Adiabatic Pair Creation
We give here the proof that pair creation in a time dependent potentials is
possible. It happens with probability one if the potential changes
adiabatically in time and becomes overcritical, that is when an eigenvalue
enters the upper spectral continuum. The potential may be assumed to be zero at
large negative and positive times. The rigorous treatment of this effect has
been lacking since the pioneering work of Beck, Steinwedel and Suessmann in

The TF Limit for Rapidly Rotating Bose Gases in Anharmonic Traps
Starting from the full many body Hamiltonian we derive the leading order
energy and density asymptotics for the ground state of a dilute, rotating Bose
gas in an anharmonic trap in the ` Thomas Fermi' (TF) limit when the
Gross-Pitaevskii coupling parameter and/or the rotation velocity tend to
infinity. Although the many-body wave function is expected to have a
complicated phase, the leading order contribution to the energy can be computed
by minimizing a simple functional of the density alone
Optical lattice quantum simulator for QED in strong external fields: spontaneous pair creation and the Sauter-Schwinger effect
Spontaneous creation of electron-positron pairs out of the vacuum due to a
strong electric field is a spectacular manifestation of the relativistic
energy-momentum relation for the Dirac fermions. This fundamental prediction of
Quantum Electrodynamics (QED) has not yet been confirmed experimentally as the
generation of a sufficiently strong electric field extending over a large
enough space-time volume still presents a challenge. Surprisingly, distant
areas of physics may help us to circumvent this difficulty. In condensed matter
and solid state physics (areas commonly considered as low energy physics), one
usually deals with quasi-particles instead of real electrons and positrons.
Since their mass gap can often be freely tuned, it is much easier to create
these light quasi-particles by an analogue of the Sauter-Schwinger effect. This
motivates our proposal of a quantum simulator in which excitations of
ultra-cold atoms moving in a bichromatic optical lattice represent particles
and antiparticles (holes) satisfying a discretized version of the Dirac
equation together with fermionic anti-commutation relations. Using the language
of second quantization, we are able to construct an analogue of the spontaneous
pair creation which can be realized in an (almost) table-top experiment.Comment: 21 pages, 10 figure
Detection of Peptide-Based Nanoparticles in Blood Plasma by ELISA
Aims The aim of the current study was to develop a method to detect peptide-linked nanoparticles in blood plasma. Materials & Methods A convenient enzyme linked immunosorbent assay (ELISA) was developed for the detection of peptides functionalized with biotin and fluorescein groups. As a proof of principle, polymerized pentafluorophenyl methacrylate nanoparticles linked to biotin-carboxyfluorescein labeled peptides were intravenously injected in Wistar rats. Serial blood plasma samples were analyzed by ELISA and by liquid chromatography mass spectrometry (LC/MS) technology. Results The ELISA based method for the detection of FITC labeled peptides had a detection limit of 1 ng/mL. We were able to accurately measure peptides bound to pentafluorophenyl methacrylate nanoparticles in blood plasma of rats, and similar results were obtained by LC/MS. Conclusions We detected FITC-labeled peptides on pentafluorophenyl methacrylate nanoparticles after injection in vivo. This method can be extended to detect nanoparticles with different chemical compositions
Male factors determining the outcome of intracytoplasmic sperm injection with epididymal and testicular spermatozoa
Summary. During a period of 8 years, 1079 intracytoplasmic sperm injection (ICSI) procedures with aspirated epididymal or testicular spermatozoa were performed. Epididymal spermatozoa were used in 172 cycles and testicular spermatozoa or spermatids in 907 cycles. Multiple biopsies were obtained from at least two different locations in the testes. Retrieved spermatozoa were used after cryopreservation (frozen) or immediately after aspiration (fresh). Three hundred patients had obstructive azoospermia (OA) or ejaculation failure. In 414 cases, azoospermia was caused by impaired spermatogenesis resulting from maldescended testes, chemotherapy/radiotherapy, or by Sertoli-cellonly syndrome, genetic disorders or unknown aetiology. Transfer rates, pregnancy rates and birth rates per ICSI cycle showed no statistically significant differences between testicular and epididymal spermatozoa in men with OA (28% average birth rates in both cases). However, birth rates differed significantly with regard to the status of spermatogenesis. Treatment of men with nonobstructive azoospermia (NOA) resulted in a birth rate of 19% per cycle. In all patient groups, there was no difference in the birth rates achieved with fresh and cryopreserved spermatozoa. While testicular volume, follicle-stimulating hormone level and age of the male patient are no statistically significant prognostic factors, the underlying cause of azoospermia is the most important factor determining the outcome of ICSI with epididymal and testicular spermatozoa. Identification of heat shock protein 32 (Hsp32) as a novel target in acute lymphoblastic leukemia
