapeutic drug sensitivity. Notably, the in vitro experiments further verified that high GALNT14 expression is closely associated with poor prognosis and malignant progress of OS.DiscussionHence, we identified and validated that the novel ICD-related signature could serve as a promising biomarker for the OS's prognosis, chemotherapy, and immunotherapy response prediction, providing guidance for personalized and accurate immunotherapy strategies for OS
An experimental study on coal gas desorption laws with different particle size
Abstract Based on the self-designed coal containing methane gas desorption law experimental system and coal gas desorption kinetics law, the gas desorption law for different particle size coal under isothermal-isobaric condition are measured by combining combined simulation measurement with the theoretical analysis. The effect of particle size on coal gas desorption laws are obtained through fitting analysis on experimental data: 1.Under the same sorption and desorption conditions, coal particle size was inversely related to total desorption gas amount in the same period. 2. For coal with any particle size, the gas desorption amount firstly increased as time, and finally reached the maximum value, which was equal to the gas sorption amount unit per mass. 3. For coal with higher gas and coal outburst risk trend, the effect of particle size on initial desorption velocity and total desorption amount with time was less. 4. Within the limit particle size, the gas initial desorption velocity had a negative relationship with particle size. The LAMOST Survey of Background Quasars in the Vicinity of the Andromeda and Triangulum Galaxies -- II. Results from the Commissioning Observations and the Pilot Surveys
We present new quasars discovered in the vicinity of the Andromeda and
Triangulum galaxies with the LAMOST during the 2010 and 2011 observational
seasons. Quasar candidates are selected based on the available SDSS, KPNO 4 m
telescope, XSTPS optical, and WISE near infrared photometric data. We present
509 new quasars discovered in a stripe of ~135 sq. deg from M31 to M33 along
the Giant Stellar Stream in the 2011 pilot survey datasets, and also 17 new
quasars discovered in an area of ~100 sq. deg that covers the central region
and the southeastern halo of M31 in the 2010 commissioning datasets. These 526
new quasars have i magnitudes ranging from 15.5 to 20.0, redshifts from 0.1 to
3.2. They represent a significant increase of the number of identified quasars
in the vicinity of M31 and M33. There are now 26, 62 and 139 known quasars in
this region of the sky with i magnitudes brighter than 17.0, 17.5 and 18.0
respectively, of which 5, 20 and 75 are newly-discovered. These bright quasars
provide an invaluable collection with which to probe the kinematics and
chemistry of the ISM/IGM in the Local Group of galaxies. A total of 93 quasars
are now known with locations within 2.5 deg of M31, of which 73 are newly
discovered. Tens of quasars are now known to be located behind the Giant
Stellar Stream, and hundreds behind the extended halo and its associated
substructures of M31. The much enlarged sample of known quasars in the vicinity
of M31 and M33 can potentially be utilized to construct a perfect astrometric
reference frame to measure the minute PMs of M31 and M33, along with the PMs of
substructures associated with the Local Group of galaxies. Those PMs are some

γH2AX foci formation in the absence of DNA damage: Mitotic H2AX phosphorylation is mediated by the DNA-PKcs/CHK2 pathway
AbstractPhosphorylated H2AX is considered to be a biomarker for DNA double-strand breaks (DSB), but recent evidence suggests that γH2AX does not always indicate the presence of DSB. Here we demonstrate the bimodal dynamic of H2AX phosphorylation induced by ionizing radiation, with the second peak appearing when G2/M arrest is induced. An increased level of γH2AX occurred in mitotic cells, and this increase was attenuated by DNA-PKcs inactivation or Chk2 depletion, but not by ATM inhibition. The phosphorylation-mimic CHK2-T68D abrogated the attenuation of mitotic γH2AX induced by DNA-PKcs inactivation. Thus, the DNA-PKcs/CHK2 pathway mediates the mitotic phosphorylation of H2AX in the absence of DNA damage

Liver-heart crosstalk controls IL-22 activity in cardiac protection after myocardial infarction.
Interleukin (IL)-22 regulates tissue inflammation and repair. Here we report participation of the liver in IL-22-mediated cardiac repair after acute myocardial infarction (MI). Methods: We induced experimental MI in mice by ligation of the left ascending artery and evaluated the effect of IL-22 on post-MI cardiac function and ventricular remodeling. Results: Daily subcutaneous injection of 100 µg/kg mouse recombinant IL-22 for seven days attenuated adverse ventricular remodeling and improved cardiac function in mice at 28 days after left anterior descending coronary artery ligation-induced MI. Pharmacological inhibition of signal transducer and activator of transcription (STAT3) muted these IL-22 activities. While cardiomyocyte-selective depletion of STAT3 did not affect IL-22 activities in protecting post-MI cardiac injury, hepatocyte-specific depletion of STAT3 fully muted these IL-22 cardioprotective activities. Hepatocyte-derived fibroblast growth factor (FGF21) was markedly increased in a STAT3-dependent manner following IL-22 administration and accounted for the cardioprotective benefit of IL-22. Microarray analyses revealed that FGF21 controlled the expression of cardiomyocyte genes that are involved in cholesterol homeostasis, DNA repair, peroxisome, oxidative phosphorylation, glycolysis, apoptosis, and steroid responses, all of which are responsible for cardiomyocyte survival. Conclusions: Supplementation of IL-22 in the first week after acute MI effectively prevented left ventricular dysfunction and heart failure. This activity of IL-22 involved crosstalk between the liver and heart after demonstrating a role of the hepatic STAT3-FGF21 axis in IL-22-induced post-MI cardiac protection
Design of the Anti-tuberculosis Drugs induced Adverse Reactions in China National Tuberculosis Prevention and Control Scheme Study (ADACS)
<p>Abstract</p> <p>Background</p> <p>More than 1 million tuberculosis (TB) patients are receiving the standard anti-TB treatment provided by China National Tuberculosis Prevention and Control Scheme (CNTS) in China every year. Adverse reactions (ADRs) induced by anti-TB drugs could both do harm to patients and lead to anti-TB treatment failure. The ADACS aimed to explore ADRs' incidences, prognoses, economical and public health impacts for TB patients and TB control, and build a DNA bank of TB patients.</p> <p>Methods/Design</p> <p>Multiple study designs were adopted. Firstly, a prospective cohort with 4488 sputum smears positive pulmonary tuberculosis patients was established. Patients were followed up for 6-9 months in 52 counties of four regions. Those suspected ADRs should be checked and confirmed by Chinese State Food and Drug Administration (SFDA). Secondly, if the suspected ADR was anti-TB drug induced liver injury (ATLI), a nested case-control study would be performed which comprised choosing a matched control and doing a plus questionnaire inquiry. Thirdly, health economical data of ADRs would be collected to analyze financial burdens brought by ADRs and cost-effectiveness of ADRs' treatments. Fourthly, a drop of intravenous blood for each patient was taken and saved in FTA card for DNA banking and genotyping. Finally, the demographic, clinical, environmental, administrative and genetic data would be merged for the comprehensive analysis.</p> <p>Discussion</p> <p>ADACS will give an overview of anti-TB drugs induced ADRs' incidences, risk factors, treatments, prognoses, and clinical, economical and public health impacts for TB patients applying CNTS regimen in China, and provide suggestions for individualized health care and TB control policy.</p
