    Pinworms of the red howler monkey (Alouatta seniculus) in Colombia. Gathering the pieces of the pinworm-primate puzzle

    Pinworms of primates are believed to be highly host specific parasites, forming co-evolutionary associations with their hosts. In order to assess the strength and reach of such evolutionary links, we need to have a broad understanding of the pinworm diversity associated with primates. Here, we employed an integrative taxonomic approach to assess pinworm diversity in red howler monkeys in Colombia. Molecular and morphological evidence validate the presence of at least four different species of Trypanoxyuris occurring in red howler monkeys: T. minutus, a widely distributed species, and three new species, T. seunimiii n. sp., T. kemuimae n. sp. and T. kotudoi n. sp. The mitochondrial COI gene and the 28S ribosomal gene were used for phylogenetic assessments through Bayesian inference. The three new species were morphologically distinct and formed reciprocally monophyletic lineages. Further molecular lineage subdivision in T. minutus and T. kotudoi n. sp. without morphological correspondence, suggests the potential scenario for the existence of cryptic species. Phylogenetic relationships imply that the different species of Trypanoxyuris occurring in each howler monkey species were acquired through independent colonization events. On-going efforts to uncover pinworm diversity will allow us to test the degree of host specificity and the co-phylogenetic hypothesis, as well as to further unravel the primate-pinworm evolutionary history puzzle

    Horticultural technique for rearing and redistribution of the sessile biological control agent, Rhizaspidiotus donacis on its host plant, Arundo donax

    Arundo donax, giant reed is an invasive weed in the riparian habitats of the Rio Grande Basin. A biological control program using specialist insects from the native range in Mediterranean Europe, including the arundo scale, Rhi-zaspidiotus donacis has been implemented. The arundo scale is a sessile insect that spends most of its life cycle affixed to its host plant, thus creating challenges for its rearing and redistribution to field sites. A horticultural technique was developed which allowed for rearing of the scale on small, potted A. donax microplants, which could later be transplanted to field sites. Female scale reach maturity on the microplants and produce mobile crawler scale, which move to adjacent uninfested A. donax plants. Arundo scale were established at 48 sites on the Rio Grande using microplants. This horticultural technique allows for movement of the arundo scale at any stage of its life cycle to field sites to conduct biological control programs for A. donax where it is invasive

    Visible and near-infrared observations of asteroid 2012 DA14 during its closest approach of February 15, 2013

    Near-Earth asteroid 2012 DA14 made its closest approach on February 15, 2013, when it passed at a distance of 27,700 km from the Earth's surface. It was the first time an asteroid of moderate size was predicted to approach that close to the Earth, becoming bright enough to permit a detailed study from ground-based telescopes. Asteroid 2012 DA14 was poorly characterized before its closest approach. We acquired data using several telescopes on four Spanish observatories: the 10.4m Gran Telescopio Canarias (GTC) and the 3.6m Telescopio Nazionale Galileo (TNG), both in the El Roque de los Muchachos Observatory (ORM, La Palma); the 2.2m CAHA telescope, in the Calar Alto Observatory (Almeria); the f/3 0.77m telescope in the La Hita Observatory (Toledo); and the f/8 1.5m telescope in the Sierra Nevada Observatory (OSN, Granada). We obtained visible and near-infrared color photometry, visible spectra and time-series photometry. Visible spectra together with color photometry of 2012 DA14 show that it can be classified as an L-type asteroid, a rare spectral type with a composition similar to that of carbonaceous chondrites. The time-series photometry provides a rotational period of 8.95 +- 0.08 hours after the closest approach, and there are indications that the object suffered a spin-up during this event. The large amplitude of the light curve suggests that the object is very elongated and irregular, with an equivalent diameter of around 18m. We obtain an absolute magnitude of H_R = 24.5 +- 0.2, corresponding to H_V = 25.0 +- 0.2. The GTC photometry also gives H_V = 25.29 +- 0.14. Both values agree with the value listed at the Minor Planet Center shortly after discovery. From the absolute photometry, together with some constraints on size and shape, we compute a geometric albedo of p_V = 0.44 +- 0.20, which is slightly above the range of albedos known for L-type asteroids (0.082 - 0.405).Comment: 7 pages, 4 figures, 1 table.     Mitigating Predatory Ants Promotes Establishment of Biological Control of Arundo by Arundo Scale in the Cattle Fever Tick Quarantine Zone

    Ant collection, identification, and control experiments were conducted to improve the establishment of the arundo scale, Rhizaspidiotus donacis, a biological control agent of Arundo donax, an invasive weed in the riparian habitats along the Rio Grande in Texas. Observational studies indicated R. donacis immatures are preyed upon by a variety of predator insects, especially ants. A survey of the principle ant species was made at sites along the Rio Grande with Arundo donax to help direct biological control strategies. We conclude that uses of ant baits can effectively control the common ant species found in these habitats and improve the establishment of R. donacis

