Dendrimers containing from 9 to 18 tryptophan residues at the peryphery have been efficiently synthesized
and tested against HIV replication. These compounds inhibit an early step of the replicative
cycle of HIV, presumably virus entry into its target cell. Our data suggest that HIV inhibition can be
achieved by the preferred interaction of the compounds herein described with glycoproteins gp120 and
gp41 of the HIV envelope preventing interaction between HIV and the (co)receptors present on the host
cells. The results obtained so far indicate that 9 tryptophan residues on the periphery are sufficient for

Resumen del trabajo presentado en el XVIII Congreso de la Sociedad Española de Química Terapéutica, celebrado en Salamanca (España), del 23 al 26 de enero de 2018Microtubules, made of ¿ß¿tubulin heterodimers, are the key components of the
cytoskeleton and play a crucial role in many cellular processes, such as cell motility,
morphogenesis and mitosis.[1] Interference with microtubule dynamics induces cell
cycle arrest during mitosis and triggers cell death. Compounds that interact with tubulin,
especially those binding at the colchicine domain, have been deeply investigated as
anticancer drugs due to their dual mechanism of action as antimitotics and as vascular
disrupting agents.[2,3] Our research group has recently described a new family of
colchicine¿domain binders, based on a cyclohexanedione skeleton, with potent
antiproliferative activity against tumor and endothelial cells.[4] Moreover, to gain
insight into the binding mode of these cyclohexanediones, we have determined the
crystal structure of ¿ß¿tubulin in complex with our hit compound (TUB075). Based on
this detailed information and by applying the affinity maps program cGRILL, a structurebased
synthesis of new cyclohexanedione derivatives has been accomplished with the
objective of improving their affinity for tubulin and their antitumor activity. Following
this approach, we have obtained new compounds with potent antiproliferative activity
against tumor and endothelial cells (IC50=8¿31 nM) and with the highest Kb value
reported for compounds binding at the colchicine site in tubulin. Additional studies have
shown that they arrest cell cycle at G2/M and disrupt a network of endothelial cells.
Moreover they keep antiproliferative activity against cell lines overexpressing P¿gp,
further supporting the potential of these compounds.The financial support

We investigated the microtubule-destabilizing, vascular-targeting, anti-tumor
and anti-metastatic activities of a new series of chalcones, whose prototype compound
is (E)-3-(3’’-amino-4’’-methoxyphenyl)-1-(5’-methoxy-3’,4’-methylendioxyphenyl)-
2-methylprop-2-en-1-one (TUB091). X-ray crystallography showed that these
chalcones bind to the colchicine site of tubulin and therefore prevent the curved-tostraight
structural transition of tubulin, which is required for microtubule formation.
Accordingly, TUB091 inhibited cancer and endothelial cell growth, induced G2/M
phase arrest and apoptosis at 1-10 nM. In addition, TUB091 displayed vascular
disrupting effects in vitro and in the chicken chorioallantoic membrane (CAM) assay
at low nanomolar concentrations. A water-soluble L-Lys-L-Pro derivative of TUB091
(i.e. TUB099) showed potent antitumor activity in melanoma and breast cancer
xenograft models by causing rapid intratumoral vascular shutdown and massive
tumor necrosis. TUB099 also displayed anti-metastatic activity similar to that of
combretastatin A4-phosphate. Our data indicate that this novel class of chalcones
represents interesting lead molecules for the design of vascular disrupting agents
(VDAs). Moreover, we provide evidence that our prodrug approach may be valuable

Background: Single-particle cryoelectron microscopy (cryoEM) has played a key role in the fight against COVID-19. The
molecular mechanisms for the action of some of the currently approved drugs targeting the SARS-CoV-2 RNA-dependent
RNA polymerase, the fast developments of the current available vaccines and antibody therapies are examples of the
impact of the knowledge gained from the cryoEM structures of SARS-CoV-2 proteins in complex with proteins (ACE2 or
antibodies/nanobodies) or small compounds. Our aim is to use this technology to understand structurally how certain
antiviral compounds and proteins targeting the spike may inhibit viral entry.
