    A One Health Framework for the Evaluation of Rabies Control Programmes: A Case Study from Colombo City, Sri Lanka

    <div><p>Background</p><p>One Health addresses complex challenges to promote the health of all species and the environment by integrating relevant sciences at systems level. Its application to zoonotic diseases is recommended, but few coherent frameworks exist that combine approaches from multiple disciplines. Rabies requires an interdisciplinary approach for effective and efficient management.</p><p>Methodology/Principal Findings</p><p>A framework is proposed to assess the value of rabies interventions holistically. The economic assessment compares additional monetary and non-monetary costs and benefits of an intervention taking into account epidemiological, animal welfare, societal impact and cost data. It is complemented by an ethical assessment. The framework is applied to Colombo City, Sri Lanka, where modified dog rabies intervention measures were implemented in 2007. The two options included for analysis were the control measures in place until 2006 (“baseline scenario”) and the new comprehensive intervention measures (“intervention”) for a four-year duration. Differences in control cost; monetary human health costs after exposure; Disability-Adjusted Life Years (DALYs) lost due to human rabies deaths and the psychological burden following a bite; negative impact on animal welfare; epidemiological indicators; social acceptance of dogs; and ethical considerations were estimated using a mixed method approach including primary and secondary data. Over the four years analysed, the intervention cost US $1.03 million more than the baseline scenario in 2011 prices (adjusted for inflation) and caused a reduction in dog rabies cases; 738 DALYs averted; an increase in acceptability among non-dog owners; a perception of positive changes in society including a decrease in the number of roaming dogs; and a net reduction in the impact on animal welfare from intermediate-high to low-intermediate.</p><p>Conclusions</p><p>The findings illustrate the multiple outcomes relevant to stakeholders and allow greater understanding of the value of the implemented rabies control measures, thereby providing a solid foundation for informed decision-making and sustainable control.</p></div

    Improving methods and indicators for evaluating coastal water eutrophication: a pilot study in the Gulf of Maine

    Study Goals and Objectives: 1) Improve existing nutrient-related eutrophication assessment methods, updating (from early 1990s to early 2000s) the eutrophication assessment for systems included in the study with the improved method. 2) Develop a human-use/socioeconomic indicator to complement the assessment indicator. The human-use indicator was developed to evaluate costs of nutrient-related degradation in coastal waters and to put the issue into a broader context relevant to the interested public and legislators as well as to scientists. 3) Project objectives included collecting existing water quality data, developing an accessible database appropriate for application to a national study, and applying the assessment methods to 14 coastal systems – nine systems north of Cape Cod and five systems south. The geographical distribution of systems was used to examine potential regional differences in condition. 4) The intent is to use the lessons learned in this pilot study on a national scale to guide completion of an update of the 1999 National Estuarine Eutrophication Assessment

    This report presents the results of twelve monthly ichthyoplankton surveys carried out from April 2010 until March 2011 in the southern North Sea. The aim of this study was to collect data on the temporal and spatial distribution of fish eggs and larvae on the Dutch Continental Shelf (NCP). However, since fish eggs and larvae are transported with the currents onto the NCP a larger area covering the majority of the southern North Sea was sampled during the surveys. Despite technical problems or bad weather conditions preventing the sampling of all planned stations during each survey, the coverage was good and the general trends in ichthyoplankton abundance became apparent. Fish eggs were found in all months but species and numbers varied per month across the southern North Sea. The highest abundance of fish eggs was found from January until May with fish eggs being found at all stations from April until July. Very few eggs were found between August and November. From December onwards abundance of eggs increased again. The pattern was the same on the NCP. In total 35 different species of fish eggs were found. Fish larvae were found in all months but species and numbers varied in a similar pattern to the fish eggs, except the highest abundance of larvae was found in December and January in the English Channel (mostly herring). Numbers of larvae in the southern North Sea increased from April until June and afterwards then gradually declined until November. After this the abundances increased again. Larvae were found at almost all stations from May until September. In October and November larvae were found at half of all the stations both in the whole sampling area and on the NCP. The patterns was the same on the NCP. In total 74 different species of fish larvae were found. Based on the results of a modelling study, a mitigating measure was issued by the Dutch authorities forbidding pile driving of offshore wind farm foundations from January to June, in order to ensure that negative effects on prey availability for birds and marine mammals within Natura 2000 areas are minimised. The results of the year-round monthly surveys show that from April until September fish larvae were found throughout the survey area, including the NCP, in varying numbers and varying species. Still in October and November larvae were found at half of the stations although at lower abundances. Even in December larvae were still found at a quarter of the stations on the NCP. Whilst the abundance of larvae was low in October and November, there are a few species with larvae present that are absent at other times of the year. These results on ichthyoplankton abundance and results of the experimental study on the effect of pile driving on fish larvae need to be combined. Ideally they should also be assessed with subsequent studies of ichthyoplankton transport and impact on the Natura 2000 sites. This unique study is the first to comprehensively sample and analyse the ichthyoplankton with monthly resolution in the southern North Sea. These data on spatial and temporal distribution of fish eggs and larvae can be used in modelling studies to assess the effects of human activities in the southern North Sea, on different fish stocks

