    The Sub-Polar Gyre Index - a community data set for application in fisheries and environment research

    Abstract. Scientific interest in the sub-polar gyre of the North Atlantic Ocean has increased in recent years. The sub-polar gyre has contracted and weakened, and changes in circulation pathways have been linked to changes in marine ecosystem productivity. To aid fisheries and environmental scientists, we present here a time series of the Sub-Polar Gyre Index (SPG-I) based on monthly mean maps of sea surface height. The established definition of the SPG-I is applied, and the first EOF (empirical orthogonal function) and PC (principal component) are presented. Sensitivity to the spatial domain and time series length are explored but found not to be important factors in terms of the SPG-I's interpretation. Our time series compares well with indices presented previously. The SPG-I time series is freely available online (http://dx.doi.org/10.7489/1806-1), and we invite the community to access, apply, and publish studies using this index time series. </jats:p

    ESTIMATING FACULTY AND STUDENT WORKLOAD FOR INTERACTION IN ONLINE GRADUATE MUSIC COURSES

    Research suggests that faculty perceive greater workload for online teaching. These perceptions have little quantitative support. This study utilized seven online graduate courses, over a three year period to estimate faculty and student workload for interaction via online discussions and electronic mail using average reading and typing speeds. Weekly faculty workload estimates for interaction did not exceed normal expectations for faculty “office hours” for six of the seven courses. Perceptions of excessive workload for communication may be better explained by the dynamics of online interaction found in this study. Online students attempted to contact their instructors, twenty-four hours per day, seven days per week, at least every fourteen hours. Further research is needed to establish the time needed for FTF teaching interaction and to validate actual typing and reading speeds for more accurate estimates of the time needed for online course interaction

    Tuberculosis interferon-gamma responses in the breast milk of human immunodeficiency virus infected mothers

    Tuberculosis (TB) cellular immune responses were examined in breastmilk of HIV-infected mothers using the T-SPOT.TB interferon gamma release assay (IGRA). Positive TB IFN-γ responses were detected in 6 of 8 (75%) valid breast milk assays. Among 7 mothers with paired breastmilk and blood assays, TB IFN- γ responses were higher in breastmilk compared to blood (p=.02). The magnitude of TB IFN-γ responses in maternal breastmilk and blood were correlated. Elucidating the influence of breastmilk TB immune responses on infant TB susceptibility and immunity may inform future maternal TB vaccine strategies

    Can 100% Pasture-Based Livestock Farming Produce Enough Ruminant Meat to Meet the Current Consumption Demand in the UK?

This is an open access article distributed under the terms of the Creative Commons Attribution License (CC BY), https://creativecommons.org/licenses/by/4.0/Grassland is grouped into temporary, permanent, and rough grazing types in the United Kingdom (UK), making up more than 60% of the national agricultural land area. It provides avenues for grazed fodder or ensiled forage contributing a large proportion of the diets consumed by cattle and sheep. The official agricultural census data in 2011 to 2020 showed that, on average, UK cattle and sheep farming can produce meat to satisfy 83.3 and 100.8% of domestic cattle beef and sheep meat consumption levels, respectively. Out of the large agricultural census datasets, we used the populations of cattle and sheep, as well as the UK definition of a standard livestock unit (SLU), to normalise the respective herd populations into a total standard livestock unit (TSLU). We then used the annual domestic meat production in dressed carcass weight to calculate cattle and sheep meat productivity per SLU. Using the potential herbal dry matter yields per year and areas of the different grassland types across the UK, the potential total available pasture feed was calculated. This potential production of herbal biomass was translated into the potential carrying capacity expressed in a TSLU. This total potential carrying capacity was partitioned into cattle and sheep sectors so that the routes of pasture-based-only options with which to produce ruminant meat to meet the current UK domestic consumption demands were assessed. The estimated mean potential annual pasture forage feed in 2011–2020 was approximately 82.0 million (M) metric tonnes (t), which can be translated into a potential carrying capacity of 17.9 M SLUs compared with the current mean 9.36 M SLUs in the survey data of the UK. With the ratio of sheep to cattle at 8.2:25 in the national TSLU, the UK national demands at present consumption levels of cattle and sheep meat can be arithmetically met with pasture grass utilisation rates at or above 65% and 50% by cattle and sheep farming systems, respectively.Peer reviewe

    Urogenital Symptoms and Pain History as Precursors of Vulvodynia: A Longitudinal Study

    Abstract Background: We sought to assess vulvodynia incidence and risk factors among those with and without premorbid urogenital symptoms. Methods: Women's Health Registry members who completed a baseline assessment in 2004 were sent a 2-year and 4-year follow-up survey containing a validated screen for vulvodynia. Subgroup analysis of vulvodynia incidence rates was performed, and risk factors associated with incidence were assessed. Results: Of 1037 original enrollees, 723 (69.7%) completed consecutive surveys (initial and 2-year or initial, 2-year, and 4-year), 660 of whom did not have current or past vulvodynia at baseline. Of these 660, 71 (10.8%) first met criteria for vulvodynia within the 4-year period, for an annual incidence rate of 3.1% (95% confidence interval [CI] 2.5-4.0). Baseline strict controls were less likely to develop criteria for vulvodynia diagnosis (annual incidence rate of 1.4%) compared to those with an intermediate phenotype (presence of dyspareunia or history of short-term vulvar pain), for whom the incidence rate was 5.6% (p<0.001). Risk factors for incident vulvodynia differed between these two groups. Among the strict controls, an increased risk was noted among younger women (incidence rate ratio) [IRR] 3.6). For those with an intermediate phenotype, risk was increased among nonwhite women and those reporting pain with or after intercourse (IRR 2.2, 3.4, and 3.1, respectively). In both control groups, incident vulvodynia risk increased among those reporting urinary burning at enrollment (IRR 4.2 and 2.8 for strict and intermediate phenotype controls, respectively). Conclusions: The annual incidence of vulvodynia is substantial (3.1%) and is greater among women reporting a history of dyspareunia or vulvar pain that did not meet criteria for vulvodynia compared to those without this history, suggesting that generalized urogenital sensitivity may be a common underlying mechanism predating the clinical presentation of vulvodynia.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/98465/1/jwh%2E2012%2E3566.pd