Methods: 1) Production of wild-type and mutated spike and ACE2 proteins using baculovirus/insect cells. 2) Spike
binding kinetics: protein-protein and protein-small compound interactions measured by BLI Biolayer interferometry
(BLI) and/or microscale Thermophoresis (MST). 3) Buffer optimization for cryoEM grid preparation of spike variants by
thermal shift assays and negative-staining electron microscopy (NSEM). These techniques are also used to adjust the
molar ratio of spike:ACE2 and spike:small-compound complexes. 4) Structural characterization by cryoEM.
Results: At IBV-CSIC we have created a pipeline for the production and characterization of several spike variants and
ACE2 decoys. While this pipeline is described in detail in other oral/poster communications, this communication is
centered around one of the pillars within this pipeline; the structural characterization of possible drug candidates
bound to the SARS-CoV-2 spike by cryoEM. In this way, we have successfully solved structures of the spike bound to:
A) protein inhibitors as ACE2 decoys; B) a small inhibitory compound; C) mixtures of proteins and small-compound
(nanobody-heparan derivative) working cooperatively as inhibitors. These protein/drug candidates were previously
selected based on the results obtained in our interactomics platform, whereas their concentration and the buffer
conditions for cryoEM grids preparation were established based on thermal shift assays and NSEM.
Conclusion: CryoEM is a powerful tool to directly visualize the effect caused by a potential drug on a protein target. In
a short period of time we have developed this technique in our institute to be applied to the SARS-CoV-2 spike protein,
not only to obtain high-resolution structures of SARS- CoV-2 spike variants of concern (see WP4) but also to obtain the
structures of complexes of the spike with various inhibitory compounds of very different nature
The glycoprotein gp120 of the HIV-1 viral envelope has a high content in mannose residues, particularly ¿-1,2-mannose oligomers. Compounds that interact with these high-mannose type glycans may disturb the interaction between gp120 and its (co)receptors and are considered potential anti-HIV agents. Previously, we demonstrated that a tripodal receptor (1), with a central scaffold of 1,3,5-triethylbenzene substituted with three 2,3,4-trihydroxybenzoyl groups, selectively recognizes ¿-1,2-mannose polysaccharides. Here we present additional studies to determine the anti-HIV-1 activity and the mechanism of antiviral activity of this compound. Our studies indicate that 1 shows anti-HIV-1 activity in the low micromolar range and has pronounced gp120 binding and HIV-1 integrase inhibitory capacity. Background: In late 2019 SARS-CoV-2 infection appeared in China, becoming a pandemic in 2020. The scientific
community reacted rapidly, characterizing the viral genome and its encoded proteins, aiming at interfering with viral
spreading with vaccines and antivirals. The receptor binding domain (RBD) of the viral spike (S) protein plays a key role
in cell entry of the virus. It interacts with the cellular receptor for SARS-CoV-2, the membrane-bound human Angiotensin
Converting Ectoenzyme 2 (ACE2). With the goal of monitoring interference with this interaction by potential antiviral
drugs, we have set up at the Institute for Biomedicine of Valencia (IBV-CSIC) an interactomics pipeline targeting the
initial step of viral entry.
Methods: For the production part of the pipeline (pure RBD/Spike variants and soluble ACE2), see parallel poster. These
proteins allowed monitoring of the RBD/Spike-ACE2 interaction in presence or absence of potential inhibitors. Thermal
shift assays (thermofluor) were used for initial detection of compound binding at different ligand/protein ratios
and media conditions (pH, buffers, chaotropic agents). Next, binding affinity and on/off kinetics were characterized
using Biolayer interferometry (BLI), Surface plasmon resonance (SPR), Microscale Thermophoresis (MST) and/or
Isothermal titration calorimetry (ITC). For protein-protein interactions, we mostly used BLI or SPR, whereas for proteinsmall
compound analysis MST was generally best. Protein aggregation-dissociation was monitored by size exclusion
chromatography with multiangle light scattering (SEC-MALS).
