    Gels under stress: the origins of delayed collapse

    Attractive colloidal particles can form a disordered elastic solid or gel when quenched into a two-phase region, if the volume fraction is sufficiently large. When the interactions are comparable to thermal energies the stress-bearing network within the gel restructures over time as individual particle bonds break and reform. Typically, under gravity such weak gels show a prolonged period of either no or very slow settling, followed by a sudden and rapid collapse - a phenomenon known as delayed collapse. The link between local bond breaking events and the macroscopic process of delayed collapse is not well understood. Here we summarize the main features of delayed collapse and discuss the microscopic processes which cause it. We present a plausible model which connects the kinetics of bond breaking to gel collapse and test the model by exploring the effect of an applied external force on the stability of a gel.Comment: Accepted version: 10 pages, 7 figure

    Fishes of the Cocos (Keeling) Islands: new records, community composition and biogeographic significance

    The Cocos (Keeling) Islands comprise the most isolated oceanic atoll in the tropical Indian Ocean and are situated 1000 km south-west of Indonesia. The remoteness of the islands has shaped the composition of marine communities but also limited scientific research. This study summarises field research on the marine fishes of the Cocos (Keeling) Islands over the last 14 years (2001–2014). Sixty-seven new records (from 28 families) are described and raise the total number of known fishes to 602 species from 84 families. New records span a variety of body sizes (3 cm TL Gobiodon unicolor to 500 cm TL Rhincodon typus), were observed in all major habitats,and found at both the Southern Atoll and at North Keeling Island. Notable new records include first records for the families Alopiidae, Coryphaenidae, Eleotridae, Gempylidae, Istiophoridae, Molidae, Polymixiidae, Rhincodontidae, Sillaginidae and Xiphiidae. Sampling from pelagic and deepwater (60–300 m) reef environments significantly increased the number of species described from these habitats. New records include species that have dispersed more than 2500 km (Centropyge acanthops) and dispersal ability appears to explain the lack of syngnathids and the high representation of acanthurids and holocentrids in the community. Some of the Indian Ocean species that have colonised the Cocos (Keeling) Islands now co-occur with their Pacific Ocean sister species, increasing the potential for hybridisation. Although the fish community of the Cocos (Keeling) Island resembles that of the Indo-West Pacific, the isolation and co-occurrence of Indian and Pacific Ocean species distinguishes it from all other locations

    Checklist and new records of Christmas Island fishes: the influence of isolation, biogeography and habitat availability on species abundance and community composition

    Christmas Island (Indian Ocean) is an oceanic high island that is situated 300 km southwest of Java, Indonesia. From 2010 to 2014, the fish community of Christmas Island was surveyed using underwater visual surveys for shallow water (0–60 m) fishes, and line fishing (bottom fishing and trolling) for deepwater (60–300 m) and pelagic fishes. Forty-seven new records (from 22 families) were identified, thereby increasing the total number of fishes described from Christmas Island to 681 (from 91 families). Notable new records include the first records for the families Alopiidae, Anomalopidae, Muraenesocidae, Tetrarogidae and Trichonotidae, and the first reports of Pacific Ocean species Plectranthias yamakawai, and Polylepion russelli in the Indian Ocean. The ten most species-rich families accounted for 58% of the community and included: Labridae (13%), Pomacentridae (8%), Epinephelidae (6%), Acanthuridae (5%), Chaetodontidae (5%), Muraenidae (5%), Gobiidae (5%), Blenniidae (4%), Apogonidae (4%) and Scorpaenidae (3%). The majority (89%) of species inhabit shallow coral reefs, with deep reefs (60–300 m) and pelagic waters only accounting for 7% and 2% of fish community. Approximately 76% of thefishes are widespread Indo-Pacific species, 12% are Pacific Ocean species, 5% are circumtropical, 4% are Indian Ocean species and approximately 1% are endemic. Abundance surveys revealed that endemic species, and species at the edge of their geographic range, do not conform to terrestrial-based predictions of low abundance. The structure and composition of the Christmas Island fish community is influenced by three main factors. Firstly, the isolation of the island means that fishes with poor dispersal abilities (e.g., syngnathids) are underrepresented. Secondly, thebiogeographic position of the island results in a unique mixing of Indian and Pacific Ocean species. Thirdly, the lack of lagoonal habitats means that fishes that use these habitats (e.g., ophichthids, lethrinids, epinephelids) are underrepresented or have low abundance

