Testing metric-affine f(R)-gravity by relic scalar gravitational waves
We discuss the emergence of scalar gravitational waves in metric-affine
f(R)-gravity. Such a component allows to discriminate between metric and
metric-affine theories The intrinsic meaning of this result is that the
geodesic structure of the theory can be discriminated. We extend the formalism
of cross correlation analysis, including the additional polarization mode, and
calculate the detectable energy density of the spectrum for cosmological relic
gravitons. The possible detection of the signal is discussed against
sensitivities of VIRGO, LIGO and LISA interferometers.Comment: 12 pages, 4 figure
Constraints on Inflation
A short introduction to structure formation is given, followed by a
discussion of the possible characteristics of the initial perturbations
assuming a generic inflationary origin. Observational data related to
large-scale structure and the cosmic microwave background radiation is then
used in an attempt to constrain the characteristics of such perturbations.
Future directions are also explored.
The possibility of direct detection of a stochastic gravitational wave
background produced during an inflationary phase in the early Universe is
briefly discussed, as well as the available evidence regarding the present
value of the total energy density in the Universe.Comment: 24 pages, Latex, no figures, based on talks given at the Cargese

Isocurvature and Adiabatic Fluctuations of Axion in Chaotic Inflation Models and Large Scale Structure
In the chaotic inflation models, quantum fluctuations for axion fields lead
to the overproduction of domain walls and too large isocurvature fluctuations
which is inconsistent with the observations of cosmic microwave background
anisotropies. These problems are solved by assuming a very flat potential for
the Peccei-Quinn scalar. As the simplest possibility, we consider a model where
the Peccei-Quinn scalar is an inflaton itself and show that the isocurvature
fluctuations can be comparable with the adiabatic ones. We investigate
cosmological implications in the case that both adiabatic and isocurvature
fluctuations exist and find that the amplitude of the matter spectrum becomes
smaller than that for the pure adiabatic case. This leads to relatively high
bias parameter () which is favoured by the current observations.Comment: 5 pages, uudecoded postscript file with figure
Rapid escape of new SARS-CoV-2 Omicron variants from BA.2-directed antibody responses
In November 2021, Omicron BA.1, containing a raft of new spike mutations, emerged and quickly spread globally. Intense selection pressure to escape the antibody response produced by vaccines or severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection then led to a rapid succession of Omicron sub-lineages with waves of BA.2 and then BA.4/5 infection. Recently, many variants have emerged such as BQ.1 and XBB, which carry up to 8 additional receptor-binding domain (RBD) amino acid substitutions compared with BA.2. We describe a panel of 25 potent monoclonal antibodies (mAbs) generated from vaccinees suffering BA.2 breakthrough infections. Epitope mapping shows potent mAb binding shifting to 3 clusters, 2 corresponding to early-pandemic binding hotspots. The RBD mutations in recent variants map close to these binding sites and knock out or severely knock down neutralization activity of all but 1 potent mAb. This recent mAb escape corresponds with large falls in neutralization titer of vaccine or BA.1, BA.2, or BA.4/5 immune serum
Self-Transcendence as a Buffer Against COVID-19 Suffering : The Development and Validation of the Self-Transcendence Measure-B
The age of COVID-19 calls for a different approach toward global well-being and flourishing through the transcendence suffering as advocated by existential positive psychology. In the present study, we primarily explained what self-transcendence is and why it represents the most promising path for human beings to flourish through the transformation of suffering in a difficult and uncertain world. After reviewing the literature on self-transcendence experiences, we concluded that the model of self-transcendence presented by Frankl is able to integrate both of the characteristics associated with self-transcendence. Afterward, we discussed how the self-transcendence paradigm proposed by Wong, an extension of the model by Frankl, may help awaken our innate capacity for connections with the true self, with others, and with God or something larger than oneself. We presented self-transcendence as a less-traveled but more promising route to achieve personal growth and mental health in troubled times. Finally, we presented the history of the development and psychometrics of the Self-Transcendence Measure-Brief (STM-B) and reported the empirical evidence that self-transcendence served as a buffer against COVID-19 suffering. The presented data in the current study suggested that the best way to overcome pandemic suffering and mental health crises is to cultivate self-transcendence
The SARS-CoV-2 variants associated with infections in India, B.1.617, show enhanced spike cleavage by furin
AbstractThe spike (S) glycoprotein of the SARS-CoV-2 virus that emerged in 2019 contained a suboptimal furin cleavage site at the S1/S2 junction with the sequence 681PRRAR/S686. This cleavage site is required for efficient airway replication, transmission, and pathogenicity of the virus. The B.1.617 lineage has recently emerged in India, coinciding with substantial disease burden across the country. Early evidence suggests that B.1.617.2 (a sublineage of B.1.617) is more highly transmissible than contemporary lineages. B.1.617 and its sublineages contain a constellation of S mutations including the substitution P681R predicted to further optimise this furin cleavage site. We provide experimental evidence that virus of the B.1.617 lineage has enhanced S cleavage, that enhanced processing of an expressed B.1.617 S protein in cells is due to P681R, and that this mutation enables more efficient cleavage of a peptide mimetic of the B.1.617 S1/S2 cleavage site by recombinant furin. Together, these data demonstrate viruses in this emerging lineage have enhanced S cleavage by furin which we hypothesise could be enhancing transmissibility and pathogenicity.</jats:p
Recurrent SARS-CoV-2 mutations in immunodeficient patients
Long-term severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections in immunodeficient patients are an important source of variation for the virus but are understudied. Many case studies have been published which describe one or a small number of long-term infected individuals but no study has combined these sequences into a cohesive dataset. This work aims to rectify this and study the genomics of this patient group through a combination of literature searches as well as identifying new case series directly from the COVID-19 Genomics UK (COG-UK) dataset. The spike gene receptor-binding domain and N-terminal domain (NTD) were identified as mutation hotspots. Numerous mutations associated with variants of concern were observed to emerge recurrently. Additionally a mutation in the envelope gene, T30I was determined to be the second most frequent recurrently occurring mutation arising in persistent infections. A high proportion of recurrent mutations in immunodeficient individuals are associated with ACE2 affinity, immune escape, or viral packaging optimisation.
There is an apparent selective pressure for mutations that aid cell–cell transmission within the host or persistence which are often different from mutations that aid inter-host transmission, although the fact that multiple recurrent de novo mutations are considered defining for variants of concern strongly indicates that this potential source of novel variants should not be discounted
