    Near-infrared spectroscopy study of tourniquet-induced forearm ischaemia in patients with coronary artery disease

    Near-Infrared Spectroscopy (NIRS) can be employed to monitor local changes in haemodynamics and oxygenation of human tissues. A preliminary study has been performed in order to evaluate the NIRS transmittance response to induced forearm ischaemia in patients with coronary artery disease (CAD). The population consists in 40 patients with cardiovascular risk factors and angiographically documented CAD, compared to a group of 13 normal subjects. By inflating and subsequently deflating a cuff placed around the patient arm, an ischaemia has been induced and released, and the patients have been observed until recovery of the basal conditions. A custom NIRS spectrometer (IRIS) has been used to collect the backscattered light intensities from the patient forearm throughout the ischaemic and the recovery phase. The time dependence of the near-infrared transmittance on the control group is consistent with the available literature. On the contrary, the magnitude and dynamics of the NIRS signal on the CAD patients show deviations from the documented normal behavior, which can be tentatively attributed to abnormal vessel stiffness. These preliminary results, while validating the performance of the IRIS spectrometer, are strongly conducive towards the applicability of the NIRS technique to ischaemia analysis and to endothelial dysfunction characterization in CAD patients with cardiovascular risk factors

    Levosimendan for patients with severely reduced left ventricular systolic function and/or low cardiac output syndrome undergoing cardiac surgery. a systematic review and meta-analysis

    Background: Previous studies have shown beneficial effects of levosimendan in high-risk patients undergoing cardiac surgery. Two large randomized controlled trials (RCTs), however, showed no advantages of levosimendan. Methods: We performed a systematic review and meta-analysis (MEDLINE and Embase from inception until March 30, 2017), investigating whether levosimendan offers advantages compared with placebo in high-risk cardiac surgery patients, as defined by preoperative left ventricular ejection fraction (LVEF) = 35% and/or low cardiac output syndrome (LCOS). The primary outcomes were mortality at longest follow-up and need for postoperative renal replacement therapy (RRT). Secondary postoperative outcomes investigated included myocardial injury, supraventricular arrhythmias, development of LCOS, acute kidney injury (AKI), duration of mechanical ventilation, intensive care unit and hospital lengths of stay, and incidence of hypotension during drug infusion. Results: Six RCTs were included in the meta-analysis, five of which investigated only patients with LVEF = 35% and one of which included predominantly patients with LCOS. Mortality was similar overall (OR 0.64 [0.37, 1.11], p = 0.11) but lower in the subgroup with LVEF < 35% (OR 0.51 [0.32, 0.82], p = 0.005). Need for RRT was reduced by levosimendan both overall (OR 0.63 [0.42, 0.94], p = 0.02) and in patients with LVEF < 35% (OR 0.55 [0.31, 0.97], p = 0.04). Among secondary outcomes, we found lower postoperative LCOS in patients with LVEF < 35% receiving levosimendan (OR 0.49 [0.27, 0.89], p = 0.02), lower overall AKI (OR 0.62 [0.42, 0.92], p = 0.02), and a trend toward lower mechanical support, both overall (p = 0.07) and in patients with LVEF < 35% (p= 0.05). Conclusions: Levosimendan reduces mortality in patients with preoperative severely reduced LVEF but does not affect overall mortality.     Two-dimensional posture evaluation in Parkinson’s disease: effect of loads on the spinal angle during gait

    Parkinson’s Disease patients present diminished coordination caused by neural degeneration. This leads to large motor difficulties during gait such as balance loss and pronounced forward inclination of the upper body. This work assessed the spinal sagittal plane angle alterations in two groups: six parkinsonian patients and six control healthy subjects. This parameter was analyzed during gait under three conditions: without external loads and with external loads applied either on the chest or on the lower back area. Results were statistically compared by means of t-test of paired samples in both groups. For patients, a significant effect was found when loads were applied on the chest.     Prion protein and copper cooperatively protect neurons by modulating NMDA receptor through S-nitrosylation