Heat shock proteins (Hsp) are increasingly employed as therapeutic targets in oncology. We have shown that Hsp32, also known as heme oxygenase-1 (HO-1), serves as survival factor and potential target in Ph+ chronic myeloid leukemia. We here report that primary cells and cell lines derived from patients with acute lymphoblastic leukemia (ALL) express Hsp32 mRNA and the Hsp32 protein in a constitutive manner. Highly enriched CD34+/CD38- ALL stem cells also expressed Hsp32. Two Hsp32-targeting drugs, pegylated zinc protoporphyrine (PEG-ZnPP) and styrene maleic acid-micelle-encapsulated ZnPP (SMA-ZnPP), induced apoptosis and growth arrest in the BCR/ABL1+ cell lines, in Ph- lymphoblastic cell lines and in primary Ph+ and Ph- ALL cells. The effects of PEG-ZnPP and SMA-ZnPP on growth of leukemic cells were dose-dependent. In Ph+ ALL, major growth-inhibitory effects of the Hsp32-targeting drugs were observed in imatinib-sensitive and imatinib-resistant cells. Hsp32-targeting drugs were found to synergize with imatinib, nilotinib, and bendamustine in producing growth inhibition and apoptosis in Ph+ ALL cells. A siRNA against Hsp32 was found to inhibit growth and survival of ALL cells and to synergize with imatinib in suppressing the growth of ALL cells. Body Composition as a Comorbidity-Independent Predictor of Survival following Nephroureterectomy for Urothelial Cancer of the Upper Urinary Tract
Radical nephroureterectomy (NUE) is the gold standard treatment for high-risk urothelial cancer of the upper urinary tract (UTUC). Besides sarcopenia and frailty, fat distribution is moving increasingly into focus. Components of body composition were assessed in patients undergoing NUE due to UTUC. The study cohort included 142 patients. By using CT-based measurements, the skeletal muscle index (SMI), subcutaneous adipose tissue index (SATI), and visceral adipose tissue index (VATI) were measured at the height of the third lumbar vertebra. Overall survival (OS) and cancer-specific survival (CSS) were estimated using univariable und multivariable Cox regression models. The prevalence of sarcopenia in the study population (n = 142) was 37%. OS and CSS were significantly reduced in sarcopenic patients. In the multivariable cox regression analysis, including age, ACE-27, T-stage, R-stage, LVI and necrosis, sarcopenia remained a significant risk factor of OS (HR, 1.77; 95% CI 1.02–3.07; p = 0.042) and CSS (HR, 2.17; 95% CI 1.18–3.99; p = 0.012). High visceral adipose tissue seems to be protective, although not statistically significant. Sarcopenia is a comorbidity-independent risk factor in patients who underwent NUE due to UTUC. Visceral fat represents a potentially protective factor. These results suggest that specific factors of body composition can be used for better risk stratification
Body Composition of Patients Undergoing Radical Cystectomy for Bladder Cancer: Sarcopenia, Low Psoas Muscle Index, and Myosteatosis Are Independent Risk Factors for Mortality
Abstract:
Background: We assessed a wide array of body composition parameters to identify those
most relevant as prognostic tools for patients undergoing radical cystectomy (RC) due to bladder
cancer (BC). Methods: In this retrospective, single-center study, preoperative computed tomography
(CT) scans of 657 patients were measured at the level of the 3rd lumbar vertebra (L3) to determine
common body composition indices including sarcopenia, myosteatosis, psoas muscle index (PMI),
subcutaneous and visceral fat index (SFI and VFI), visceral-to-subcutaneous fat ratio (VSR), and visceral
obesity. Predictors of overall survival (OS) and cancer-specific survival (CSS) were identified in
univariate and multivariate survival analysis. Results: Sarcopenia and a low PMI were independently
associated with shorter OS (Sarcopenia: HR 1.30; 95% CI 1.02–1.66; p = 0.04 and a low PMI: HR 1.32;
95% CI 1.02–1.70; p = 0.03) and CSS (Sarcopenia: HR 1.64; 95% CI 1.19–2.25; p < 0.01 and a low PMI:
HR 1.41; 95% CI 1.02–1.96; p = 0.04). Myosteatosis, measured as decreasing average Hounsfield units
of skeletal muscle, was an independent risk factor for OS (HR 0.98; 95% CI 0.97–1.00; p = 0.01) and
CSS (HR 0.98; 95% CI 0.96–1.00; p < 0.05). The assessed adipose tissue indices were not significant
predictors for OS and CSS. Conclusions: Sarcopenia, a low PMI, and myosteatosis are independent
predictors for OS and CSS in patients undergoing radical cystectomy for bladder cancer

Biallelic loss-of-function mutation in NIK causes a primary immunodeficiency with multifaceted aberrant lymphoid immunity
Primary immunodeficiency disorders enable identification of genes with crucial roles in the human immune system. Here we study patients suffering from recurrent bacterial, viral and Cryptosporidium infections, and identify a biallelic mutation in the MAP3K14 gene encoding NIK (NF- B-inducing kinase). Loss of kinase activity of mutant NIK, predicted by in silico analysis and confirmed by functional assays, leads to defective activation of both canonical and non-canonical NF- B signalling. Patients with mutated NIK exhibit B-cell lymphopenia, decreased frequencies of class-switched memory B cells and hypogammaglobulinemia due to impaired B-cell survival, and impaired ICOSL expression. Although overall T-cell numbers are normal, both follicular helper and memory T cells are perturbed. Natural killer (NK) cells are decreased and exhibit defective activation, leading to impaired formation of NK-cell immunological synapses. Collectively, our data illustrate the non-redundant role for NIK in human immune responses, demonstrating that loss-of-function mutations in NIK can cause multiple aberrations of lymphoid immunity
