    The long and eventful discovery voyage of Juan Ponce de León, the Spanish conquistador and explorer, has not received the amount of in-depth study and analysis that this historically significant voyage so richly deserves. Perhaps a reason for this can be found in the fact that sixteenth-century (and later) historians, in dwelling on the fact that the largely apocryphal purpose of the voyage was to find a fountain of youth, perverted the factual reporting of the real purpose of the voyage and its several significant discoveries. This distorted version continues in many contemporary history books because of the proclivity of writers simply to paraphrase previous authors. To avoid this I have examined original source documents wherever possible for information and data untainted by unsubstantiated conjecture and fiction

    Distribution of DDS-cerberus Authenticated Facial Recognition Streams

    Successful missions in the field often rely upon communication technologies for tactics and coordination. One middleware used in securing these communication channels is Data Distribution Service (DDS) which employs a publish-subscribe model. However, researchers have found several security vulnerabilities in DDS implementations. DDS-Cerberus (DDS-C) is a security layer implemented into DDS to mitigate impersonation attacks using Kerberos authentication and ticketing. Even with the addition of DDS-C, the real-time message sending of DDS also needs to be upheld. This paper extends our previous work to analyze DDS-C’s impact on performance in a use case implementation. The use case covers an artificial intelligence (AI) scenario that connects edge sensors across a commercial network. Specifically, it characterizes how DDS-C performs between unmanned aerial vehicles (UAV), the cloud, and video streams for facial recognition. The experiments send a set number of video frames over the network using DDS to be processed by AI and displayed on a screen. An evaluation of network traffic using DDS-C revealed that it was not statistically significant compared to DDS for the majority of the configuration runs. The results demonstrate that DDS-C provides security benefits without significantly hindering the overall performance

    Quantifying DDS-cerberus Network Control Overhead

    Securing distributed device communication is critical because the private industry and the military depend on these resources. One area that adversaries target is the middleware, which is the medium that connects different systems. This paper evaluates a novel security layer, DDS-Cerberus (DDS-C), that protects in-transit data and improves communication efficiency on data-first distribution systems. This research contributes a distributed robotics operating system testbed and designs a multifactorial performance-based experiment to evaluate DDS-C efficiency and security by assessing total packet traffic generated in a robotics network. The performance experiment follows a 2:1 publisher to subscriber node ratio, varying the number of subscribers and publisher nodes from three to eighteen. By categorizing the network traffic from these nodes into either data message, security, or discovery+ with Quality of Service (QoS) best effort and reliable, the mean security traffic from DDS-C has minimal impact to Data Distribution Service (DDS) operations compared to other network traffic.     Explaining decision-making in government: the neo-Durkheimian institutional framework

    In understanding styles of political judgement in government decision-making, explanatory limitations of rational choice, prospect theoretic, historical institutional, groupthink, and other approaches suggest that there is space for developing other frameworks. This article argues that the neo-Durkheimian institutional theoretical framework deserves serious consideration. It shows that it offers a powerful causally explanatory framework for generating theories of decision-making in government which can be examined using historical comparative research designs. The value of the concept of a ‘thought style’ for understanding political judgement is demonstrated, and contrasted sharply with ideology. The theory argues that informal institutions explain thought styles. Well-known cases from the Cuban missile crisis, and the Wilson and Heath governments illustrate the argument. The article rebuts criticisms offered of the neo-Durkheimian institutional framework in the literature.     Metformin Blunts Muscle Hypertrophy in Response to Progressive Resistance Exercise Training in Older Adults: A Randomized, Double‐Blind, Placebo‐Controlled, Multicenter Trial: The MASTERS Trial

    Progressive resistance exercise training (PRT) is the most effective known intervention for combating aging skeletal muscle atrophy. However, the hypertrophic response to PRT is variable, and this may be due to muscle inflammation susceptibility. Metformin reduces inflammation, so we hypothesized that metformin would augment the muscle response to PRT in healthy women and men aged 65 and older. In a randomized, double-blind trial, participants received 1,700 mg/day metformin (N = 46) or placebo (N = 48) throughout the study, and all subjects performed 14 weeks of supervised PRT. Although responses to PRT varied, placebo gained more lean body mass (p = .003) and thigh muscle mass (p \u3c .001) than metformin. CT scan showed that increases in thigh muscle area (p = .005) and density (p = .020) were greater in placebo versus metformin. There was a trend for blunted strength gains in metformin that did not reach statistical significance. Analyses of vastus lateralis muscle biopsies showed that metformin did not affect fiber hypertrophy, or increases in satellite cell or macrophage abundance with PRT. However, placebo had decreased type I fiber percentage while metformin did not (p = .007). Metformin led to an increase in AMPK signaling, and a trend for blunted increases in mTORC1 signaling in response to PRT. These results underscore the benefits of PRT in older adults, but metformin negatively impacts the hypertrophic response to resistance training in healthy older individuals.     A Reverse Genetics Platform That Spans the Zika Virus Family Tree

