USP45 and Spindly are part of the same complex implicated in cell migration
Abstract Ubiquitylation is a protein modification implicated in several cellular processes. This process is reversible by the action of deubiquinating enzymes (DUBs). USP45 is a ubiquitin specific protease about which little is known, aside from roles in DNA damage repair and differentiation of the vertebrate retina. Here, by using mass spectrometry we have identified Spindly as a new target of USP45. Our data show that Spindly and USP45 are part of the same complex and that their interaction specifically depends on the catalytic activity of USP45. In addition, we describe the type of ubiquitin chains associated with the complex that can be cleaved by USP45, with a preferential activity on K48 ubiquitin chain type and potentially K6. Here, we also show that Spindly is mono-ubiquitylated and this can be specifically removed by USP45 in its active form but not by the catalytic inactive form. Lastly, we identified a new role for USP45 in cell migration, similar to that which was recently described for Spindly
Layered double hydroxides-indomethacin nanohybrids: intercalation, pH influence, stability and release properties
In this work, three pH values (8, 9 and 10) were studied for the insertion of Indomethacin (Indo) molecules into Layered Double Hydroxides (LDHs). The obtained results showed that the LDH materials have been a good storage for the drug. LDHs provide thermal stability with an increase in the thermal decomposition of the drug around 100°C more. Indo into LDHs exhibited higher photostability to UV light irradiation. In vitro drug release experiments in a phosphate buffer solution (pH = 7.4) have been carried out. The loading amount of intercalated Indo was increased to 66 % at pH 8, and showed a profile of sustained release of 97 % in 8h. The release profiles were fitted by mathematical models, which describe various kinetic models that served to investigate the drug release mechanism, being the Bhaskar kinetics model the most appropriate. Virtual Instruments in Dimensional Metrology
During the last five years, in order to improve understanding of content related to "Coordinate Metrology", the Laboratorio de Metrología y Metrotecnia (LMM) from the Polytechnic University of Madrid offers its PhD students, as a course work, the construction of a virtual instrument. This virtual instrument simulates the imaging of a part to be measured by optical dimensional metrology instruments (microscopes, profile projectors, vision machines). The LMM provides students with images similar to those they would obtain with real instrumentation for the instrument adjustment and calibration process. Working with these images, students should determine the adjustment parameters of the virtual instrument. Once these parameters are set, the student can perform the proper calibration of the virtual instrument. Beyond this process, the instrument is already able to perform traceable measurement. In order to do that, LMM offers students some images of parts. Students should perform some measurements using those images and estimate the corresponding uncertainties
Agilidad, flexibilidad de producción e innovación en la empresa manufacturera española
Este artículo analiza en una muestra de empresas manufactureras españolas la relación de la producción ágil con el esfuerzo interno y la cooperación externa en innovación tecnológica. Los resultados del estudio indican que las empresas con mayor capacidad de agilidad utilizan con más intensidad un amplio conjunto de tecnologías agiles de desarrollo, fabricación y suministro. Las empresas más ágiles también cooperan externamente más en innovación y dicha cooperación modera la flexibilidad de producción de las empresas, indicando con ello que, gracias a la cooperación externa en innovación las empresas con menor flexibilidad de producción pueden aun así conseguir mayor capacidad de agilidad
The Nuclear Stellar Cluster in the Seyfert~1 Galaxy NGC 3227: High Angular Resolution NIR Imaging and Spectroscopy
NIR high angular resolution speckle imaging and imaging spectroscopy of the
nuclear region (10'' ~ 840pc) of the Seyfert1 galaxy NGC3227 are presented. A
nuclear stellar cluster is slightly resolved in the J and H band with
increasing contribution to the NIR continuum from the K to the J band. The
stellar absorption lines are extended compared to the neighboring continuum
suggesting a cluster size of ~ 70pc FWHM. Analysis of those lines suggests that
the stars are contributing about 65% (40%) of the total continuum emission in
the H (K) band in a 3.6'' aperture. Population synthesis in conjunction with
NIR spectral synthesis indicates an age of 25 to 50 Myr when red supergiants
contribute most to the NIR light. This is supported by published optical data
on the MgIb line and the CaII triplet. Although a higher age of ~ 0.5 Gyr where
AGB stars dominate the NIR light can not be excluded, the observed parameters
are at the limit of those expected for a cluster dominated by AGB stars.
However, in either case the resolved stellar cluster contributes only about ~
15 % of the total dynamical mass in the inner 300pc implying another much older
stellar population. Pure constant star formation over the last 10 Gyr can be
excluded. Therefore, at least two star formation/starburst events took place in
the nucleus of NGC3227. Since such sequences in the nuclear star formation
history are also observed in the nuclei of other galaxies a link between the
activity of the star formation and the AGN itself seems likely.Comment: accepted for publication in the Astrophysical Journal, 46 pages, 15
