    WOU Community Health + Polk County Family Thrive Kits: Social Marketing Campaign To Prevent COVID-19 Among Latinx Families

    The WOU Community Health + Polk County COVID-19 Project is a partnership between Western Oregon University and Polk County Public Health Services that provides contact tracing and outreach efforts to Polk County, OR. During this pandemic, Latinx families have been disproportionately affected by COVID-19. For this reason, our team has developed Family Thrive kits that follow health literacy guidelines and provide culturally appropriate information for families in English and Spanish. The purpose of the Family Thrive kit is to provide information and materials that encourage safe COVID-19 practices and family well-being. Information provided is tailored to the needs of families with children and focusing on several health issues and local resources. As of June 2021, we will have collaborated with more than 20 community-based organizations (CBO’s) and provided more than 2,246 kits to families throughout Polk County. We continue developing materials and building relationships with new CBO’s to raise awareness of COVID-19 prevention strategies

    Fate mapping melanoma persister cells through regression and into recurrent disease in adult zebrafish

    Melanoma heterogeneity and plasticity underlie therapy resistance. Some tumour cells possess innate resistance, while others reprogramme during drug exposure and survive to form persister cells, a source of potential cancer cells for recurrent disease. Tracing individual melanoma cell populations through tumour regression and into recurrent disease remains largely unexplored, in part, because complex animal models are required for live imaging of cell populations over time. Here, we applied tamoxifen-inducible cre(ERt2)/loxP lineage tracing to a zebrafish model of MITF-dependent melanoma regression and recurrence to image and trace cell populations in vivo through disease stages. Using this strategy, we show that melanoma persister cells at the minimal residual disease site originate from the primary tumour. Next, we fate mapped rare MITF-independent persister cells and demonstrate that these cells directly contribute to progressive disease. Multiplex immunohistochemistry confirmed that MITF-independent persister cells give rise to Mitfa(+) cells in recurrent disease. Taken together, our work reveals a direct contribution of persister cell populations to recurrent disease, and provides a resource for lineage-tracing methodology in adult zebrafish cancer models

    The WOU Community Health + Polk County COVID-19 project is a local example of how Academic Health Departments can serve as public health training sites for students of public health and address the public health needs of the local community. An academic health department (AHD) represents a formal affiliation between an academic institution and a public health practice organization, such as a local health department. The WOU Community Health + Polk County COVID-19 project team consists of WOU faculty in Community Health, recent alumni, undergraduate students, and Polk County Public Health. This session will review the outcomes, challenges, and lessons learned from a one-year health communication and contact tracing intervention in Polk County, Oregon. How can undergraduate students, faculty, and community partners build a network for student success and community well-being? We share our publicly available health communication campaign materials and discuss students’ perspectives on professional development and networking opportunities within the project

    Loss of the chromatin modifier Kdm2aa causes BrafV600E-independent spontaneous melanoma in zebrafish.

    KDM2A is a histone demethylase associated with transcriptional silencing, however very little is known about its in vivo role in development and disease. Here we demonstrate that loss of the orthologue kdm2aa in zebrafish causes widespread transcriptional disruption and leads to spontaneous melanomas at a high frequency. Fish homozygous for two independent premature stop codon alleles show reduced growth and survival, a strong male sex bias, and homozygous females exhibit a progressive oogenesis defect. kdm2aa mutant fish also develop melanomas from early adulthood onwards which are independent from mutations in braf and other common oncogenes and tumour suppressors as revealed by deep whole exome sequencing. In addition to effects on translation and DNA replication gene expression, high-replicate RNA-seq in morphologically normal individuals demonstrates a stable regulatory response of epigenetic modifiers and the specific de-repression of a group of zinc finger genes residing in constitutive heterochromatin. Together our data reveal a complex role for Kdm2aa in regulating normal mRNA levels and carcinogenesis. These findings establish kdm2aa mutants as the first single gene knockout model of melanoma biology
