    Salmonellosis in people is a costly disease, much of it occurring because of food associated exposure. We develop a farm-to-fork model which estimates the pork associated Salmonella risk and human health costs. This analysis focuses on the components of the pork production chain up to the point of producing a chilled pork carcass. Sensitivity and scenario analysis show that changes that occur in Salmonella status during processing are substantially more important for human health risk and have a higher benefit/cost ratio for application of strategies that control Salmonella compared with on-farm strategies.Food Consumption/Nutrition/Food Safety,

    Diet-induced obesity impairs mammary development and lactogenesis in murine mammary gland

    We have developed a mouse model of diet-induced obesity that shows numerous abnormalities relating to mammary gland function. Animals ate 40% more calories when offered a high-fat diet and gained weight at three times the rate of controls. They exhibited reduced conception rates, increased peripartum pup mortality, and impaired lactogenesis. The impairment of lactogenesis involved lipid accumulation in the secretory epithelial cells indicative of an absence of copius milk secretion. Expression of mRNAs for -casein, whey acid protein, and -lactalbumin were all decreased immediately postpartum but recovered as lactation was established over 2–3 days. Expression of acetyl-CoA carboxylase (ACC)- mRNA was also decreased at parturition as was the total enzyme activity, although there was a compensatory increase in the proportion in the active state. By day 10 of lactation, the proportion of ACC in the active state was also decreased in obese animals, indicative of suppression of de novo fatty acid synthesis resulting from the supply of preformed fatty acids in the diet. Although obese animals consumed more calories in the nonpregnant and early pregnant states, they showed a marked depression in fat intake around day 9 of pregnancy before food intake recovered in later pregnancy. Food intake increased dramatically in both lean and obese animals during lactation although total calories consumed were identical in both groups. Thus, despite access to high-energy diets, the obese animals mobilized even more adipose tissue during lactation than their lean counterparts. Obese animals also exhibited marked abnormalities in alveolar development of the mammary gland, which may partially explain the delay in differentiation evident during lactogenesis

    Phylogeography of the crown-of-thorns starfish in the Indian Ocean

    Background: Understanding the limits and population dynamics of closely related sibling species in the marine realm is particularly relevant in organisms that require management. The crown-of-thorns starfish Acanthaster planci, recently shown to be a species complex of at least four closely related species, is a coral predator infamous for its outbreaks that have devastated reefs throughout much of its Indo-Pacific distribution. Methodology/Principal Findings: In this first Indian Ocean-wide genetic study of a marine organism we investigated the genetic structure and inferred the paleohistory of the two Indian Ocean sister-species of Acanthaster planci using mitochondrial DNA sequence analyses. We suggest that the first of two main diversification events led to the formation of a Southern and Northern Indian Ocean sister-species in the late Pliocene-early Pleistocene. The second led to the formation of two internal clades within each species around the onset of the last interglacial. The subsequent demographic history of the two lineages strongly differed, the Southern Indian Ocean sister-species showing a signature of recent population expansion and hardly any regional structure, whereas the Northern Indian Ocean sister-species apparently maintained a constant size with highly differentiated regional groupings that were asymmetrically connected by gene flow. Conclusions/Significance: Past and present surface circulation patterns in conjunction with ocean primary productivity were identified as the processes most likely to have shaped the genetic structure between and within the two Indian Ocean lineages. This knowledge will help to understand the biological or ecological differences of the two sibling species and therefore aid in developing strategies to manage population outbreaks of this coral predator in the Indian Ocean

    Contrasting Demographic History and Phylogeographic Patterns in Two Indo-Pacific Gastropods

    Marine species with ranges that span the Indo-Australian Archipelago (IAA) exhibit a range of phylogeographic patterns, most of which are interpreted in the context of vicariance between Indian and Pacific Ocean populations during Pliocene and Pleistocene low sea level stands. However, patterns often vary among ecologically similar taxa, sometimes even within genera. This study compares phylogeographic patterns in two species of highly dispersive Neritid gastropod, Nerita albicilla and Nerita plicata, with nearly sympatric ranges that span the Indo-Pacific. Mitochondrial COI sequences from \u3e 1000 individuals from 97 sites reveal similar phylogenies in both species (two divergent clades differing by 3.2% and 2.3%, for N. albicilla and N. plicata respectively). However, despite ecological similarity and congeneric status, the two species exhibit phylogeographic discordance, with N. albicilla maintaining reciprocal monophyly of Indian and Pacific Ocean populations, while N. plicata is panmictic between oceans, but displays a genetic cline in the Central Pacific. Although this difference might be explained by qualitatively different demographic histories, parameter estimates from three coalescent models indicate that both species have high levels of gene flow between demes (2Ne m \u3e 75), and share a common history of population expansion that is likely associated with cyclical flooding of continental shelves and island lagoons following low sea level stands. Results indicate that ecologically similar co-distributed species may respond very differently to shared environmental processes, suggesting that relatively minor differences in traits such as pelagic larval duration or microhabitat association may profoundly impact phylogeographic structure

    Universality in the partially anisotropic three-dimensional Ising lattice

    Using transfer-matrix extended phenomenological renormalization-group methods the critical properties of spin-1/2 Ising model on a simple-cubic lattice with partly anisotropic coupling strengths J=(J,J,J){\vec J}=(J',J',J) are studied. Universality of both fundamental critical exponents yty_t and yhy_h is confirmed. It is shown that the critical finite-size scaling amplitude ratios U=Aχ(4)Aκ/Aχ2U=A_{\chi^{(4)}}A_\kappa/A_\chi^2, Y1=Aκ/AχY_1=A_{\kappa^{''}}/A_\chi, and Y2=Aκ(4)/Aχ(4)Y_2=A_{\kappa^{(4)}}/A_{\chi^{(4)}} are independent of the lattice anisotropy parameter Δ=J/J\Delta=J'/J. By this for the last above invariant of the three-dimensional Ising universality class we give the first quantitative estimate: Y22.013Y_2\simeq2.013 (shape L×L×L\times L\times\infty, periodic boundary conditions in both transverse directions).Comment: 11 pages in latex; no figure

    Regional differentiation and post-glacial expansion of the Atlantic silverside, Menidia menidia, an annual fish with high dispersal potential

    The coastal marine environment of the Northwest Atlantic contains strong environmental gradients that create distinct marine biogeographic provinces by limiting dispersal, recruitment, and survival. This region has also been subjected to numerous Pleistocene glacial cycles, resulting in repeated extirpations and recolonizations in northern populations of marine organisms. In this study, we examined patterns of genetic structure and historical demography in the Atlantic silverside, Menidia menidia, an annual marine fish with high dispersal potential but with well-documented patterns of clinal phenotypic adaptation along the environmental gradients of the Northwest Atlantic. Contrary to previous studies indicating genetic homogeneity that should preclude regional adaptation, results demonstrate subtle but significant (F(ST) = 0.07; P < 0.0001) genetic structure among three phylogeographic regions that partially correspond with biogeographic provinces, suggesting regional limits to gene flow. Tests for non-equilibrium population dynamics and latitudinal patterns in genetic diversity indicate northward population expansion from a single southern refugium following the last glacial maximum, suggesting that phylogeographic and phenotypic patterns have relatively recent origins. The recovery of phylogeographic structure and the partial correspondence of these regions to recognized biogeographic provinces suggest that the environmental gradients that shape biogeographic patterns in the Northwest Atlantic may also limit gene flow in M. menidia, creating phylogeographic structure and contributing to the creation of latitudinal phenotypic clines in this species.
