    The Vatican and ecospirituality: tensions, promises and possibilities for fostering an emerging green Catholic spirituality

    A partir del mensaje de Juan Pablo II en el Día Mundial de la Paz de en 1990 sobre la crisis ecológica, han sido frecuentes los esfuerzos de divulgación del Vaticano con respecto a temas relacionados al medio ambiente. Basándose en una selección de documentos de la doctrina Social Católica a estos temas, este artículo explora las reacciones del Vaticano sobre argumentos relacionados a la ecoespiritualidad. A través de un lente crítico basado en la ética ecoteológica asimismo que explorando una versión particular de ecoespiritualidad particularmente referida a la condenación del biocentrismo, se pondrá de manifiesto los esfuerzos oficiales de reverdecer la política de la Iglesia Apostólica Romana y las prácticas de la fe que a este tema se refieren. Tomando como punto de partida la prerrogativa de una distancia crítica desde una enfoque Norteamericano, esta ponencia sugiere maneras en las que el Vaticano puede fomentar un reverdecimiento mas integral y pacífico de las doctrina Social Católica y de la Espiritualidad Católica recurriendo a fuentes dentro de la tradición intelectual Católica asimismo que basándose en otras expresiones de ecoespiritualidad, ecoteología y documentos de Educación Social Católica de conferencias de obispos en Europa y la Américas.Since John Paul II’s 1990 World Day of Peace Message on the ecological crisis, green themes have been a recurring feature of the Vatican’s public teachings. Working with a selection of Catholic Social Teaching documents, this article explores the Vatican’s reactions to and accommodations of ecospirituality. A critical lens informed by ecotheological ethics is employed to analyse the Vatican’s specific brand of ecospirituality, particularly as it relates to its condemnation of biocentrism, while also acknowledging official efforts to green the Roman Catholic Church’s doctrine and faith-based practices. With the advantage of the critical distance that a North American perspective provides, this article suggests ways that the Vatican can foster a more integral and substantively peaceful greening of Catholic Social Teaching and Catholic spirituality by drawing on resources from within Catholic intellectual tradition, as well as other expressions of ecospirituality, ecotheology, and Catholic Social Teaching documents from local bishops’ conferences in Europe and the Americas

    Molecular and functional correction of a deep intronic splicing mutation in CFTR by CRISPR-Cas9 gene editing

    Cystic fibrosis (CF) is an autosomal recessive disorder caused by mutations in the CFTR gene. The 10th most common mutation, c.3178-2477C>T (3849+10kb C>T), involves a cryptic, intronic splice site. This mutation was corrected in CF primary cells homozygous for this mutation by delivering pairs of guide RNAs (gRNAs) with Cas9 protein in ribonucleoprotein (RNP) complexes that introduce double-strand breaks to flanking sites to excise the 3849+10kb C>T mutation, followed by DNA repair by the non-homologous end-joining pathway, which functions in all cells of the airway epithelium. RNP complexes were delivered to CF basal epithelial cell by a non-viral, receptor-targeted nanocomplex comprising a formulation of targeting peptides and lipids. Canonical CFTR mRNA splicing was, thus, restored leading to the restoration of CFTR protein expression with concomitant restoration of electrophysiological function in airway epithelial air-liquid interface cultures. Off-target editing was not detected by Sanger sequencing of in silico-selected genomic sites with the highest sequence similarities to the gRNAs, although more sensitive unbiased whole genome sequencing methods would be required for possible translational developments. This approach could potentially be used to correct aberrant splicing signals in several other CF mutations and other genetic disorders where deep-intronic mutations are pathogenic

    Complete genome sequence of Rhizobium leguminosarum bv trifolii strain WSM2304, an effective microsymbiont of the South American clover Trifolium polymorphum.

    Rhizobium leguminosarum bv trifolii is the effective nitrogen fixing microsymbiont of a diverse range of annual and perennial Trifolium (clover) species. Strain WSM2304 is an aerobic, motile, non-spore forming, Gram-negative rod, isolated from Trifolium polymorphum in Uruguay in 1998. This microsymbiont predominated in the perennial grasslands of Glencoe Research Station, in Uruguay, to competitively nodulate its host, and fix atmospheric nitrogen. Here we describe the basic features of WSM2304, together with the complete genome sequence, and annotation. This is the first completed genome sequence for a nitrogen fixing microsymbiont of a clover species from the American center of origin. We reveal that its genome size is 6,872,702 bp encoding 6,643 protein-coding genes and 62 RNA only encoding genes. This multipartite genome was found to contain 5 distinct replicons; a chromosome of size 4,537,948 bp and four circular plasmids of size 1,266,105 bp, 501,946 bp, 308,747 bp and 257,956 bp

    Effects of a home-based intervention on diet and physical activity behaviours for rural adults with or at risk of metabolic syndrome: a randomised controlled trial

    BACKGROUND: This study aimed to determine whether a home-based 6-month lifestyle intervention program complemented by motivational interviewing could improve diet and physical activity behaviours in 50-69 year olds with or at risk of metabolic syndrome, residing in a disadvantaged rural Western Australian community. METHODS: Participants from the City of Albany and surrounding towns (n?=?401) were recruited into a 6 month randomised controlled trial. They were screened for metabolic syndrome and randomly allocated to intervention (n?=?201) or control group (n?=?200). Baseline and post-test data collection for both groups included a self-report questionnaire which incorporated the Fat and Fibre Barometer and the International Physical Activity Questionnaire Short Form. The intervention group received the program materials at baseline and the control group was waitlisted. Generalised estimating equation models assessed repeated outcome measures over time. RESULTS: A total of 151 (75.1 %) intervention and 159 (79.5 %) control group participants completed post-test and were included in the analysis. After controlling for confounders, the intervention group achieved a marginally significant increase in their metabolic equivalent (MET) minutes of moderate intensity physical activity per week (p?=?0.049), and significantly improved fibre intake (p?<?0.001), fat intake (p?=?0.003), and vegetable serves per day (p?=?0.002) from baseline to post-test relative to the control group. CONCLUSION: A home-based, low-cost intervention with motivational support can effectively improve the physical activity and dietary behaviours of adults aged 50-69 years with or at risk of metabolic syndrome residing in a disadvantaged rural area.     Feasibility of a Transient Dynamic Design Analysis Method

    This article addresses the degree of success that may be achieved by using simple equipment–vehicle models that produce time history responses whose equipment fixed base modal maximum response values are equivalent to those found in the U.S. Navy's dynamic design analysis method. The criteria of success is measured by a comparison of the results with typical interim shock design values. The equipment models reported are limited to two- degree and three degrees of freedom systems; the model of the vehicle to which the equipment is attached consists solely of a rigid mass and an elastic spring; and the shock excitation is produced by an ideal impulse that is applied to the vehicle mass