    High-resolution melt (HRM) analysis for detection of SNPs associated with pyrethroid resistance in the southern cattle fever tick, Rhipicephalus (Boophilus) microplus (Acari: Ixodidae)

    The southern cattle fever tick, Rhipicephalus (Boophilus) microplus, is the most economically important ectoparasite of cattle worldwide. A limitation for sustainable control and eradication is the emergence of acaricide resistance among tick populations. Molecular diagnostic tools offer the opportunity to detect resistance rapidly, which can be complemented with confirmatory bioassays with larvae and adult ticks that are more resource and time consuming to generate. Synthetic pyrethroid resistance is one of the most prevalent and well-studied forms of resistance in arthropods, being linked with target site alterations in the sodium ion channel gene. Here, we report research on a novel molecular method to detect mutations in the para-sodium channel gene of R. microplus associated with acaricide resistance that is based on quantitative PCR high-resolution melt (HRM) analysis. Genomic DNA fragments of domains II and III of the para-sodium channel gene were amplified by real-time PCR in the presence of EVA®Green dye to test resistant and susceptible reference ticks from the U.S., Brazil, and Mexico. Larval packet tests with discriminating doses and a modified lethal time analysis were performed to confirm resistance to permethrin, cypermethrin, deltamethrin, and flumethrin in laboratory strains. Tick specimens collected from cattle that were inspected at the United States Port-of-Entry at the Texas-Mexico border were also genotyped. Previously described mutations associated with pyrethroid resistance (T170C, C190A, G184C, and T2134A) were successfully detected by qPCR-HRM in different genotypes and confirmed by sequencing. A novel non-synonymous SNP located at domain III (C2136A) and the G215T mutation in domain II, previously described only in Asian R. microplus and R. australis, were also detected with the HRM and confirmed by sequencing. This technique could be adapted for high-throughput screening, detection, and discovery of allele-specific mutations in cattle tick outbreak populations to inform eradication strategies in the USA. This knowledge could also be applied to integrated control programs in other parts of the world where R. microplus is endemic and where similar SNPs have been identified associated with pyrethroid resistance.     Identification of Light Sources using Machine Learning

    The identification of light sources represents a task of utmost importance for the development of multiple photonic technologies. Over the last decades, the identification of light sources as diverse as sunlight, laser radiation and molecule fluorescence has relied on the collection of photon statistics or the implementation of quantum state tomography. In general, this task requires an extensive number of measurements to unveil the characteristic statistical fluctuations and correlation properties of light, particularly in the low-photon flux regime. In this article, we exploit the self-learning features of artificial neural networks and naive Bayes classifier to dramatically reduce the number of measurements required to discriminate thermal light from coherent light at the single-photon level. We demonstrate robust light identification with tens of measurements at mean photon numbers below one. Our work demonstrates an improvement in terms of the number of measurements of several orders of magnitude with respect to conventional schemes for characterization of light sources. Our work has important implications for multiple photonic technologies such as LIDAR and microscopy.Comment: 8 pages, 10 figure

    The Impact of Opuntia ficus-indica and Other Vegetables on Serum Cholesterol and Triglycerides: A Cross-Sectional Analysis

    Background: Cholesterol is primarily synthesized in the liver. Treatment with statins and diet regulation are commonly prescribed for people with hypercholesterolemia. Previous claims suggest that the consumption of nopal and other vegetables may have a significant effect on diabetes but scarce is known about its relationship with cholesterol and triglycerides. The aim of this study was to evaluate the impact of nopal and other vegetables on serum cholesterol and triglyceride level in population not related with hypercholesterolemia. Methods: We analyzed a dataset comprising of students from UMAN and their relatives totaling 198 participants. Participants completed an informed consent, filled out questionnaires, anthropometric and serum lipid measurements. A semi-quantitative food questionnaire assessed the frequency and amount of consumption for 56-specific foods. We did a factor analysis with varimax rotation using 7 specific foods we were interested. We conducted linear regression analyses with total cholesterol, LDL-c, and triglycerides as dependent variables. Age, sex, BMI, body fat percentage, and consumption factors (fruits, vegetables, and grains) were included as predictors. Collinearity was assessed using VIF calculations. Results: Approximately 48% of participants consumed nopal on a weekly basis, with most considering their intake to be of median to large amounts, while only 11% reported never consuming nopal. Participants with diabetes showed higher nopal consumption [3.1 (s.e.m. 0.24) vs non-diabetic 2.7 (0.22) vs unknown 1.7 (0.28), p\u3c0.04]. A slight correlation was observed between high self-efficacy in diet and nopal ingestion (rho 0.15, p=0.02), but no significant correlations were found for fruits or grains. Regression analysis revealed that LDL-c was associated with obesity (b=-20, p=0.04) and marginally with body fat percentage (adjusted b=0.83, p=0.069), but not with any of the consumption factors. Total cholesterol was explained by age (adjusted b=0.5, p=0.002) and body fat percentage (b=1.05, p=0.024). HDL-c was marginally associated with sex (Male b=-8.3, p=0.08), while triglycerides were associated with age and obesity (overweight and Class-I, b=0.04, p=0.001 and b=0.04, p=0.04, respectively). No collinearity was found in the analyzed regressions (VIF between 0.1 and 3.4). Conclusion: Our findings suggest that cholesterol and other lipid traits are primarily influenced by genetic factors, with diet playing a minor role. Nopal, vegetables, and fruit showed no significant effects on serum lipid levels in our study. A limitation of our study is its cross-sectional design, and future research could benefit from longitudinal studies with controlled amounts of nopal and other foods

