    Twists of opposite handedness on a scroll wave

    The dynamic interaction of scroll waves in the Belousov-Zhabotinsky reaction with a vertically orientated gradient of excitability is studied by optical tomography. This study focuses on scroll waves, whose filaments were oriented almost perpendicular to the gradient. Whereas scroll waves with filaments exactly perpendicular to the gradient remain unaffected, filaments with a component parallel to the gradient develop a twist. Scroll waves with U-shaped filaments exhibit twists starting from both of its ends, resulting in scroll waves whose filaments display a pair of twists of opposite handedness. These twists are separated by a nodal plane where the filament remains straight and untwisted. The experimental findings were reproduced by numerical simulations using the Oregonator model and a linear gradient of excitability almost perpendicular to the orientation of the filament.Peer ReviewedPostprint (published version

    Farmacovigilancia de psicofármacos en una farmacia de A Coruña (España)

    Objetivos: Realizar una actividad de farmacovigilancia (FV) en una farmacia comunitaria, registrando las reacciones adversas (RAM) y derivando a los pacientes a su médico si fuese necesario.Material y métodos: Estudio observacional transversal. En un cuestionario ad hoc se recogieron datos demográficos de los pacientes sobre la utilización de los medicamentos, percepción sobre efectividad y seguridad de los fármacos a estudiar. Se seleccionaron cuatro principios activos: duloxetina, venlafaxina, quetiapina y olanzapina por ser de dispensación frecuente en la farmacia y de reciente comercialización.Resultados: 47 pacientes utilizaban uno de los antidepresivos estudiados. 34 (72,3%) mujeres y 13 (27,7%) hombres. 21 (55,3%) tomaban venlafaxina y 26 (44,7%) duloxetina. Media de edad 52,9 (DE= 13,7) años (21-80). 36 en tratamiento con los dos antipsicóticos del estudio. 21 (58,3%) utilizaban olanzapina y 15 (41,8%) quetiapina. Edad media 41,6 (DE=13,4) años (27-70).174 RAM en los antidepresivos y 120 en los antipsicóticos. Sequedad de boca, temblores, hipersudoración, flatulencia y ansiedad son las más frecuentes en los dos antidepresivos y sequedad de boca, reducción de la libido y aumento de peso en los dos antipsicóticos.Se notificó 1 caso al Centro Gallego de Farmacovigilancia. Se derivaron dos pacientes al médico por problemas relacionados con la medicación (PRM).Conclusiones: Mediante una actividad de FV en una farmacia comunitaria de A Coruña se ha detectado un elevado número de RAM, contribuyendo a un resultado más seguro de la farmacoterapia de los pacientes

    Mechanism of action studies on new ligands for soluble guanylate cyclase.

    This thesis will comprise two topics. The first part relates to the application of photoaffinity labelling to soluble guanylate cyclase (sGC), which belongs to a family of enzymes that catalyse the conversion of guanosine-5'-triphosphate (GTP) to cyclic guanosine-3', 5'- monophosphate (cGMP). By formation of the intracellular signalling molecule cGMP, sGC plays a key role in smooth muscle relaxation and inhibition of platelet aggregation and in many signal transduction pathways. Although there are two types of activators of sGC, NO-dependent (nitrovasodilators such as sodium nitroprusside, DEANO, etc.) and NO-independent (YC-1, l-benzyl-3- 5'-hydroxymethyl-2'-furyl indazole), the mechanism of action of only the former is understood. This thesis describes an attempt to locate residues that make up the binding sites for these activators. This comprises the design, synthesis and biological evaluation of small molecule activators, and their photolabeled analogues, followed by photoafiinity labelling experiments using tryptic digestion and mass spectroscopy (MALDI) for the identification of peptide fragments covalently bonded to activator analogue. The second part involves the investigation of a traceless linker designed for use in the solid phase synthesis of sGC modulators. The resulting method could also be applied in the synthesis of a variety of biologically active molecules. We wanted to design a linker which would combine the advantages of a traceless and a "safety-catch" linker. Therefore we investigated the application of a benzoisothiazol-based linker as this heterocycle is stable until activated by oxidation of the sulphur atom, allowing facile cleavage in the last step of the synthesis
