Presence of Gardnerella vaginalis in healthy portuguese women: a pilot study
Bacterial vaginosis (BV) has an important position worldwide, as the leading vaginal disorder in women, and affects 30-50% of African women and 10-20% of White women of reproductive age. This condition although not mortal causes great discomfort and may lead to other complications such as pre-term labour or increase susceptibility for HIV infection.
During BV occur a decrease of Lactobacillus spp. present in the vaginal epithelium and an increase in the number of anaerobic microorganisms like Gardnerella vaginalis, Pretovella spp., Mobilincus spp.; Mycoplasma hominis and Atopobium vaginae. Gardnerella vaginalis is also responsible for the formation of a biofilm in the vaginal epithelium in sick womenâs. However, the direct correlation between the pathology and the causing agent (or agents) has not been clearly established.
Currently there is only one article in PubMed (Guerreiro et. al, 1998) referring to the prevalence of bacterial vaginosis in Portugal and we aim to extend the research in this field specifically to the portuguese population. As part of this effort one of our aims it to characterise the bacterial population of portuguese women both healthy and diagnosed with BV. As such we collected swab samples of vaginal fluids from protuguese women with the help of health professionals and using self collection. The swabs were collected and treated within 24 hours at the University of Minho for the characterization of the bacterial population present, by using conventional microbiological growth techniques, PNA-FISH microscopy and 16S PCR.
It was found that about 20% of the samples tested possessed G. vaginalis and all possessed Lactobacillus spp. using all 3 identification techniques described. This result is consistent with previous reports of G. vaginalis prevalence althought slightly lower, and shows that traditional microbiological techniques, microscopy and molecular methods were consistent in terms of results
Access to infertility consultations: what women tell us about it?
The main objective of the present paper is to evaluate the perception of women concerning the barriers and access to infertility consultations. Socio cultural and economic access to infertility consultations is detached and three municipalities of the northwest of Portugal were chosen as an example of a peripheral country. A quantitative/qualitative study was done with 60 women. Three dimensions were evaluated: geographic and structural and functional access; economic access; and sociocultural access. The main barriers were mainly identified in the last two dimensions. The economic access was the less well evaluated by women being the cost of treatment (medication, and concentration of costs in a short period) difficult to bear. This can justify a greater involvement of the Portuguese Government, by developing policies for the reimbursement of part of the costs. Also, some changes in structural and functional access must be done with special regard to the separation of the infertility consultations from the reproductive medicine section. The setting of the teams, with a follow-up by the same team of health professionals is also needed
Searching for molecular markers in head and neck squamous cell carcinomas (HNSCC) by statistical and bioinformatic analysis of larynx-derived SAGE libraries
Background: Head and neck squamous cell carcinoma (HNSCC) is one of the most common malignancies in humans. The average 5-year survival rate is one of the lowest among aggressive cancers, showing no significant improvement in recent years. When detected early, HNSCC has a good prognosis, but most patients present metastatic disease at the time of diagnosis, which significantly reduces survival rate. Despite extensive research, no molecular markers are currently available for diagnostic or prognostic purposes. Methods: Aiming to identify differentially-expressed genes involved in laryngeal squamous cell carcinoma (LSCC) development and progression, we generated individual Serial Analysis of Gene Expression (SAGE) libraries from a metastatic and non-metastatic larynx carcinoma, as well as from a normal larynx mucosa sample. Approximately 54,000 unique tags were sequenced in three libraries. Results: Statistical data analysis identified a subset of 1,216 differentially expressed tags between tumor and normal libraries, and 894 differentially expressed tags between metastatic and non-metastatic carcinomas. Three genes displaying differential regulation, one down-regulated (KRT31) and two up-regulated (BST2, MFAP2), as well as one with a non-significant differential expression pattern (GNA15) in our SAGE data were selected for real-time polymerase chain reaction (PCR) in a set of HNSCC samples. Consistent with our statistical analysis, quantitative PCR confirmed the upregulation of BST2 and MFAP2 and the downregulation of KRT31 when samples of HNSCC were compared to tumor-free surgical margins. As expected, GNA15 presented a non-significant differential expression pattern when tumor samples were compared to normal tissues. Conclusion: To the best of our knowledge, this is the first study reporting SAGE data in head and neck squamous cell tumors. Statistical analysis was effective in identifying differentially expressed genes reportedly involved in cancer development. The differential expression of a subset of genes was confirmed in additional larynx carcinoma samples and in carcinomas from a distinct head and neck subsite. Study protocol for the multicentre cohorts of Zika virus infection in pregnant women, infants, and acute clinical cases in Latin America and the Caribbean: The ZIKAlliance consortium
Background: The European Commission (EC) Horizon 2020 (H2020)-funded ZIKAlliance Consortium designed a multicentre study including pregnant women (PW), children (CH) and natural history (NH) cohorts. Clinical sites were selected over a wide geographic range within Latin America and the Caribbean, taking into account the dynamic course of the ZIKV epidemic. Methods: Recruitment to the PW cohort will take place in antenatal care clinics. PW will be enrolled regardless of symptoms and followed over the course of pregnancy, approximately every 4 weeks. PW will be revisited at delivery (or after miscarriage/abortion) to assess birth outcomes, including microcephaly and other congenital abnormalities according to the evolving definition of congenital Zika syndrome (CZS). After birth, children will be followed for 2 years in the CH cohort. Follow-up visits are scheduled at ages 1-3, 4-6, 12, and 24 months to assess neurocognitive and developmental milestones. In addition, a NH cohort for the characterization of symptomatic rash/fever illness was designed, including follow-up to capture persisting health problems. Blood, urine, and other biological materials will be collected, and tested for ZIKV and other relevant arboviral diseases (dengue, chikungunya, yellow fever) using RT-PCR or serological methods. InfluĂȘncia do grupamento muscular na recuperação da frequĂȘncia cardĂaca apĂłs o exercĂcio resistido
INTRODUĂĂO: O exercĂcio resistido (ER) Ă© um tipo de exercĂcio amplamente praticado, sendo recomendado para a manutenção ou aprimoramento da força e massa musculares e utilizado com fins estĂ©ticos e de saĂșde. Apesar disto, pouco se sabe sobre o impacto deste tipo de exercĂcio sobre o controle autonĂŽmico cardĂaco, tampouco da influĂȘncia do grupamento muscular nesta resposta. OBJETIVO: Verificar a influĂȘncia do grupamento muscular utilizado durante o ER, na recuperação da frequĂȘncia cardĂaca (REC-FC) pĂłs-exercĂcio. MĂTODOS: Participaram deste estudo 14 indivĂduos do sexo masculino (27,4 ± 6,1 anos; 79,4 ± 10,4 kg; 1,77 ± 0,1 m; 10,5 ± 4,6 %G) experientes na prĂĄtica de ER. O protocolo experimental constou da realização de teste e reteste de 1RM nos exercĂcios supino horizontal e meio agachamento para determinação da força dinĂąmica mĂĄxima e execução do nĂșmero mĂĄximo de repetiçÔes a 80% de 1RM com avaliação da REC-FC durante um minuto pĂłs-exercĂcio. RESULTADOS: Os resultados encontrados indicam menor REC-FC nos 10, 20, 30 e 40 segundos apĂłs o exercĂcio meio agachamento em comparação ao supino horizontal.
