    Impiego delle tecniche proteomiche nello studio del pattern proteico del siero di pazienti nefropatici con insufficienza renale cronica.

    RIASSUNTO L’Albumina (HSA) è la proteina più abbondante nel plasma della maggior parte delle specie,dai primati all’uomo e per questo è stata oggetto di grandi studi in passato. Ha una struttura semplice costituita da 585 aminoacidi organizzati in una struttura terziaria plastica grazie a 17 ponti disolfuro. Svolge una serie di funzioni fra cui il mantenimento della pressione oncotica del plasma,la regolazione del tono vascolare attraverso il trasporto e rilascio di NO,il trasporto di ormoni ed altre molecole biologiche quali la bilirubina e gli acidi grassi,il trasporto di metalli pesanti e ioni e la difesa contro i radicali dell’Ossigeno. Probabilmente la lista non è completa ed ulteriori studi permetteranno di aggiungere ulteriori dettagli. Queste funzioni sono consentite dalla plasticità di questa proteina che può realizzare una serie di microtransizioni cambiando la pKs dei gruppi ionizzabili in relazione al pH o al ligando con cui interagisce. Le possibili conformazioni che ne conseguono possono determinare diversi tipi di frammentazione. La variabilità di proteine e/o frammenti di Albumina del siero si riflettono nella variabilità dei contenuti proteici delle urine. Nella pratica clinica nefrologica lo studio di queste differenze è ampiamente impiegato a fini diagnostici. Lo scopo di questa tesi è l’individuazione di markers, soprattutto i frammenti di Albumina, in sieri patologici,utili ai fini di diagnosi precoce. La fase iniziale del lavoro di tesi è consistita nella messa a punto di un sistema che potesse mettere in risalto le proteine seriche che normalmente sono mascherate dall’Albumina. Questo risultato è stato ottenuto utilizzando una colonnina cibacron blue F3G-A che trattiene l’Albumina. Successivamente le proteine sono state separate attraverso la 2D-PAGE. Lo step successivo consiste nel confrontare i diversi pattern di espressione che si osservano nei pazienti affetti da malattie renali e nelle persone sane. Da un punto di vista biologico esiste un crescente interesse nello studio del metabolismo dell’Albumina e la scoperta di frammenti di questa proteina nei pazienti con insufficienza renale e la loro caratterizzazione può costituire un’importante base di partenza per fini diagnostici.     Myocardial perfusion in chronic diabetic mice by the up-regulation of pLKB1 and AMPK signaling

    Previous studies related impaired myocardial microcirculation in diabetes to oxidative stress and endothelial dysfunction. Thus, this study was aimed to determine the effect of up-regulating pAMPK-pAKT signaling on coronary microvascular reactivity in the isolated heart of diabetic mice. We measured coronary resistance in wild-type and streptozotocin (STZ)-treated mice, during perfusion pressure changes. Glucose, insulin, and adiponectin levels in plasma and superoxide formation, NOx levels and heme oxygenase (HO) activity in myocardial tissue were determined. In addition, the expression of HO-1, 3-nitrotyrosine, pLKB1, pAMPK, pAKT, and peNOS proteins in control and diabetic hearts were measured. Coronary response to changes in perfusion pressure diverged from control in a time-dependent manner following STZ administration. The responses observed at 28 weeks of diabetes (the maximum time examined) were mimicked by L-NAME administration to control animals and were associated with a decrease in serum adiponectin and myocardial pLKB1, pAMPK, pAKT, and pGSK-3 expression. Cobalt protoporphyrin treatment to induce HO-1 expression reversed the microvascular reactivity seen in diabetes towards that of controls. Up-regulation of HO-1 was associated with an increase in adiponectin, pLKB1, pAKT, pAMPK, pGSK-3, and peNOS levels and a decrease in myocardial superoxide and 3-nitrotyrosine levels. In the present study we describe the time course of microvascular functional changes during the development of diabetes and the existence of a unique relationship between the levels of serum adiponectin, pLKB1, pAKT, and pAMPK activation in diabetic hearts. The restoration of microvascular function suggests a new therapeutic approach to even advanced cardiac microvascular derangement in diabetes

    International audienceWe propose a reading of Kreisel's informal rigour as a dialectic interplay between a formal and an intuitive level in the study of logically relevant notions. To substantiate our reading, we provide two case-studies: Prawitz semantics and Walton's analysis of argument schemes. Prawitz's semantics suggests an interaction between informal presuppositions, confirmed and refined by results about formal setups, and formal setups, which can be improved by suitably formalising informal presuppositions. In Walton's Argumentation Theory, argument schemes are introduced to account for arguments that support their conclusion in a presumptive way. The possibility of developing such informal notion through the formal means provided by research in AI shows the importance of the interaction between these two levels in the analysis of defeasibility

    Aim Bacterial adherence to implants is considered to be an important event in the pathogenesis of bacterial infections. In fact, this infection process is a first stage of peri-implant mucositis and peri-implantitis, and a positive correlation has been found between oral hygiene and marginal bone loss around implants in the edentulous mandible. Surface properties of transgingival implant components are important determinants in bacterial adhesion. The purpose of this study was to characterize the biofilm formation, in vivo, on healing screws made of titanium alloy or coated with a combination of anatase and Bactercline® product. Materials and methods Twenty-five patients, between 21- 37 years, in excellent systemic health, participated in this study. In each of the 25 participants, one anatase-Bactercline® coated healing screw (Test) and one titanium alloy (TI6Al4V) healing screw (Control) were adapted to two different implants. Quantitative and qualitative biofilm formation on healing abutments was analyzed by culture method.Results Bacterial adherence to the two different healing screws used in this study were compared. Statistically significant differences were found between the Control and the Test group for both aerobic and anaerobic bacterial counts (p<0,05). The microflora consisted both of Gram-positive and Gram-negative bacteria, and displayed a high variability. The anaerobic S. intermedius, potentially “pathogenic”, was isolated only from the Control group. Both healing screws harbored primarily Gram-positive rods as Actinomyces spp, A. naeslundii, A. viscosus and the Gram-negative rods (Fusobacterium spp, Prevotella spp, Capnocythophaga spp) were mostly found on the Control healing screws.Conclusion Anatase-Bactercline® coated healing screws reduce the number of initially adhering bacteria, formed mainly of Gram-positive microorgnisms, while, on the contrary, the microflora covering the titanium alloy healing screws was, for the most part, Gram-negative
