Malarial Hemozoin Is a Nalp3 Inflammasome Activating Danger Signal

BACKGROUND: Characteristic symptoms of malaria include recurrent fever attacks and neurodegeneration, signs that are also found in patients with a hyperactive Nalp3 inflammasome. Plasmodium species produce a crystal called hemozoin that is generated by detoxification of heme after hemoglobin degradation in infected red blood cells. Thus, we hypothesized that hemozoin could activate the Nalp3 inflammasome, due to its particulate nature reminiscent of other inflammasome-activating agents. METHODOLOGY/PRINCIPAL FINDINGS: We found that hemozoin acts as a proinflammatory danger signal that activates the Nalp3 inflammasome, causing the release of IL-1beta. Similar to other Nalp3-activating particles, hemozoin activity is blocked by inhibiting phagocytosis, K(+) efflux and NADPH oxidase. In vivo, intraperitoneal injection of hemozoin results in acute peritonitis, which is impaired in Nalp3-, caspase-1- and IL-1R-deficient mice. Likewise, the pathogenesis of cerebral malaria is dampened in Nalp3-deficient mice infected with Plasmodium berghei sporozoites, while parasitemia remains unchanged. SIGNIFICANCE/CONCLUSIONS: The potent pro-inflammatory effect of hemozoin through inflammasome activation may possibly be implicated in plasmodium-associated pathologies such as cerebral malaria

Terrestrial Very-Long-Baseline Atom Interferometry:Workshop Summary

This document presents a summary of the 2023 Terrestrial Very-Long-Baseline Atom Interferometry Workshop hosted by CERN. The workshop brought together experts from around the world to discuss the exciting developments in large-scale atom interferometer (AI) prototypes and their potential for detecting ultralight dark matter and gravitational waves. The primary objective of the workshop was to lay the groundwork for an international TVLBAI proto-collaboration. This collaboration aims to unite researchers from different institutions to strategize and secure funding for terrestrial large-scale AI projects. The ultimate goal is to create a roadmap detailing the design and technology choices for one or more km-scale detectors, which will be operational in the mid-2030s. The key sections of this report present the physics case and technical challenges, together with a comprehensive overview of the discussions at the workshop together with the main conclusions

La préparation des peintures magdaléniennes des cavernes ariégeoises

ABSTRACT Analyses of Upper Palaeolithic paintings in some Ariège caves, particularly Niaux, show that Magdale- nian artists used a real paint. Their workshops had some elaborate recipes at their disposal and followed them scrupulously. The chronology of these recipes, based upon analyses of well-dated mobiliáry art works covered with paint, enables the authors to conclude that the Niaux cave was decorated over a much longer time than was previously thought.RESUME L'analyse de la matière picturale des oeuvres de la fin du Paléolithique supérieur ariégeois met en lumière la préparation d'une vraie peinture par les artistes magdaléniens. Les ateliers avaient à leur disposition des recettes élaborées qu'ils suivaient scrupuleusement. Les peintures schématiques postglaciaires du Sud-Est de la France : analyses pigmentaires

ABSTRACT The painted schematic art of the Chalcolithic period is the expression of its authors' philosophical beliefs but varies considerably according to local characteristics. The study of particularisms frequently allows an approach of ritual practices related to paintings. Analysis of the pigments choosen by the artists shows that there was a great diversity in drawing materials, even if the number of colours used was limited. Some modern over-paintings executed with lead and zinc have been detected.RÉSUMÉ L'art schématique peint daté du Chalcolithique exprime les conceptions philosophiques de ses auteurs tout en admettant de nombreuses variations à mettre au compte des particularismes locaux. L'étude de ces derniers permet souvent d'approcher les pratiques rituelles liées aux peintures. L'analyse des colorants choisis par les artistes montre une grande diversité de matériaux utilisés en dépit d'un nombre restreint de couleurs. La gravure sur os au Magdalénien : étude microscopique d'une côte de la grotte de La Vache (commune d'Alliat, Ariège)

International audienceFor several years research on mobiliary artefacts from the Paleolithic has been following a new path where the artist engraver's behaviour is more involved. We do believe that the engravings, and the information which may be associated with them, help us visualise the links between the artist's hand, the tool he used, and the line of the engraved figure. Our present work consists in the study of the techniques of engraving on bone based on good knowledge of the different materials involved (bone and flint). The aim of such research is to look for macromorphological singularities of the lines, to re-discover the hand of the engraver through them : all things that bear witness to the artist's know-how.L'étude de l'art mobilier paléolithique nous démontre, depuis quelques années, que la reconstitution du geste et du comportement du graveur est importante. Nous pensons, en effet, que les gravures et les informations qu'elles recèlent, nous aident à établir les relations existantes entre la main, l'outil et le trait. Dans ce but, nous recherchons les particularités morphologiques des tracés, uniquement sur os. Notre recherche, basée sur une connaissance préalable fine des divers matériaux mis en jeu (l'os — support de la gravure — et le silex — outil à graver — ), suit une double démarche : dans un premier temps, une phase expérimentale, pour comprendre l'interaction entre les matériaux et pour définir les différents paramètres de la technique artistique liés aux particularités morphologiques des tracés ; dans un second temps, nous procédons à l'application sur les objets archéologiques. Nous tentons ainsi d'approcher la main du graveur à travers la dynamique du trait afin de révéler le savoir-faire des artistes
