    Change blindness: size matters

    It is easy to detect a small change between two sequential presentations of a visual stimulus, but, if they are separated by a blank interval, performance is around chance. This change blindness (CB) can be rectified, or improved, by cueing the spatial location of the change either in the first stimulus or the interval; however, no advantage is conferred when the cue appears during the second presentation of the stimulus. This supports the idea that a representation of the first stimulus is formed and persists through the course of the interval before being 'overwritten' by the second presentation of the stimulus (Landman et al, 2003 Vision Research 43 149 - 164). We were interested in the time course of the cueing effect during the interval. Following Landman et al, our first stimulus was an array of eight rectangles defined by texture and there was a 50% chance that one of the rectangles would change orientation in the second stimulus. Five cues were used, one within the first stimulus, three across the interval, and one in the second stimulus. Only one of these cues appeared in each trial. The cued rectangle was the one that would change between the first and second stimulus when a change occurred. The cue was a yellow line. Eighty-five observers showed the characteristic cueing performance supporting 'overwriting', but performance decreased over the duration of the interval suggesting that the initial representation of the first stimulus fades over time. However, when the size of the rectangles was increased, performance across the interval improved significantly. We consider two possible explanations: one is that simply by increasing rectangle size we raise the storage capacity for the number of rectangles in our representation, the other is that storage is related to task difficulty

    First paragraph: As any fish farmer knows, feed is usually the most important variable production cost. A simple objective is therefore to minimize waste from uneaten food, which has the added benefit of reducing the risk of environmental degradation. However, decreasing feed level risks reducing growth rate, leadìng to a rise in other costs per unit of production. The optimum biological feeding rate is thus rarely the same as the optimum economic rate. In practice, these calculations are complicated as feed requirement and efficiency of conversion varies with changing environmental conditions including water temperature, oxygen concentration, water quality, current speed, light intensity and day length. Feed utilisation also varies with diet quality and physiological factors such as age/size, life-stage, stress level and endogenous rhythms. lt is therefore not surprising that these factors contrìbute towards an element of uncertaìnty regarding the amount of feed required, often leading to under or over feeding of stock and resultant under performance of the system

    BACKGROUND: The analysis of internet search traffic may present the opportunity to gain insights into general trends and patterns in information seeking behaviour related to medical conditions at a population level. For prevalent and widespread problems such as foot and ankle pain, this information has the potential to improve our understanding of seasonality and trends within these conditions and their treatments, and may act as a useful proxy for their true incidence/prevalence characteristics. This study aimed to explore seasonal effects, general trends and relative popularity of internet search terms related to foot and ankle pain over the past decade. METHODS: We used the Google Trends tool to obtain relative search engine traffic for terms relating to foot and ankle pain and common treatments from Google search and affiliated pages for major northern and southern hemisphere English speaking nations. Analysis of overall trends and seasonality including summer/winter differences was carried out on these terms. RESULTS: Searches relating to general foot pain were on average 3.4 times more common than those relating to ankle pain, and twice as common as searches relating to heel pain. Distinct seasonal effects were seen in the northern hemisphere, with large increases in search volumes in the summer months compared to winter for foot (pā€‰=ā€‰0.004, 95Ā % CI [22.2ā€“32.1]), ankle (pā€‰=ā€‰0.0078, 95Ā % CI [20.9ā€“35.5]), and heel pain (pā€‰=ā€‰0.004, 95Ā % CI [29.1ā€“45.6]). These seasonal effects were reflected by data from Australia, with the exception of ankle pain. Annual seasonal effects for treatment options were limited to terms related to foot surgery and ankle orthoses (pā€‰=ā€‰0.031, 95Ā % CI [3.5ā€“20.9]; pā€‰=ā€‰0.004, 95Ā % CI [7.6ā€“25.2] respectively), again increasing in the summer months. CONCLUSIONS: A number of general trends and annual seasonal effects were found in time series internet search data for terms relating to foot and ankle pain. This data may provide insights into these conditions at population levels.     The impacts of suspended mariculture on coastal zones in China and the scope for Integrated Multi-Trophic Aquaculture

