    Re-engineering a nanodosimetry Monte Carlo code into Geant4: software design and first results

    A set of physics models for nanodosimetry simulation is being re-engineered for use in Geant4-based simulations. This extension of Geant4 capabilities is part of a larger scale R&D project for multi-scale simulation involving adaptable, co-working condensed and discrete transport schemes. The project in progress reengineers the physics modeling capabilities associated with an existing FORTRAN track-structure code for nanodosimetry into a software design suitable to collaborate with an object oriented simulation kernel.     Publication patterns in HEP computing

    An overview of the evolution of computing-oriented publications in high energy physics following the start of operation of LHC. Quantitative analyses are illustrated, which document the production of scholarly papers on computing-related topics by high energy physics experiments and core tools projects, and the citations they receive. Several scientometric indicators are analyzed to characterize the role of computing in high energy physics literature. Distinctive features of software-oriented and hardware-oriented scholarly publications are highlighted.     An analysis of publications related to high energy physics computing in refereed journals is presented. The distribution of papers associated to various fields of computing relevant to high energy physics is critically analyzed. The relative publication rate of software papers is evaluated in comparison to other closely related physics disciplines, such as nuclear physics, radiation protection and medical physics, and to hardware publications. The results hint to the fact that, in spite of the significant effort invested in high energy physics computing and its fundamental role in the experiments, this research area is underrepresented in scientific literature; nevertheless the analysis of citations highlights the significant impact of software publications in experimental research

    Unwanted hormonal and metabolic effects of postoperative adjuvant mitotane treatment for adrenocortical cancer

    Mitotane is widely used for the treatment of adrenocortical cancer (ACC), although the drug-related toxicity complicates its use. The aim of this study is to assess comprehensively the different endocrine and metabolic unwanted effects of the drug, and to provide data on the supportive therapies. We retrospectively analyzed 74 ACC patients adjuvantly treated with mitotane for ≥12 months. During the treatment period (40 months, 12–195), 32.4% of patients needed replacement therapy for mineralocorticoid deficit, 36.2% for hypothyroidism and 34.3% for male hypogonadism. In fertile women, hypogonadism was uncommon, while 65.4% of women developed ovarian cysts. Although no significant change in low-density lipoprotein (LDL) was observed, statins were started in 50% of patients for a significant increase in total cholesterol and triglycerides. Dyslipidemia occurred early, after a median time of 6 months from mitotane start. Conversely, testosterone replacement was usually started after >2 years. In many cases, ranging from 29.4% to 50% according to the side effect, toxicity occurred well before the achievement of the target mitotane concentrations. Supportive therapies were able to revert the biochemical alterations induced by mitotane, although higher doses were needed for a likely pharmacokinetic interaction of exogenous steroids and statins with mitotane. In conclusion, adjuvant mitotane therapy is associated with a spectrum of unwanted effects encompassing the function of different endocrine glands and requires a careful clinical and biochemical assessment associated with the therapeutic drug monitoring

    A quick pipeline for the isolation of 3D cell culture-derived extracellular vesicles

    Recent advances in cell biology research regarding extracellular vesicles have highlighted an increasing demand to obtain 3D cell culture-derived EVs, because they are considered to more accurately represent EVs obtained in vivo. However, there is still a grave need for efficient and tunable methodologies to isolate EVs from 3D cell cultures. Using nanofibrillar cellulose (NFC) scaffold as a 3D cell culture matrix, we developed a pipeline of two different approaches for EV isolation from cancer spheroids. A batch method was created for delivering high EV yield at the end of the culture period, and a harvesting method was created to enable time-dependent collection of EVs to combine EV profiling with spheroid development. Both these methods were easy to set up, quick to perform, and they provided a high EV yield. When compared to scaffold-free 3D spheroid cultures on ultra-low affinity plates, the NFC method resulted in similar EV production/cell, but the NFC method was scalable and easier to perform resulting in high EV yields. In summary, we introduce here an NFC-based, innovative pipeline for acquiring EVs from 3D cancer spheroids, which can be tailored to support the needs of variable EV research objectives.Peer reviewe