Results: Candidates proven by thermal shift assays to bind to RBD/spike protein without affecting the integrity of
these proteins were subjected to quantitative affinity measurements. We successfully demonstrated that BLI, SPR and
MST can be used to follow the interactions between SARS-CoV- 2 proteins and the putative drug candidates, as well
as to monitor the interference with Spike-Ace2 binding of potential drug candidates. While BLI and SPR displayed
reproducible results in the measurement of protein-protein interaction (applied to soluble ACE2 used as a decoy),
they were less suitable for measuring the binding of small molecules. The fact that most small compounds were only
soluble in organic solvents made difficult to obtain a low signal/noise while using BLI, necessary for the assessment
of the binding. We overcame that problem by using MST. After dilution of the compounds to the final experimental
concentrations, the technique could detect a significant binding signal enough to calculate binding parameters. MST
also allowed to measure the degree of interference that each compound was having on RBD/Spike-ACE2 interaction.
The pipeline has been customized and validated with compounds of very different nature provided by different groups
belonging to the PTI and other external laboratories, as well as with different Ace2 decoys designed at the IBV.
Conclusions: The interactomics platform at the IBV has been used to successfully develop two different antiviral
approaches in order to fight COVID-19. It has allowed technical specialization of the staff as well as the development,
in a very short period of time, of two ambitious projects. We have demonstrated that we can perform interactomic
characterization for challenging projects as well as provide information about binding of antivirals to potential new
SARS-CoV-2 variants of concern
The flavivirus life cycle is strictly dependent on cellular lipid metabolism. Polyphenols like gallic acid and its derivatives are promising lead compounds for new therapeutic agents as they can exert multiple pharmacological activities, including the alteration of lipid metabolism. The evaluation of our collection of polyphenols against West Nile virus (WNV), a representative medically relevant flavivirus, led to the identification of N,N'-(dodecane-1,12-diyl)bis(3,4,5-trihydroxybenzamide) and its 2,3,4-trihydroxybenzamide regioisomer as selective antivirals with low cytotoxicity and high antiviral activity (half-maximal effective concentrations [EC50s] of 2.2 and 0.24 μM, respectively, in Vero cells; EC50s of 2.2 and 1.9 μM, respectively, in SH-SY5Y cells). These polyphenols also inhibited the multiplication of other flaviviruses, namely, Usutu, dengue, and Zika viruses, exhibiting lower antiviral or negligible antiviral activity against other RNA viruses. The mechanism underlying their antiviral activity against WNV involved the alteration of sphingolipid metabolism. These compounds inhibited ceramide desaturase (Des1), promoting the accumulation of dihydrosphingomyelin (dhSM), a minor component of cellular sphingolipids with important roles in membrane properties. The addition of exogenous dhSM or Des1 blockage by using the reference inhibitor GT-11 {N-[(1R,2S)-2-hydroxy-1-hydroxymethyl-2-(2-tridecyl-1-cyclopropenyl)ethyl]octanamide} confirmed the involvement of this pathway in WNV infection. These results unveil the potential of novel antiviral strategies based on the modulation of the cellular levels of dhSM and Des1 activity for the control of flavivirus infection.We thank Theodore C. 26 páginas, 6 figuras, 2 tablas.Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causes COVID-19, by infecting cells via the interaction of its spike protein (S) with the primary cell receptor angiotensin-converting enzyme (ACE2). To search for inhibitors of this key step in viral infection, we screened an in-house library of multivalent tryptophan derivatives. Using VSV-S pseudoparticles, we identified compound 2 as a potent entry inhibitor lacking cellular toxicity. Chemical optimization of 2 rendered compounds 63 and 65, which also potently inhibited genuine SARS-CoV-2 cell entry. Thermofluor and microscale thermophoresis studies revealed their binding to S and to its isolated receptor binding domain (RBD), interfering with the interaction with ACE2. High-resolution cryoelectron microscopy structure of S, free or bound to 2, shed light on cell entry inhibition mechanisms by these compounds. La Agenda 2030 para el Desarrollo Sostenible, aprobada por la Organización de las Naciones Unidas en el 2015, tiene como objetivo el mejoramiento de la vida de las personas en sus distintas dimensiones y en respeto de su hábitat, desde lo local hasta lo planetario. Para hacerlo realidad, la organización articula el propósito en 17 objetivos, los cuales se desglosan a su vez en 169 retos (metas) y se relacionan con las siguientes temáticas: fin de la pobreza; hambre cero, salud y bienestar, educación de calidad; igualdad de género; agua limpia y saneamiento; energía asequible y no contaminante; trabajo decente y crecimiento económico; industria, innovación e infraestructura; reducción de las desigualdades; ciudades y comunidades sostenibles; producción y consumo responsables; acción por el clima; vida submarina; vida de ecosistemas terrestres; paz, justicia e instituciones sólidas y alianzas para lograr los objetivos.
