    Ramsey goodness of trees in random graphs

    For a graph GG, we write G(Kr+1,T(n,D))G\rightarrow \big(K_{r+1},\mathcal{T}(n,D)\big) if every blue-red colouring of the edges of GG contains either a blue copy of Kr+1K_{r+1}, or a red copy of each tree with nn edges and maximum degree at most DD. In 1977, Chv\'atal proved that for any integers r,n,D2r,n,D \ge 2, KN(Kr+1,T(n,D))K_N \rightarrow \big(K_{r+1},\mathcal{T}(n,D)\big) if and only if Nrn+1N \ge rn+1. We prove a random analogue of Chv\'atal's theorem for bounded degree trees, that is, we show that for each r,D2r,D\ge 2 there exist constants C,C>0C,C'>0 such that if pCn2/(r+2)p \ge C{n}^{-2/(r+2)} and Nrn+C/pN \geq rn + C'/p, then G(N,p)(Kr+1,T(n,D))G(N,p) \rightarrow \big(K_{r+1},\mathcal{T}(n,D)\big) with high probability as nn\to \infty. The proof combines a stability argument with the embedding of trees in expander graphs. Furthermore, the proof of the stability result is based on a sparse random analogue of the Erd\H{o}s--S\'os conjecture for trees with linear size and bounded maximum degree, which may be of independent interest.Comment: 30 pages, 3 figure

    Visiones y valoraciones subculturales de la locura en España (1981-2010)

    El concepto de “locura” -más amplio y menos definido que el de “psicosis”- continúa siendo de uso popular, pese a haberse abandonado en medicina por su carga peyorativa. Las manifestaciones culturales y artísticas pueden reflejar las representaciones sociales de la enfermedad mental, a la vez que difundir, amplificar, matizar y/o metabolizar los sentidos circulantes. Atendiendo al estigma y la discriminación como barreras para el acceso a los tratamientos psiquiátricos, el objetivo de este trabajo es la aproximación a las visiones y valoraciones subculturales de la “locura”. Para ello, se presentan y discuten los resultados del análisis de contenido de 3.653 canciones punk españolas (1981-2010). Mediante este proceso se pretende delinear un retrato subcultural del “loco” y “la locura”. Hipotetizamos que una parte de las visiones estigmatizantes presentes en la población general pueden hallarse contenidas en los discursos lítero-musicales del punk español, aunque con matices valorativos que pueden diferir respecto a la cultura mayoritaria

    Global Multidimensional Poverty Index 2021: Unmasking disparities by ethnicity, caste and gender

    This report provides a comprehensive picture of acute multidimensional poverty to inform the work of countries and communities building a more just future for the global poor. Part I focuses on where we are now. It examines the levels and composition of multidimensional poverty across 109 countries covering 5.9 billion people. It also discusses trends among more than 5 billion people in 80 countries, 70 of which showed a statistically significant reduction in Multidimensional Poverty Index value during at least one of the time periods presented. While the COVID-19 pandemic's impact on developed countries is already an active area of research, this report offers a multidimensional poverty perspective on the experience of developing countries. It explores how the pandemic has affected three key development indicators (social protection, livelihoods and school attendance), in association with multidimensional poverty, with a focus predominantly on Sub-Saharan Africa. Part II profiles disparities in multidimensional poverty with new research that scrutinizes estimates disaggregated by ethnicity or race and by caste to identify who and how people are being left behind. It also explores the proportion of multidimensionally poor people who live in a household in which no female member has completed at least six years of schooling and presents disparities in multidimensional poverty by gender of the household head. Finally, it probes interconnections between the incidence of multidimensional poverty and intimate partner violence against women and girls

    Gain-of-function mutation in ubiquitin ligase KLHL24 causes desmin degradation and dilatation in hiPSC-derived engineered heart tissues

    The start codon c.1A>G mutation in KLHL24, encoding ubiquitin ligase KLHL24, results in the loss of 28 N-terminal amino acids (KLHL24-ΔN28) by skipping the initial start codon. In skin, KLHL24-ΔN28 leads to gain of function, excessively targeting intermediate filament keratin-14 for proteasomal degradation and ultimately causing epidermolysis bullosa simplex (EBS). The majority of patients with EBS are also diagnosed with dilated cardiomyopathy (DCM), but the pathological mechanism in the heart is unknown. As desmin is the cardiac homolog of keratin-14, we hypothesized that KLHL24-ΔN28 leads to excessive degradation of desmin, resulting in DCM. Dynamically loaded engineered heart tissues (dyn-EHTs) were generated from human-induced pluripotent stem cell–derived (hiPSC-derived) cardiomyocytes from 2 patients and 3 nonfamilial controls. Ten-fold lower desmin protein levels were observed in patient-derived dyn-EHTs, in line with diminished desmin levels detected in patients’ explanted heart. This was accompanied by tissue dilatation, impaired mitochondrial function, decreased force values, and increased cardiomyocyte stress. HEK293 transfection studies confirmed KLHL24-mediated desmin degradation. KLHL24 RNA interference or direct desmin overexpression recovered desmin protein levels, restoring morphology and function in patient-derived dyn-EHTs. To conclude, presence of KLHL24-ΔN28 in cardiomyocytes leads to excessive degradation of desmin, affecting tissue morphology and function, which can be prevented by restoring desmin protein levels

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field