    Representational predicaments at three Hong Kong sites

    Representational predicaments arise when a job incumbent believes that attributions and images assumed by dominant authorities unfavourably ignore, or disproportionately and unfavourably emphasize, aspects of the incumbent\u27s own work and social identity. This is likely to happen when the incumbent does not have a close relationship with a dominant authority, and when power asymmetries give the former relatively little control over which aspects of their work and social identity are made visible or invisible to the latter. We draw on critical incident interviews from three organizations to illustrate a typology of six types of representational predicament: invasive spotlighting, idiosyncratic spotlighting, embedded background work, paradoxical social visibility, standardization of work processes, and standardization of work outputs. We analyse responses to representational predicaments according to whether they entailed exit, voice, loyalty, or neglect. Incumbents tended to respond with loyalty if they felt able and willing to accommodate their work behaviour and/or social identity to the dominant representations, and if there were sufficient compensatory factors, such as intrinsic rewards from the work or solidarity with colleagues. Exit or neglect appeared to reflect the belief that it was impossible to accommodate. Power asymmetries appeared to deter voice. Individual employees with a close and cordial working relationship with a member of a dominant authority group, or who were relationally networked to one, appeared not to experience representational predicaments

    The Accuracy of Clinical Staging of Stage I-IIIa Non-Small Cell Lung Cancer : An Analysis Based on Individual Participant Data

    BACKGROUND: Clinical staging of non-small cell lung cancer (NSCLC) helps determine the prognosis and treatment of patients; few data exist on the accuracy of clinical staging and the impact on treatment and survival of patients. We assessed whether participant or trial characteristics were associated with clinical staging accuracy as well as impact on survival. METHODS: We used individual participant data from randomized controlled trials (RCTs), supplied for a meta-analysis of preoperative chemotherapy (+/- radiotherapy) vs surgery alone (+/- radiotherapy) in NSCLC. We assessed agreement between clinical TNM (cTNM) stage at randomization and pathologic TNM (pTNM) stage, for participants in the control group. RESULT: Results are based on 698 patients who received surgery alone (+/- radiotherapy) with data for cTNM and pTNM stage. Forty-six percent of cases were cTNM stage I, 23% were cTNM stage II, and 31% were cTNM stage IIIa. cTNM stage disagreed with pTNM stage in 48% of cases, with 34% clinically understaged and 14% clinically overstaged. Agreement was not associated with age (P = .12), sex (P = .62), histology (P = .82), staging method (P = .32), or year of randomization (P = .98). Poorer survival in understaged patients was explained by the underlying pTNM stage. Clinical staging failed to detect T4 disease in 10% of cases and misclassified nodal disease in 38%. CONCLUSION: This study demonstrates suboptimal agreement between clinical and pathologic staging. Discrepancies between clinical and pathologic T and N staging could have led to different treatment decisions in 10% and 38% of cases, respectively. There is therefore a need for further research into improving staging accuracy for patients with stage I-IIIa NSCLC.Peer reviewe

    Tissue Glucocorticoid Metabolism in Adrenal Insufficiency:A Prospective Study of Dual-release Hydrocortisone Therapy

