    Plasma cortisol level attributable to physical exercise in endurance horses

    O objetivo deste trabalho foi relacionar a intensidade do exercício físico e as concentrações de cortisol plasmático em cavalos de enduro, uma competição em que somente animais experientes podem competir nas provas mais longas. Foram utilizados 30 equinos Puro Sangue Árabe e mestiços Árabe, machos ou fêmeas participantes de provas de enduro. Foram divididos em três grupos de 10 animais: (G1): percorreram mais de 100km, (G2): percorreram menos de 100km, e (G3): desqualificados por causa metabólica. Foram realizadas dosagens de cortisol plasmático em três momentos diferentes: (t0): dia anterior à competição, (t1): 30 a 60 minutos após o término da prova e, (t2): 90 a 120 minutos após o término da prova. Concluiu-se que o enduro leva ao aumento do cortisol plasmático; animais que percorrem maiores distâncias apresentam menor aumento das concentrações de cortisol; animais desqualificados por\ud causa metabólica, que passam por situações de extremo esforço físico, tendem a valores de cortisol mais elevados e animais menos experientes apresentam valores de cortisol mais elevados mesmo tendo percorrido menores distâncias.The objective of this study was to establish a relationship between exercise and plasma cortisol levels in endurance horses. A race in which only experienced animals may run longer distances. Thirty male and female, Arabians and crossbred Arabian horses that compete in endurance races were used. They were divided into three groups of 10 animals each: G1, ran more than 100km; G2, ran less than 100km; and G3, disqualified due to metabolic problems. Plasma cortisol was quantified at three different moments: t0, the day before the competition; t1, 30 to 60 minutes after\ud the end of the circuit; and t2, 90 to 120 minutes after the end of the circuit.     Factors Associated With Red Blood Cell Transfusions In Very-low-birth-weight Preterm Infants In Brazilian Neonatal Units

    Background: Preterm infants in neonatal intensive care units frequently receive red blood cells (RBC) transfusions due to the anemia of prematurity. A number of variables related to gestational age, severity of illness and transfusion practices adopted in the neonatal unit where the neonate was born may contribute to the prescription of RBC transfusions. This study aimed to analyse the frequency and factors associated with RBC transfusions in very-low-birth-weight preterm infants. Methods: A prospective cohort of 4283 preterm infants (gestational age: 29.9 +/- 2.9 weeks; birth weight: 1084 +/- 275 g) carried out at 16 university hospitals in Brazil between January 2009 and December 2011 was analysed. Factors associated with RBC transfusions were evaluated using univariate and multiple logistic regression analysis. Results: A total of 2208 (51.6 %) infants received RBC transfusions (variation per neonatal unit: 34.1 % to 66.4 %). RBC transfusions were significantly associated with gestational age (OR: -1.098; 95% CI: -1.12 to -1.04), SNAPPE II score (1.01; 1.00-1.02), apnea (1.69; 1.34-2.14), pulmonary hemorrhage (2.65; 1.74-4.031), need for oxygen at 28 days of life (1.56; 1.17-2.08), clinical sepsis (3.22; 2.55-4.05), necrotising enterocolitis (3.80; 2.26-6.41), grades III/IV intraventricular hemorrhage (1.64; 1.05-2.58), mechanical ventilation (2.27; 1.74-2.97), use of umbilical catheter (1.86; 1.35-2.57), parenteral nutrition (2.06; 1.27-3.33), > 60 days of hospitalization (5.29; 4.02-6.95) and the neonatal unit where the neonate was born. Conclusions: The frequency of RBC transfusions varied among neonatal intensive care units. Even after adjusting for adverse health conditions and therapeutic interventions, the neonatal unit continued to influence transfusion practices in very-low birth-weight infants.1

    Comparison between traditional judgement and jumping kinematic evaluation of Brazilian Sport horses