Cada uno de los objetivos está conformados a su vez por objetivos cuantificables que servirán de sustento para ponderar el progreso conseguido en la materia. Naturalmente, las condiciones de partida de las distintas regiones del mundo no son iguales.
De ahí que cada una de las áreas precisa de una estrategia para abordar y cumplir los retos, atendiendo a las particularidades de las zonas y de los países que la componen.
La Comisión Económica para América Latina y el Caribe (CEPAL) es la que monitorea los avances en esta región del Continente Americano. No obstante, para comprender la urgencia de la implementación de ciertos procesos, es necesario analizar el complejo contexto nacional, regional y local, tanto para encontrar vulnerabilidades como modelos o estrategias que se podrían aprovechar a mayor escala.
La publicación de esta obra fue financiada por el Consejo Nacional de Ciencia y Tecnología (CONACYT) a través del proyecto de investigación “Fundamentos ecológico-culturales vs. principios geográficos clásicos: Nuevos enfoques para investigar la sostenibilidad y los sistemas agrícolas en México”, con referencia número 320107 de la convocatoria Ciencia Básica y/o Ciencia de Frontera, modalidad: Paradigmas y Controversias de la Ciencia 2022 y por el Colegio de Ciencias Geográficas del Estado de México, A.C., asimismo, con aportaciones de los coordinadores (responsable técnico y

El libro “Los objetivos del desarrollo sostenible: Perspectivas y retos para la protección del ambiente y el desarrollo humano en México” aporta, en este sentido, la importante visión local de las estrategias, retos particulares y perspectivas del territorio mexicano en su camino hacia el cumplimiento de los Objetivos y las Metas. La variedad de las especialidades y experiencias de los autores y las autoras que contribuyen al presente volumen permite vislumbrar distintas realidades y estudios de caso para llamar la atención sobre aspectos que necesitan ser examinados con mayor detalle para avanzar en la consecución y cumplimiento de los objetivos.
En el capítulo primero, “Agricultura tradicional y seguridad alimentaria en comunidades rurales del sur del Estado de México” el investigador José Manuel Pérez Sánchez analiza la diversidad de estrategias de las comunidades rurales del sur del Estado de México para garantizar la seguridad alimentaria, desde los sistemas tradicionales de cultivo hasta la comercialización de la cosecha, atendiendo a la complejidad de factores (geológico-geomorfológicos, climáticos, agrícolas, económicos, sociales, familiares) que la determinan y moldean. Esto desde la perspectiva del cumplimiento del reto 2.4 que enfatiza asegurar la sostenibilidad de los sistemas de producción de alimentos y aplicar prácticas agrícolas resilientes para incrementar la productividad y la producción de alimentos. El estudio incide en la importancia del manejo del ambiente y las prácticas agrícolas tradicionales como estrategias sostenibles para hacer frente a las adversidades, notablemente contra los procesos de cambio de uso del suelo y el cambio
climático.