    Personality Disorders and Health Problems Distinguish Suicide Attempters from Completers in a Direct Comparison

    Background Whether suicide attempters and completers represent the same population evaluated at different points along a progression towards suicide death, overlapping populations, or completely different populations is a problem still unresolved. Methods 446 Adult suicide attempters and knowledgeable collateral informants for 190 adult suicide probands were interviewed. Sociodemographic and clinical data was collected for both groups using semi-structured interviews and structured assessments. Univariate analyses and logistic regression models were conducted to explore the similarities and differences between suicide attempters and completers. Results Univariate analyses yielded significant differences in sociodemographics, recent life events, impulsivity, suicide intent, and distribution of Axis I and II disorders. A logistic regression model aimed at distinguishing suicide completers from attempters properly classified 90% of subjects. The most significant variables that distinguished suicide from attempted suicide were the presence of narcissistic personality disorder (OR=21.4; 95% CI=6.8–67.7), health problems (OR=20.6; 95% CI=5.6–75.9), male sex (OR=9.6; 95% CI=4.42–20.9), and alcohol abuse (OR=5.5; 95% CI=2.3–14.2). Limitations Our study shares the limitations of studies comparing suicide attempters and completers, namely that information from attempters can be obtained from the subject himself, whereas the assessment of completers depends on information from close family or friends. Furthermore, different semi-structured instruments assessed Axis I and Axis II disorders in suicide attempters and completers. Finally, we have no data on inter-rater reliability data. Conclusions Suicide completers are more likely to be male and suffer from alcohol abuse, health problems (e.g. somatic illness), and narcissistic personality disorder. The findings emphasize the importance of implementing suicide prevention programs tailored to suicide attempters and completers

    Docetaxel-induced prostate cancer cell death involves concomitant activation of caspase and lysosomal pathways and is attenuated by LEDGF/p75

    <p>Abstract</p> <p>Background</p> <p>Hormone-refractory prostate cancer (HRPC) is characterized by poor response to chemotherapy and high mortality, particularly among African American men when compared to other racial/ethnic groups. It is generally accepted that docetaxel, the standard of care for chemotherapy of HRPC, primarily exerts tumor cell death by inducing mitotic catastrophe and caspase-dependent apoptosis following inhibition of microtubule depolymerization. However, there is a gap in our knowledge of mechanistic events underlying docetaxel-induced caspase-independent cell death, and the genes that antagonize this process. This knowledge is important for circumventing HRPC chemoresistance and reducing disparities in prostate cancer mortality.</p> <p>Results</p> <p>We investigated mechanistic events associated with docetaxel-induced death in HRPC cell lines using various approaches that distinguish caspase-dependent from caspase-independent cell death. Docetaxel induced both mitotic catastrophe and caspase-dependent apoptosis at various concentrations. However, caspase activity was not essential for docetaxel-induced cytotoxicity since cell death associated with lysosomal membrane permeabilization still occurred in the presence of caspase inhibitors. Partial inhibition of docetaxel-induced cytotoxicity was observed after inhibition of cathepsin B, but not inhibition of cathepsins D and L, suggesting that docetaxel induces caspase-independent, lysosomal cell death. Simultaneous inhibition of caspases and cathepsin B dramatically reduced docetaxel-induced cell death. Ectopic expression of lens epithelium-derived growth factor p75 (LEDGF/p75), a stress survival autoantigen and transcription co-activator, attenuated docetaxel-induced lysosomal destabilization and cell death. Interestingly, LEDGF/p75 overexpression did not protect cells against DTX-induced mitotic catastrophe, and against apoptosis induced by tumor necrosis factor related apoptosis inducing ligand (TRAIL), suggesting selectivity in its pro-survival activity.</p> <p>Conclusion</p> <p>These results underscore the ability of docetaxel to induce concomitantly caspase-dependent and independent death pathways in prostate cancer cells. The results also point to LEDGF/p75 as a potential contributor to cellular resistance to docetaxel-induced lysosomal destabilization and cell death, and an attractive candidate for molecular targeting in HRPC.</p
