    Energy Estimates and Gravitational Collapse

    We study the cancelations in the energy estimates for Einstein vacuum equations in order to prove the formation of black holes along evolutions. The novelty of the paper is that, we completely avoid using rotation vector fields to establish the global existence theorem of the solution. More precisely, we use only canonical null directions as commutators to derive energy estimates at the level of one derivatives of null curvature components.Comment: 2 figures, 40 page

    HCT116 colorectal liver metastases exacerbate muscle wasting in a mouse model for the study of colorectal cancer cachexia

    Colorectal cancer (CRC) is often accompanied by formation of liver metastases (LM) and skeletal muscle wasting, i.e. cachexia. Despite affecting the majority of CRC patients, cachexia remains underserved, understudied and uncured. Animal models for the study of CRC-induced cachexia, in particular models containing LM, are sparse; therefore, we aimed to characterize two new models of CRC cachexia. Male NSG mice were injected subcutaneously (HCT116) or intrasplenically (mHCT116) with human HCT116 CRC tumor cells to disseminate LM, whereas experimental controls received saline (n=5-8/group). Tumor growth was accompanied by loss of skeletal muscle mass (HCT116: -20%; mHCT116: -31%; quadriceps muscle) and strength (HCT116: -20%; mHCT116: -27%), with worsened loss of skeletal muscle mass in mHCT116 compared with HCT116 (gastrocnemius: -19%; tibialis anterior: -22%; quadriceps: -21%). Molecular analyses revealed elevated protein ubiquitination in HCT116, whereas mHCT116 also displayed elevated Murf1 and atrogin-1 expression, along with reduced mitochondrial proteins PGC1α, OPA1, mitofusin 2 and cytochrome C. Further, elevated IL6 levels were found in the blood of mHCT116 hosts, which was associated with higher phosphorylation of STAT3 in skeletal muscle. To clarify whether STAT3 was a main player in muscle wasting in this model, HCT116 cells were co-cultured with C2C12 myotubes. Marked myotube atrophy (-53%) was observed, along with elevated phospho-STAT3 levels (+149%). Conversely, inhibition of STAT3 signaling by means of a JAK/STAT3 inhibitor was sufficient to rescue myotube atrophy induced by HCT116 cells (+55%). Overall, our results indicate that the formation of LM exacerbates cachectic phenotype and associated skeletal muscle molecular alterations in HCT116 tumor hosts

    TRIP13PCH-2 promotes Mad2 localization to unattached kinetochores in the spindle checkpoint response.

    The spindle checkpoint acts during cell division to prevent aneuploidy, a hallmark of cancer. During checkpoint activation, Mad1 recruits Mad2 to kinetochores to generate a signal that delays anaphase onset. Yet, whether additional factors contribute to Mad2's kinetochore localization remains unclear. Here, we report that the conserved AAA+ ATPase TRIP13(PCH-2) localizes to unattached kinetochores and is required for spindle checkpoint activation in Caenorhabditis elegans. pch-2 mutants effectively localized Mad1 to unattached kinetochores, but Mad2 recruitment was significantly reduced. Furthermore, we show that the C. elegans orthologue of the Mad2 inhibitor p31(comet)(CMT-1) interacts with TRIP13(PCH-2) and is required for its localization to unattached kinetochores. These factors also genetically interact, as loss of p31(comet)(CMT-1) partially suppressed the requirement for TRIP13(PCH-2) in Mad2 localization and spindle checkpoint signaling. These data support a model in which the ability of TRIP13(PCH-2) to disassemble a p31(comet)/Mad2 complex, which has been well characterized in the context of checkpoint silencing, is also critical for spindle checkpoint activation

    On Varieties of Ordered Automata

    The Eilenberg correspondence relates varieties of regular languages to pseudovarieties of finite monoids. Various modifications of this correspondence have been found with more general classes of regular languages on one hand and classes of more complex algebraic structures on the other hand. It is also possible to consider classes of automata instead of algebraic structures as a natural counterpart of classes of languages. Here we deal with the correspondence relating positive C\mathcal C-varieties of languages to positive C\mathcal C-varieties of ordered automata and we present various specific instances of this correspondence. These bring certain well-known results from a new perspective and also some new observations. Moreover, complexity aspects of the membership problem are discussed both in the particular examples and in a general setting

    Guessing a password over a wireless channel (on the effect of noise non-uniformity)

    A string is sent over a noisy channel that erases some of its characters. Knowing the statistical properties of the string's source and which characters were erased, a listener that is equipped with an ability to test the veracity of a string, one string at a time, wishes to fill in the missing pieces. Here we characterize the influence of the stochastic properties of both the string's source and the noise on the channel on the distribution of the number of attempts required to identify the string, its guesswork. In particular, we establish that the average noise on the channel is not a determining factor for the average guesswork and illustrate simple settings where one recipient with, on average, a better channel than another recipient, has higher average guesswork. These results stand in contrast to those for the capacity of wiretap channels and suggest the use of techniques such as friendly jamming with pseudo-random sequences to exploit this guesswork behavior.Comment: Asilomar Conference on Signals, Systems & Computers, 201