    O objetivo do presente trabalho foi descrever as características cinemáticas do salto de cavalos da raça Brasileiro de Hipismo, avaliar o julgamento realizado pelos juízes durante uma competição de salto em liberdade e comparar essas avaliações. Foram utilizados 13 equinos da raça Brasileiro de Hipismo, machos não castrados (40,15±4,24 meses, 491,1±40,89 kg). Os animais foram filmados saltando um obstáculo oxer durante uma competição de saltos em liberdade. Trajetórias do centro de massa corporal (CM) foram calculadas através da posição de pontos anatômicos que foram rastreados manualmente desde a decolagem até o pouso. Dessa trajetória, variáveis biomecânicas foram calculadas: velocidade vertical e horizontal, ângulos da velocidade e do deslocamento do CM na decolagem, altura máxima, deslocamento vertical e horizontal do CM, altura e fração de encurtamento dos membros sobre o obstáculo. Um polinômio de segunda ordem foi calculado através da trajetória do CM para obter três constantes com base na equação resultante. Os cavalos foram avaliados por três juízes: medidas zoométricas, funcionalidade, morfologia, salto, genealogia e modelo de garanhão. As notas de julgamento são consistentes entre si, mas são tendenciosas. As variáveis biomecânicas apresentaram alta repetibilidade e baixo erro. Nenhuma das notas de funcionalidade, genealogia e salto se correlacionam com as variáveis biomecânicas de salto. O julgamento do salto realizado não apresenta relação com as variáveis biomecânicas medidas. A análise cinemática não apresentou viés e foi muito consistente, podendo ser uma ferramenta para tornar o julgamento mais objetivo.The objective of this study was to describe the jump kinematic characteristics of Brazilian Sport horses, evaluate the judgment conducted during a free jump competition and compare these assessments. Thirteen Brazilian Sport Horses stallions (40.15 ± 4.24 months, 491.1 ± 40.89 kg) were filmed jumping an obstacle during a competition. Body center of mass (CM) trajectories were calculated by the position of anatomical landmarks that were manually tracked from takeoff to landing. From this trajectory, biomechanical variables were calculated: vertical and horizontal velocities, angles of the velocity and of the displacement of the CM at takeoff, maximum height, vertical and horizontal displacement of the CM, height and shortening rate of the limbs over the obstacle. A second order polynomial was calculated to obtain three coefficients. All horses were evaluated by three judges: body measures, functionality, morphology, jumping, genealogy and stallion model. Judges are consistent with each other, but they are biased. The biomechanical variables showed high repeatability and low error. None of the functionality, genealogy and jumping scores correlate with jumping biomechanical variables. The judgement is not associated with the jumping biomechanical variables measured. Kinematic analysis showed no bias and was very consistent therefore being helpful for future sport horses judgement

    Avaliação ecocardiográfica do desenvolvimento cardíaco de cavalos atletas: comparação entre atividades físicas distintas

    A capacidade aeróbica dos equinos atletas pode ser explicada, em partes, pelo seu desenvolvimento cardíaco associado ao treinamento físico. O exame ecocardiográfico de equinos atletas permite a visualização destas alterações e da função cardíaca destes animais. Buscou-se avaliar ecocardiograficamente equinos de diferentes modalidades esportivas, para verificar se o tipo de atividade física desempenhada por cada grupo de animais promove diferenças estruturais ou funcionais cardíacas. 196 equinos foram submetidos à avaliação ecocardiográfica em repouso, sendo divididos em três grupos: grupo Polo, 44 equinos participantes de provas de Polo; grupo QM, 49 equinos da raça Quarto de Milha, participantes de provas de baliza e tambor, e grupo PSI, 103 equinos da raça Puro Sangue Inglês participantes de provas de corridas. Foram avaliados os seguintes índices cardíacos: SIV, DIVE e PLVE em sístole e diástole, AE em sístole, Ao em diástole, relação AE:Ao, FEj, VEj, VSFVE, VDFVE, FS%, E-S, TEVE, DC e FC. Os animais do grupo PSI apresentaram os maiores volumes de ventrículo esquerdo, tendo apresentado proporcionalmente menor eficiência quando analisado o VEj. Os animais do grupo Polo e QM apresentaram melhores valores quando analisadas as variáveis de função cardíaca. A atividade física mais intensa desempenhada pelos animais do grupo QM e Polo permitiu melhor desenvolvimento funcional do coração destes animais, tendo o grupo PSI, apresentado valores que sugerem que sua capacidade aeróbica ainda pode ser melhor explorada através de melhor orientação de seu treinamento físico

    Differential expression of apoptosis-related genes from death receptor pathway in chronic myeloproliferative diseases

    Background Chronic myeloproliferative disorders (MPDs) are clonal haematopoietic stem cell malignancies characterised by an accumulation of mature myeloid cells in bone marrow and peripheral blood. Deregulation of the apoptotic machinery may be associated with MPD physiopathology. Aims To evaluate expression of death receptors` family members, mononuclear cell apoptosis resistance, and JAK2 allele burden. Subjects and Methods Bone marrow haematopoietic progenitor CD34 cells were separated using the Ficoll-hypaque protocol followed by the Miltenyi CD34 isolation kit, and peripheral blood leukocytes were separated by the Haes-Steril method. Total RNA was extracted by the Trizol method, the High Capacity Kit was used to synthesise cDNA, and real-time PCR was performed using SybrGreen in ABIPrism 7500 equipment. The results of gene expression quantification are given as 2(-Delta Delta Ct). The JAK2 V617F mutation was detected by real-time allelic discrimination PCR assay. Peripheral blood mononuclear cells (PBMCs) were isolated by the Ficoll-hypaque protocol and cultured in the presence of apoptosis inducers. Results In CD34 cells, there was mRNA overexpression for fas, faim and c-flip in polycythaemia vera (PV), essential thrombocythaemia (ET) and primary myelofibrosis (PMF), as well as fasl in PMF, and dr4 levels were increased in ET. In leukocytes, fas, c-flip and trail levels were increased in PV, and dr5 expression was decreased in ET. There was an association between dr5 and fasl expression and JAK2V617F mutation. PBMCs from patients with PV, ET or PMF showed resistance to apoptosis inducers.
