The influence of ecophysiological factors on growth, aflR gene expression and aflatoxin B1 production by a type strain of Aspergillus flavus
Maize is prone to infection by Aspergillus flavus, which can contaminate the product with aflatoxins. The objective of this study was to examine the impact that interactions between water activity (aw) and temperature may have on growth, the expression of a biosynthetic regulatory gene (aflR) and aflatoxin B1 (AFB1) production by a strain of A. flavus on a maize-based medium. Results showed that there were some differences between lag phases and growth rates of A. flavus. The optimum growth rate for A. flavus was at 30 °C and 0.99 aw. No growth occurred at 0.90 aw and 20 °C. Both temperature and aw had an influence on the relative aflR gene expression and AFB1 production by A. flavus; however, the results for AFB1 production were not consistent with the effects on gene expression or growth. These findings provide data that are useful to better understand the conditions which represent higher risks from AFB1 production. However, the aflR expression was not a good indicator of AFB1 production alone. Thus, further molecular studies of other AF-related genes should be done. These results are discussed in the context of harvesting and storage of maize and the prevailing environmental conditions to minimise AFB1 contamination
Virological Correlates of IgM–IgG Patterns of Response to SARS-CoV-2 Infection According to Targeted Antigens
The virological meaning of the different patterns of serology in COVID-19 has been little examined in clinical settings. Asymptomatic subjects with IgM-spike (S) and IgG-nucleocapsid (N) determinations by chemiluminescence were studied for SARS-CoV-2 shedding in respiratory secretions by transcription-mediated amplification (TMA). In subjects showing IgM-S positive and IgG-N negative, IgG-S was determined by lateral flow assay. A total of 712 individuals were tested: 30.0% presented IgM-S(+)/IgG-N(−), 25.8% had IgM-S(+)/IgG-N(+) and 44.2% had IgM-S(−)/IgG-N(+); the proportion with TMA(+) were comparable in these three groups: 12.1, 8.7 and 10.5%, respectively. In individuals with IgM-S(+)/IgG-N(−), IgG-S(+) was detected in 66.5%. The frequency of IgM-S(+)/IgG-S(−) in the total population was 10.0%, of whom 24.1% had TMA(+); the chances for TMA(+) in subjects with an IgM-S(+) alone pattern were 2.4%. Targeting of the same SARS-CoV-2 antigen seems to be better for the characterization of IgM/IgG patterns of response. Association Between Preexisting Versus Newly Identified Atrial Fibrillation and Outcomes of Patients With Acute Pulmonary Embolism
Background Atrial fibrillation (AF) may exist before or occur early in the course of pulmonary embolism (PE). We determined the PE outcomes based on the presence and timing of AF. Methods and Results Using the data from a multicenter PE registry, we identified 3 groups: (1) those with preexisting AF, (2) patients with new AF within 2 days from acute PE (incident AF), and (3) patients without AF. We assessed the 90-day and 1-year risk of mortality and stroke in patients with AF, compared with those without AF (reference group). Among 16 497 patients with PE, 792 had preexisting AF. These patients had increased odds of 90-day all-cause (odds ratio [OR], 2.81; 95% CI, 2.33-3.38) and PE-related mortality (OR, 2.38; 95% CI, 1.37-4.14) and increased 1-year hazard for ischemic stroke (hazard ratio, 5.48; 95% CI, 3.10-9.69) compared with those without AF. After multivariable adjustment, preexisting AF was associated with significantly increased odds of all-cause mortality (OR, 1.91; 95% CI, 1.57-2.32) but not PE-related mortality (OR, 1.50; 95% CI, 0.85-2.66). Among 16 497 patients with PE, 445 developed new incident AF within 2 days of acute PE. Incident AF was associated with increased odds of 90-day all-cause (OR, 2.28; 95% CI, 1.75-2.97) and PE-related (OR, 3.64; 95% CI, 2.01-6.59) mortality but not stroke. Findings were similar in multivariable analyses. Conclusions In patients with acute symptomatic PE, both preexisting AF and incident AF predict adverse clinical outcomes. Estrategias para el control de mohos toxigénicos en derivados cárnicos curado-madurados utilizando microorganismos y factores ambientales
Tesis doctoral con la mención de "Doctor Internacional"El desarrollo en los derivados cárnicos curado-madurados de mohos productores de micotoxinas y la consiguiente contaminación de estos productos puede suponer un problema de seguridad alimentaria. Por ello, es necesario desarrollar estrategias que limiten el desarrollo de mohos toxigénicos en su superficie durante el proceso de maduración. Para ello, en primer lugar, se validó un método de detección multitoxinas de HPLC-MS/MS y se confirmó que las micotoxinas difunden al interior de estos productos cárnicos. Además, se desarrolló un método de extracción y análisis basado en la metodología QuEChERS y uHPLC-MS/MS para la detección y cuantificación de ácido ciclopiazónico en productos cárnicos. En segundo lugar, se estudió una estrategia para minimizar el desarrollo de mohos toxigénicos evaluando el efecto de las condiciones ambientales (temperatura y actividad de agua) habituales en el proceso de elaboración de derivados cárnicos curado-madurados sobre el