    Bacteriophage Therapy: Clinical Trials and Regulatory Hurdles

    Increasing reports of antimicrobial resistance and limited new antibiotic discoveries and development have fuelled innovation in other research fields and led to a revitalization of bacteriophage (phage) studies in the Western world. Phage therapy mainly utilizes obligately lytic phages to kill their respective bacterial hosts, while leaving human cells intact and reducing the broader impact on commensal bacteria that often results from antibiotic use. Phage therapy is rapidly evolving and has resulted in cases of life-saving therapeutic use and multiple clinical trials. However, one of the biggest challenges this antibiotic alternative faces relates to regulations and policy surrounding clinical use and implementation beyond compassionate cases. This review discusses the multi-drug resistant Gram-negative pathogens of highest critical priority and summarizes the current state-of-the-art in phage therapy targeting these organisms. It also examines phage therapy in humans in general and the approaches different countries have taken to introduce it into clinical practice and policy. We aim to highlight the rapidly advancing field of phage therapy and the challenges that lie ahead as the world shifts away from complete reliance on antibiotics

    How data visualisation using historical medical journals can contribute to current debates around antibiotic use and antimicrobial resistance in primary care

    BackgroundThe early years of antibiotic use in primary care (c1950-1969) has received little attention. Medical journals provide a rich source for studying historic healthcare practitioners’ views and interests, with the potential to inform contemporary debate around issues of overuse and antimicrobial resistance. AimsPilot study to test the application of digital methods to interrogate historical medical journal data in relation to antibiotic use.Methods / ApproachMeta-data and scanned articles were extracted from the online British Journal of General Practice (BJGP) archive from inception (1953) to 1969. Searchable text was generated using an application called ABBYY optical character recognition, and Python used to generate data visualisations exploring (1) how BJGP changed during the period, (2) mentions of terms ‘antibiotic(s)’, ‘penicillin’, ‘resistance/resistant’ and mapping when and where they occurred.Results / EvaluationFrom 1953-1969, BJGP expanded in terms of number of annual issues (4 to 17) and annual pages (&lt;25 to &gt;1100). Heatmap visualisations were used to facilitate understanding of the frequency with which use of the term ‘antibiotic(s)’ occurred. By 1969 an article mentioning ‘antibiotic(s)’ was published monthly. Bigram searches found ‘treatment’ and ‘therapy’ to be the two most common terms that appeared with ‘antibiotic(s)’. The fourth and seventh most common terms were ‘resistant’ (first appearing in 1955) and ‘resistance’ (1962).ConclusionsThis pilot work shows that primary care publications increased considerably between 1953-1969. Articles on antibiotics featured frequently in relation to therapeutic intervention, and concerns around resistance occurred at an early stage. This approach provides new insights into how attitudes and behaviours around antibiotic use by primary care have evolved over time. It may also have the potential to inform study of the future use of antibiotics in primary care. <br/

    Sterol Carrier Protein-2 Directly Interacts with Caveolin-1 in Vitro and in Vivo

    HDL-mediated reverse-cholesterol transport as well as phosphoinositide signaling are mediated through plasma membrane microdomains termed caveolae/lipid rafts. However, relatively little is known regarding mechanism(s) whereby these lipids traffic to or are targeted to caveolae/lipid rafts. Since sterol carrier protein-2 (SCP-2) binds both cholesterol and phosphatidylinositol, the possibility that SCP-2 might interact with caveolin-1 and caveolae was examined. Double immunolabeling and laser scanning fluorescence microscopy showed that a small but significant portion of SCP-2 colocalized with caveolin-1 primarily at the plasma membrane of L-cells and more so within intracellular punctuate structures in hepatoma cells. In SCP-2 overexpressing L-cells, SCP-2 was detected in close proximity to caveolin, 48 ± 4 Å, as determined by fluorescence resonance energy transfer (FRET) and immunogold electron microscopy. Cell fractionation of SCP-2 overexpressing L-cells and Western blotting detected SCP-2 in purified plasma membranes, especially in caveolae/ lipid rafts as compared to the nonraft fraction. SCP-2 and caveolin-1 were coimmunoprecipitated from cell lysates by anti-caveolin-1 and anti-SCP-2. Finally, a yeast two-hybrid assay demonstrated that SCP-2 directly interacts with caveolin-1 in vivo. These interactions of SCP-2 with caveolin-1 were specific since a functionally related protein, phosphatidyinositol transfer protein (PITP), colocalized much less well with caveolin-1, was not in close proximity to caveolin-1 (i.e., \u3e120 Å), and was not coimmunoprecipitated by anti-caveolin-1 from cell lysates. In summary, it was shown for the first time that SCP-2 (but not PITP) selectively interacted with caveolin-1, both within the cytoplasm and at the plasma membrane. These data contribute significantly to our understanding of the role of SCP-2 in cholesterol and phosphatidylinositol targeted from intracellular sites of synthesis in the endoplasmic reticulum to caveolae/lipid rafts at the cell surface plasma membrane