    Introduction: China is responsible for more than 60\% of global aquaculture production. As the frontiers of food production have expanded, the cultivation of marine organisms in coastal zones and the open ocean has grown rapidly. The dominant mariculture industry in China is suspended mariculture, which uses net cages, ropes, or other structures suspended in the water column to cultivate aquatic organisms. This systematic, quantitative review provides a clear and comprehensive account of research that has investigated the adverse impacts of suspended mariculture in China and reviews research that has applied Integrated Multi-Trophic Aquaculture (IMTA) systems for mitigating impacts. This work builds on 218 peer reviewed papers that have been published in English-language journals. Outcomes: Eighteen impacts were identified, including chemical, ecological, physical, and socioeconomic impacts. Eighteen measures for improving suspended mariculture were recommended consisting of government department, farm management, and ecological engineering measures. IMTA was the most frequently recommended measure. The capabilities of IMTA for bioremediation and increased farm production were the most frequently studied advantages. Seven other benefits have been explored but remain understudied. The current challenges facing the expansion of commercial IMTA include limited use of new technology, limited skills development, decreasing production of low trophic-level species, biogeographic and temporal barriers, and negative system feedbacks. Conclusion: Despite challenges, implementing commercial IMTA is a promising measure for reducing the impacts of suspended mariculture because it presents a range of secondary benefits that can improve the overall sustainability of aquaculture in the coastal zone

    The Feasibility of Integrating the Noble Scallop Mimachlamys nobilis with Existing Fish Monoculture Farms in the South China Sea: A Bioeconomic Assessment from Hong Kong

    The environmental implications of integrated multitrophic aquaculture have been well studied in China, but few investigations have empirically explored potential economic benefits. This study investigated the technical and economic feasibility of physically integrating the noble scallop Mimachlamys nobilis (Reeve, 1852) with existing fish monoculture farms in Hong Kong. Scallops were grown for 201 days from June to December in lantern nets hung directly from fish farm platforms at treatment depths of 1, 3.5, and 6 m. Only the 1-m treatment attained the target mean height-at-harvest of 80 mm. Fitted von Bertalanffy growth functions showed significant differences in growth performance between depths. The von Bertalanffy growth function projected that the 3.5- and 6-m treatments would require an additional 26 and 59 days of culture to reach 80 mm. Mortality was significantly lower at 1 m (53% Ā± 12.5%) compared with those at 3.5 m (70% Ā± 9.0%) and 6 m (83% Ā± 4.5%). The slower growth and higher mortality at 3.5 and 6mwere probably due to periodically low oxygen which dropped to 4.96, 3.08, and 1.73 mg L-1 at 1, 3.5, and 6 m, respectively, in midsummer. A bioeconomic assessments of two typical farm sizes was conducted; small (45 m2) and large (315 m2). The initial investment, discounted payback time, and 10-y net present value of the projects was US5,485.51,3y,andUS5,485.51, 3 y, and US20,211.33, respectively, for the small farm and US27,659.03,2y,andUS27,659.03, 2 y, and US227,406.49, respectively, for the large farm. Sensitivity analysis revealed that the profitability of operations was sensitive to changes in mortality and sales price. This study has confirmed that physically integrating M. nobilis at existing fish farms is technically and economically feasible

    Extracellular Localisation of the C-Terminus of DDX4 Confirmed by Immunocytochemistry and Fluorescence-Activated Cell Sorting