crecimiento, la expresión de genes implicados en la biosíntesis de las micotoxinas y su producción. A partir de estos datos se diseñaron modelos matemáticos de predicción de la concentración de micotoxinas y del momento en que se inicia su síntesis. Finalmente, se evaluó el efecto de agentes de biocontrol, como levaduras o mohos antifúngicos en el crecimiento y producción de micotoxinas. La información obtenida en esta Tesis Doctoral resultaría de gran utilidad en el contexto de la seguridad alimentaria ya que permitiría establecer acciones preventivas adecuadas en los sistemas APPCC de las industrias elaboradoras de derivados cárnicos curado-madurados para reducir el riesgo asociado con la acumulación de micotoxinas.The development of mycotoxin-producing moulds and the consequent contamination with mycotoxins in dry-cured meat products could be a food safety problem. Therefore, it is necessary to develop strategies that limit the growth of toxigenic moulds on its Surface during the ripening process. For this, in the first place, a multitoxin detection method by HPLC-MS/MS was validated and the mycotoxins diffuse into these meat products was confirmed. In addition, an extraction method based on the QuEChERS methodology and analysis by uHPLC-MS/MS for the detection and quantification of cyclopiazonic acid in meat products were developed. Secondly, a strategy to minimize the development of toxigenic moulds by evaluating the effect of the environmental conditions (temperatura and water activity) usual in the process of elaboration of dry-cured meat products on growth, the expression of genes involved in the biosynthesis of mycotoxins and their production were studied. From these data, mathematical models to predict the concentration of mycotoxins and the moment when their synthesis begins were designed. Finally, the effect of biocontrol agents, such as yeasts or antifungal moulds on the growth and production of mycotoxins was evaluated. Enterococcus faecium: a promising protective culture to control growth of ochratoxigenic moulds and mycotoxin production in dry-fermented sausages
Moulds positively contribute to the development of typical characteristic flavour and aroma of dry-fermented sausages. However, some mould species, such as Penicillium nordicum and Penicillium verrucosum, may contaminate this product with ochratoxin A (OTA). For this reason, the control of toxigenic moulds is needed. Strategies based on the use of antifungal microorganisms present in the native microbial population in the dry-fermented sausage processing could be a promising strategy. The aim of this work was to study the effect of Enterococcus faecium strains on P. nordicum and P. verrucosum growth and OTA production in a dry-fermented sausage-based medium at conditions of temperature and water activity similar to those occurring during the ripening of these meat products. Six strains were screened to evaluate their growth capacity and antifungal activity against P. nordicum and P. verrucosum at three fixed temperatures related to the sausage ripening. The two E. faecium strains that decreased growth of both species were chosen to further evaluate their effect on growth of P. verrucosum and P. nordicum and their mycotoxin production under conditions simulating the dry-fermented sausage ripening. The presence of E. faecium SE920 significantly reduced OTA production of P. nordicum although it did not affect P. verrucosum. Antifungal Effect of Autochthonous Aromatic Plant Extracts on Two Mycotoxigenic Strains of <i>Aspergillus flavus</i>
This study identified the compounds obtained from four native Dehesa plants, which were holm oak, elm, blackberry and white rockrose, and evaluated their ability to inhibit the growth and production of aflatoxins B1 and B2 of two strains of mycotoxigenic Aspergillus flavus. For this purpose, phenolic compounds present in the leaves and flowers of the plants were extracted and identified, and subsequently, the effect on the growth of A. flavus, aflatoxin production and the expression of a gene related to its synthesis were studied. Cistus albidus was the plant with the highest concentration of phenolic compounds, followed by Quercus ilex. Phenolic acids and flavonoids were mainly identified, and there was great variability among plant extracts in terms of the type and quantity of compounds. Concentrated and diluted extracts were used for each individual plant. The influence on mold growth was not very significant for any of the extracts. However, those obtained from plants of the genus Quercus ilex, followed by Ulmus sp., were very useful for inhibiting the production of aflatoxin B1 and B2 produced by the two strains of A. flavus. Expression studies of the gene involved in the aflatoxin synthesis pathway did not prove to be effective. The results indicated that using these new natural antifungal compounds from the Dehesa for aflatoxin production inhibition would be desirable, promoting respect for the environment by avoiding the use of chemical fungicides. However, further studies are needed to determine whether the specific phenolic compounds responsible for the antifungal activity of Quercus ilex and Ulmus sp. produce the antifungal activity in pure form, as well as to verify the action mechanism of these compounds