    Background: Patients with adrenal insufficiency (AI) require life-long glucocorticoid (GC) replacement therapy. Within tissues, cortisol (F) availability is under the control of the isozymes of 11β-hydroxysteroid dehydrogenase (11β-HSD). We hypothesize that corticosteroid metabolism is altered in patients with AI because of the nonphysiological pattern of current immediate release hydrocortisone (IR-HC) replacement therapy. The use of a once-daily dual-release hydrocortisone (DR-HC) preparation, (Plenadren®), offers a more physiological cortisol profile and may alter corticosteroid metabolism in vivo.Study Design and Methods: Prospective crossover study assessing the impact of 12 weeks of DR-HC on systemic GC metabolism (urinary steroid metabolome profiling), cortisol activation in the liver (cortisone acetate challenge test), and subcutaneous adipose tissue (microdialysis, biopsy for gene expression analysis) in 51 patients with AI (primary and secondary) in comparison to IR-HC treatment and age- and BMI-matched controls.Results: Patients with AI receiving IR-HC had a higher median 24-hour urinary excretion of cortisol compared with healthy controls (72.1 µg/24 hours [IQR 43.6-124.2] vs 51.9 µg/24 hours [35.5-72.3], P = .02), with lower global activity of 11β-HSD2 and higher 5-alpha reductase activity. Following the switch from IR-HC to DR-HC therapy, there was a significant reduction in urinary cortisol and total GC metabolite excretion, which was most significant in the evening. There was an increase in 11β-HSD2 activity. Hepatic 11β-HSD1 activity was not significantly altered after switching to DR-HC, but there was a significant reduction in the expression and activity of 11β-HSD1 in subcutaneous adipose tissue.Conclusion: Using comprehensive in vivo techniques, we have demonstrated abnormalities in corticosteroid metabolism in patients with primary and secondary AI receiving IR-HC. This dysregulation of pre-receptor glucocorticoid metabolism results in enhanced glucocorticoid activation in adipose tissue, which was ameliorated by treatment with DR-HC

    Assessing smoking status in disadvantaged populations: is computer administered self report an accurate and acceptable measure?

    Background: Self report of smoking status is potentially unreliable in certain situations and in high-risk populations. This study aimed to determine the accuracy and acceptability of computer administered self-report of smoking status among a low socioeconomic (SES) population. Methods: Clients attending a community service organisation for welfare support were invited to complete a cross-sectional touch screen computer health survey. Following survey completion, participants were invited to provide a breath sample to measure exposure to tobacco smoke in expired air. Sensitivity, specificity, positive predictive value and negative predictive value were calculated. Results: Three hundred and eighty three participants completed the health survey, and 330 (86%) provided a breath sample. Of participants included in the validation analysis, 59% reported being a daily or occasional smoker. Sensitivity was 94.4% and specificity 92.8%. The positive and negative predictive values were 94.9% and 92.0% respectively. The majority of participants reported that the touch screen survey was both enjoyable (79%) and easy (88%) to complete. Conclusions: Computer administered self report is both acceptable and accurate as a method of assessing smoking status among low SES smokers in a community setting. Routine collection of health information using touch-screen computer has the potential to identify smokers and increase provision of support and referral in the community setting

    A Novel Signaling Network Essential for Regulating Pseudomonas aeruginosa Biofilm Development

    The important human pathogen Pseudomonas aeruginosa has been linked to numerous biofilm-related chronic infections. Here, we demonstrate that biofilm formation following the transition to the surface attached lifestyle is regulated by three previously undescribed two-component systems: BfiSR (PA4196-4197) harboring an RpoD-like domain, an OmpR-like BfmSR (PA4101-4102), and MifSR (PA5511-5512) belonging to the family of NtrC-like transcriptional regulators. These two-component systems become sequentially phosphorylated during biofilm formation. Inactivation of bfiS, bfmR, and mifR arrested biofilm formation at the transition to the irreversible attachment, maturation-1 and -2 stages, respectively, as indicated by analyses of biofilm architecture, and protein and phosphoprotein patterns. Moreover, discontinuation of bfiS, bfmR, and mifR expression in established biofilms resulted in the collapse of biofilms to an earlier developmental stage, indicating a requirement for these regulatory systems for the development and maintenance of normal biofilm architecture. Interestingly, inactivation did not affect planktonic growth, motility, polysaccharide production, or initial attachment. Further, we demonstrate the interdependency of this two-component systems network with GacS (PA0928), which was found to play a dual role in biofilm formation. This work describes a novel signal transduction network regulating committed biofilm developmental steps following attachment, in which phosphorelays and two sigma factor-dependent response regulators appear to be key components of the regulatory machinery that coordinates gene expression during P. aeruginosa biofilm development in response to environmental cues