    The MeerKAT 1.3 GHz Survey of the Small Magellanic Cloud

    We present new radio continuum images and a source catalogue from the MeerKAT survey in the direction of the Small Magellanic Cloud (SMC). The observations, at a central frequency of 1.3 GHz across a bandwidth of 0.8 GHz, encompass a field of view ~7 x 7 degrees and result in images with resolution of 8 arcsec. The median broad-band Stokes I image Root Mean Squared noise value is ~11 microJy/beam. The catalogue produced from these images contains 108,330 point sources and 517 compact extended sources. We also describe a UHF (544-1088 MHz) single pointing observation. We report the detection of a new confirmed Supernova Remnant (SNR) (MCSNR J0100-7211) with an X-ray magnetar at its centre and 10 new SNR candidates. This is in addition to the detection of 21 previously confirmed SNRs and two previously noted SNR candidates. Our new SNR candidates have typical surface brightness an order of magnitude below those previously known, and on the whole they are larger. The high sensitivity of the MeerKAT survey also enabled us to detect the bright end of the SMC Planetary Nebulae (PNe) sample - point-like radio emission is associated with 38 of 102 optically known PNe, of which 19 are new detections. Lastly, we present the detection of three foreground radio stars amidst 11 circularly polarised sources, and a few examples of morphologically interesting background radio galaxies from which the radio ring galaxy ESO 029-G034 may represent a new type of radio object

    Standardized volumetric 3D-analysis of SPECT/CT imaging in orthopaedics: overcoming the limitations of qualitative 2D analysis

    <p>Abstract</p> <p>Background</p> <p>SPECT/CT combines high resolution anatomical 3D computerized tomography (CT) and single photon emission computerized tomography (SPECT) as functional imaging, which provides 3D information about biological processes into a single imaging modality. The clinical utility of SPECT/CT imaging has been recognized in a variety of medical fields and most recently in orthopaedics; however, clinical adoption has been limited due to shortcomings of analytical tools available. Specifically, SPECT analyses are mainly qualitative due to variation in overall metabolic uptake among patients. Furthermore, most analyses are done in 2D, although rich 3D data are available. Consequently, it is difficult to quantitatively compare the position, size, and intensity of SPECT uptake regions among patients, and therefore difficult to draw meaningful clinical conclusions.</p> <p>Methods</p> <p>We propose a method for normalizing orthopaedic SPECT/CT data that enables standardised 3D volumetric quantitative measurements and comparison among patients. Our method is based on 3D localisation using clinically relevant anatomical landmarks and frames of reference, along with intensity value normalisation using clinically relevant reference regions. Using the normalised data, we describe a thresholding technique to distinguish clinically relevant hot spots from background activity.</p> <p>Results</p> <p>Using an exemplar comparison of two patients, we demonstrate how the normalised, 3D-rendered data can provide a richer source of clinical information and allow quantitative comparison of SPECT/CT measurements across patients. Specifically, we demonstrate how non-normalized SPECT/CT analysis can lead to different clinical conclusions than the normalized SPECT/CT analysis, and that normalized quantitative analysis can be a more accurate indicator of pathology.</p> <p>Conclusions</p> <p>Conventional orthopaedic frames of reference, 3D volumetric data analysis and thresholding are used to distinguish clinically relevant hot spots from background activity. Our goal is to facilitate a standardised approach to quantitative data collection and comparison of clinical studies using SPECT/CT, enabling more widespread clinical use of this powerful imaging tool.</p

    Cbl Enforces Vav1 Dependence and a Restricted Pathway of T Cell Development

    Extensive studies of pre-TCR- and TCR-dependent signaling have led to characterization of a pathway deemed essential for efficient T cell development, and comprised of a cascade of sequential events involving phosphorylation of Lck and ZAP-70, followed by phosphorylation of LAT and SLP-76, and subsequent additional downstream events. Of interest, however, reports from our lab as well as others have indicated that the requirements for ZAP-70, LAT, and SLP-76 are partially reversed by inactivation of c-Cbl (Cbl), an E3 ubiquitin ligase that targets multiple molecules for ubiquitination and degradation. Analysis of signaling events in these Cbl knockout models, including the recently reported analysis of SLP-76 transgenes defective in interaction with Vav1, suggested that activation of Vav1 might be a critical event in alternative pathways of T cell development. To extend the analysis of signaling requirements for thymic development, we have therefore assessed the effect of Cbl inactivation on the T cell developmental defects that occur in Vav1-deficient mice. The defects in Vav1-deficient thymic development, including a marked defect in DN3-DN4 transition, were completely reversed by Cbl inactivation, accompanied by enhanced phosphorylation of PLC-Îł1 and ERKs in response to pre-TCR/TCR cross-linking of Vav1-/-Cbl-/- DP thymocytes. Taken together, these results suggest a substantially modified paradigm for pre-TCR/TCR signaling and T cell development. The observed consensus pathways of T cell development, including requirements for ZAP-70, LAT, SLP-76, and Vav1, appear to reflect the restriction by Cbl of an otherwise much broader set of molecular pathways capable of mediating T cell development