    ABSTRACT Zika virus (ZIKV), a mosquito-borne flavivirus discovered in 1947, has only recently caused large outbreaks and emerged as a significant human pathogen. In 2015, ZIKV was detected in Brazil, and the resulting epidemic has spread throughout the Western Hemisphere. Severe complications from ZIKV infection include neurological disorders such as Guillain-Barré syndrome in adults and a variety of fetal abnormalities, including microcephaly, blindness, placental insufficiency, and fetal demise. There is an urgent need for tools and reagents to study the pathogenesis of epidemic ZIKV and for testing vaccines and antivirals. Using a reverse genetics platform, we generated six ZIKV infectious clones and derivative viruses representing diverse temporal and geographic origins. These include three versions of MR766, the prototype 1947 strain (with and without a glycosylation site in the envelope protein), and H/PF/2013, a 2013 human isolate from French Polynesia representative of the virus introduced to Brazil. In the course of synthesizing a clone of a circulating Brazilian strain, phylogenetic studies identified two distinct ZIKV clades in Brazil. We reconstructed viable clones of strains SPH2015 and BeH819015, representing ancestral members of each clade. We assessed recombinant virus replication, binding to monoclonal antibodies, and virulence in mice. This panel of molecular clones and recombinant virus isolates will enable targeted studies of viral determinants of pathogenesis, adaptation, and evolution, as well as the rational attenuation of contemporary outbreak strains to facilitate the design of vaccines and therapeutics. IMPORTANCE Viral emergence is a poorly understood process as evidenced by the sudden emergence of Zika virus in Latin America and the Caribbean. Malleable reagents that both predate and span an expanding epidemic are key to understanding the virologic determinants that regulate pathogenesis and transmission. We have generated representative cDNA molecular clones and recombinant viruses that span the known ZIKV family tree, including early Brazilian isolates. Recombinant viruses replicated efficiently in cell culture and were pathogenic in immunodeficient mice, providing a genetic platform for rational vaccine and therapeutic design

This research explores the role of postgraduate level professional development in contributing to professional identity. The research utilises a case study approach and adopts multiple tools to provide an in-depth examination of practitioners’ perceptions of themselves as professionals within their lived world experience. It presents a group of practitioners struggling to define themselves as professionals due to changing occupational nomenclature resulting from shifting government policy. Postgraduate professional development generated a perceived enhancement in professional identity through exposure to theory, policy and opportunities for reflection, thus contributing to more confident and empowered practitioners.     Early satellite cell communication creates a permissive environment for long-term muscle growth

    Using in vivo muscle stem cell (satellite cell)-specific extracellular vesicle (EV) tracking, satellite cell depletion, in vitro cell culture, and single-cell RNA sequencing, we show satellite cells communicate with other cells in skeletal muscle during mechanical overload. Early satellite cell EV communication primes the muscle milieu for proper long-term extracellular matrix (ECM) deposition and is sufficient to support sustained hypertrophy in adult mice, even in the absence of fusion to muscle fibers. Satellite cells modulate chemokine gene expression across cell types within the first few days of loading, and EV delivery of miR 206 to fibrogenic cells represses Wisp1 expression required for appropriate ECM remodeling. Late-stage communication from myogenic cells during loading is widespread but may be targeted toward endothelial cells. Satellite cells coordinate adaptation by influencing the phenotype of recipient cells, which extends our understanding of their role in muscle adaptation beyond regeneration and myonuclear donation

    Why do banks promise to pay par on demand?

    We survey the theories of why banks promise to pay par on demand and examine evidence about the conditions under which banks have promised to pay the par value of deposits and banknotes on demand when holding only fractional reserves. The theoretical literature can be broadly divided into four strands: liquidity provision, asymmetric information, legal restrictions, and a medium of exchange. We assume that it is not zero cost to make a promise to redeem a liability at par value on demand. If so, then the conditions in the theories that result in par redemption are possible explanations of why banks promise to pay par on demand. If the explanation based on customers’ demand for liquidity is correct, payment of deposits at par will be promised when banks hold assets that are illiquid in the short run. If the asymmetric-information explanation based on the difficulty of valuing assets is correct, the marketability of banks’ assets determines whether banks promise to pay par. If the legal restrictions explanation of par redemption is correct, banks will not promise to pay par if they are not required to do so. If the transaction explanation is correct, banks will promise to pay par value only if the deposits are used in transactions. After the survey of the theoretical literature, we examine the history of banking in several countries in different eras: fourth-century Athens, medieval Italy, Japan, and free banking and money market mutual funds in the United States. We find that all of the theories can explain some of the observed banking arrangements, and none explain all of them