    Putative oogonial stem cells (OSCs) have been isolated by fluorescence-activated cell sorting (FACS) from adult human ovarian tissue using an antibody against DEAD-box helicase 4 (DDX4). DDX4 has been reported to be germ cell specific within the gonads and localised intracellularly. White et al. (2012) hypothesised that the C-terminus of DDX4 is localised on the surface of putative OSCs but is internalised during the process of oogenesis. This hypothesis is controversial since it is assumed that RNA helicases function intracellularly with no extracellular expression. To determine whether the C-terminus of DDX4 could be expressed on the cell surface, we generated a novel expression construct to express full-length DDX4 as a DsRed2 fusion protein with unique C- and N-terminal epitope tags. DDX4 and the C-terminal myc tag were detected at the cell surface by immunocytochemistry and FACS of non-permeabilised human embryonic kidney HEK 293T cells transfected with the DDX4 construct. DDX4 mRNA expression was detected in the DDX4-positive sorted cells by RT-PCR. This study clearly demonstrates that the C-terminus of DDX4 can be expressed on the cell surface despite its lack of a conventional membrane-targeting or secretory sequence. These results validate the use of antibody-based FACS to isolate DDX4-positive putative OSCs

    Deferiprone versus Deferoxamine in Sickle Cell Disease: Results from a 5-year long-term Italian multi-center randomized clinical trial.

    Blood transfusion and iron chelation currently represent a supportive therapy to manage anemia, vasculopathy and vaso-occlusion crises in Sickle-Cell-Disease. Here we describe the first 5-year long-term randomized clinical trial comparing Deferiprone versus Deferoxamine in patients with Sickle-Cell-Disease. The results of this study show that Deferiprone has the same effectiveness as Deferoxamine in decreasing body iron burden, measured as repeated measurements of serum ferritin concentrations on the same patient over 5-years and analyzed according to the linear mixed-effects model (LMM) (p=0.822). Both chelators are able to decrease, significantly, serum ferritin concentrations, during 5-years, without any effect on safety (p=0.005). Moreover, although the basal serum ferritin levels were higher in transfused compared with non-transfused group (p=0.031), the changes over time in serum ferritin levels were not statistically significantly different between transfused and non-transfused cohort of patients (p=0.389). Kaplan-Meier curve, during 5-years of study, suggests that Deferiprone does not alter survival in comparison with Deferoxamine (p=0.38). In conclusion, long-term iron chelation therapy with Deferiprone was associated with efficacy and safety similar to that of Deferoxamine. Therefore, in patients with Sickle-Cell-Disease, Deferiprone may represent an effective long-term treatment option

    Identification and functional analysis of SKA2 interaction with the glucocorticoid receptor

    Glucocorticoid (GC) receptors (GRs) have profound anti-survival effects on human small cell lung cancer (SCLC). To explore the basis of these effects, protein partners for GRs were sought using a yeast two-hybrid screen. We discovered a novel gene, FAM33A, subsequently identified as a SKA1 partner and involved in mitosis, and so renamed Ska2. We produced an anti-peptide antibody that specifically recognized full-length human SKA2 to measure expression in human cell lines and tissues. There was a wide variation in expression across multiple cell lines, but none was detected in the liver cell line HepG2. A xenograft model of human SCLC had intense staining and archival tissue revealed SKA2 in several human lung and breast tumours. SKA2 was found in the cytoplasm, where it co-localized with GR, but nuclear expression of SKA2 was seen in breast tumours. SKA2 overexpression increased GC transactivation in HepG2 cells while SKA2 knockdown in A549 human lung epithelial cells decreased transactivation and prevented dexamethasone inhibition of proliferation. GC treatment decreased SKA2 protein levels in A549 cells, as did Staurosporine, phorbol ester and trichostatin A; all agents that inhibit cell proliferation. Overexpression of SKA2 potentiated the proliferative response to IGF-I exposure, and knockdown with shRNA caused cells to arrest in mitosis. SKA2 has recently been identified in HeLa S3 cells as part of a complex, which is critical for spindle checkpoint silencing and exit from mitosis. Our new data show involvement in cell proliferation and GC signalling, with implications for understanding how GCs impact on cell fate