El capítulo segundo, elaborado en coautoría por los doctores José Carmen García Flores, María de Jesús Ordoñez Díaz y José Isabel Juan Pérez, enfatiza en el problema de sostenibilidad, enfocándose específicamente en el Objetivo 11 (ciudades y comunidades sostenibles). En el estudio titulado “El huerto familiar para el bienestar humano en Jojutla, Morelos” los autores analizan el huerto familiar urbano en el asentamiento humano mencionado, ponderándolo no sólo como una estrategia para autoabastecimiento, sino como un factor importante para el bienestar familiar. La investigación
llevada a cabo en tiempos de la pandemia SARS-COV-2, permitió analizar y destacar elementos que contribuyeron positivamente a la salud mental de las familias en situación
de emergencia.
En el capítulo “Enfermedades raras en México. Retos para objetivos del desarrollo sostenible” Agata Joanna Bak analiza la reciente resolución de la ONU concerniente a las personas que viven con una enfermedad rara en el contexto familiar, en vistas a examinar los retos que esta comunidad confronta en México. Esta investigación, que destaca la falta de una legislación específica, el problema de registro y reconocimiento de las enfermedades poco frecuentes y la inexistencia de un Plan Nacional y redes de atención específicas, visibiliza algunos de los problemas, no sólo nacionales, que este grupo de las enfermedades experimenta en el camino del cumplimiento de la meta 3.8 que se enfoca a lograr la cobertura sanitaria universal.
Los capítulos cuarto, quinto y sexto constituyen una importante reflexión en relación con el Objetivo 4 relacionado con educación de calidad. En el primero de ellos, “En torno al 4º. ODS y los indicadores de calidad educativa en el nivel superior en México”, la doctora Rosenda Ivett Vilchis Torres y los doctores Gustavo Antonio Segura Lazcano y Francisco Argüello Zepeda, llaman la atención sobre la necesidad de establecer indicadores de calidad educativa en el nivel superior en México, adecuándolos a los postulados de la Agenda 2030. El examen al que someten la educación superior resulta en la constatación de la falta de evaluación en este nivel de estudios, en la disparidad entre los criterios de calidad en la educación superior en México y los criterios internacionales y sugiere una profunda reflexión acerca de la pertinencia de las políticas
educativas y el establecimiento de los indicadores que reflejen de forma fidedigna los retos locales y nacionales en materia de educación. El capítulo quinto, “Educación Ambiental: conceptos básicos, el reto de las creencias actuales y la posibilidad de la pedagogía crítica” a cargo de la doctora Cristina María Guadalupe García-Rendón Arteaga, revisa la historia y la noción de la educación ambiental ante la crisis climática a la que se enfrenta la sociedad globalmente. Este análisis destaca la llamada de atenciónsobre el pensamiento a contracorriente de la educación ambiental, concebido entérminos de unas determinadas creencias acerca de la relación entre el ser humano y la naturaleza, la satisfacción de necesidades, entre otras. Una de las formas de paliar esta situación consistiría en incorporar, según la autora, elementos de pedagogía crítica
en la educación formal para ser capaz de trabajar con este tipo de creencias. El valor del pensamiento crítico se pondera también en el capítulo seis “La filosofía como vía para el desarrollo del pensamiento crítico: una propuesta para mejorar el mundo”, en donde los investigadores María del Carmen Magallanes Méndez y Raúl Juárez Toledo, valoran el rol que la filosofía en tanto herramienta crítica puede tener en la formación de actitudes que contemplen la complejidad, estilos de vida que tengan en cuenta lo global y aprendizajes que permitan contribuir al cumplimiento de los Objetivos del
Desarrollo Sostenible.
“Valor del agua en la producción agrícola bajo riego en México”, firmado por el doctor José Luis Montesillo Cedillo, calcula el valor del agua en la producción agrícola de bajo riego (la más extendida en México) en el territorio mexicano. En esta exploración, que establece el valor promedio en
