--Klausurerfolg,Veranstaltungsteilnahme,E-Learning
Bactericidal Efficacy of Cold Plasma at Different Depths of Infected Root Canals In Vitro
Objectives: Cold plasma (CP) has been shown to be effective even against multiresistant microorganisms. As previous investigations on the effect of CP in root canals showed promising results, the aim of the present study was to analyze the bactericidal efficacy of CP in different depths of infected dentin. Methods: 32 standardized root canals of human mandibular premolars were infected with Enterococcus faecalis and incubated for one week. Specimens were randomly selected for one of four disinfection methods: control (5mL NaCl), 5mL chlorhexidine (CHX), CP alone (CP), and a combination of 5mL CHX and cold plasma (CHX+CP). CHX was ultrasonically activated for 30s, while cold plasma was used for 60s in the root canals. Dentin samples at depths of 300, 500 and 800 µm were obtained and diluted serially. Colony forming units (CFUs) were counted on agar plates after 24h of incubation. Results: The highest overall logarithmic reduction factors (RF) were obtained from CHX+CP (log RF 3.56 p<0.01; Mann-Whitney U test), followed by CP (log RF 3.27 p<0.01) and CHX alone (log RF 2.65 p<0.01) related to the control. All disinfection methods showed significantly lower CFU counts compared to the control group in 300 µm and 800 µm (both p<0.01, Kruskal-Wallis test). Discussion: The adjuvant use of CP might be beneficial in highly infected root canals to improved disinfection. However, the disinfection effect against Enterococcus faecalis of CP is comparable to ultrasonically activated CHX
usage of differences in the reaction of prokaryotes and eukaryotic cells to plasma for caries therapy
Plasma, also known as the 4th state of matter, is a partially or completely
ionized gas. Through its electrical conductivity it has a few specific
characteristics and is used as cold or warm plasma in medical fields. It’s not
completely researched if cold plasma, due to its antimicrobial effect, is able
to inactive carious processes and application could be indicated
prophylactically. This has been part of the present investigations. In this
context antimicrobial effect in root canals and on implant surfaces was
investigated as a progressing caries may require root canal treatment. After
tooth loss implant restoration could be a therapy option. For this purpose,
bacterial incubated samples (root surface, root canals, implants) were treated
with cold plasma (CP). Furthermore, in an ex vivo approach freshly extracted
teeth with carious lesions were treated with plasma. On carious dentin
surfaces combination of CHX + CP (2,67 log CFU/ml) showed significant better
results compared to CHX group (4,45 log CFU/ml). Moreover, plasma adjuvantly
used to scaling and root planning (SRP) on infected dentin surfaces led to a
significant difference compared to the control (K) (median SRP+CP: 0 log
CFU/ml, control: 3,85 log CFU/ml). In root canals reduction factors (RF) of
3,56 log RF (p<0,01) for CHX + CP, 3,27 log RF(p<0,01) for CP and 2,66 log RF
(p<0,01) for CHX were observed. Also, significant differences on implant
surfaces compared to laser usage (DL) were determined (K= 4,59 log CFU/ml, DL=
4 log CFU/ml, CP60= 2,38 log CFU/ml and CP120= 2,66 log CFU/ml). The usage of
plasma for caries therapy seems promising, based on the lower resistance of
prokaryotes compared to eukaryotic cells. However, further clinical studies
are needed to examine obtained results validity under oral cavity conditions