    AIMS: Several neurodegenerative disorders show alterations in glutamatergic synapses and increased susceptibility to excitotoxicity. Mounting evidence suggests a central role for the cellular prion protein (PrP(C)) in neuroprotection. Therefore, the loss of PrP(C) function occurring in prion disorders may contribute to the disease progression and neurodegeneration. Indeed, PrP(C) modulates N-methyl-d-aspartate receptors (NMDAR), thus preventing cell death. In this study, we show that PrP(C) and copper cooperatively inhibit NMDAR through S-nitrosylation, a post-translational modification resulting from the chemical reaction of nitric oxide (NO) with cysteines. RESULTS: Comparing wild-type Prnp (Prnp(+/+)) and PrP(C) knockout (Prnp(0/0)) mouse hippocampi, we found that GluN1 and GluN2A S-nitrosylation decrease in Prnp(0/0). Using organotypic hippocampal cultures, we found that copper chelation decreases NMDAR S-nitrosylation in Prnp(+/+) but not in Prnp(0/0). This suggests that PrP(C) requires copper to support the chemical reaction between NO and thiols. We explored PrP(C)-Cu neuroprotective role by evaluating neuron susceptibility to excitotoxicity in Prnp(+/+) and Prnp(0/0) cultures. We found that (i) PrP(C)-Cu modulates GluN2A-containing NMDAR, those inhibited by S-nitrosylation; (ii) PrP(C) and copper are interdependent to protect neurons from insults; (iii) neuronal NO synthase inhibition affects susceptibility in wild-type but not in Prnp(0/0), while (iv) the addition of a NO donor enhances Prnp(0/0) neurons survival. INNOVATION AND CONCLUSIONS: Our results show that PrP(C) and copper support NMDAR S-nitrosylation and cooperatively exert neuroprotection. In addition to NMDAR, PrP(C) may also favor the S-nitrosylation of other proteins. Therefore, this mechanism may be investigated in the context of the different cellular processes in which PrP(C) is involved

    Biomimetic hydroxyapatite nanocrystals are an active carrier for Salmonella bacteriophages

    open access articlePurpose: The use of bacteriophages represents a valid alternative to conventional antimicrobial treatments, overcoming the widespread bacterial antibiotic resistance phenomenon. In this work, we evaluated whether biomimetic hydroxyapatite (HA) nanocrystals are able to enhance some properties of bacteriophages. The final goal of this study was to demonstrate that biomimetic HA nanocrystals can be used for bacteriophage delivery in the context of bacterial infections, and contribute – at the same time – to enhance some of the biological properties of the same bacteriophages such as stability, preservation, antimicrobial activity, and so on. Materials and methods: Phage isolation and characterization were carried out by using Mitomycin C and following double-layer agar technique. The biomimetic HA water suspension was synthesized in order to obtain nanocrystals with plate-like morphology and nanometric dimensions. The interaction of phages with the HA was investigated by dynamic light scattering and Zeta potential analyses. The cytotoxicity and intracellular killing activities of the phage–HA complex were evaluated in human hepatocellular carcinoma HepG2 cells. The bacterial inhibition capacity of the complex was assessed on chicken minced meat samples infected with Salmonella Rissen. Results: Our data highlighted that the biomimetic HA nanocrystal–bacteriophage complex was more stable and more effective than phages alone in all tested experimental conditions. Conclusion: Our results evidenced the important contribution of biomimetic HA nanocrystals: they act as an excellent carrier for bacteriophage delivery and enhance its biological characteristics.     Chronic kidney disease and urological disorders: systematic use of uroflowmetry in nephropathic patients

    Background. Chronic kidney disease (CKD) is a highly prevalent condition. Urologic disorders are known causes of CKD, but often remain undiagnosed and underestimated also for their insidious onset and slow progression. We aimed to evaluate the prevalence of urological unrecognized diseases in CKD patients by uroflowmetry. Methods. We enrolled consecutive stable CKD outpatients. The patients carried out two questionnaires, the International Prostate Symptom Score and Incontinence Questionnaire-Short Form, and they also underwent uroflowmetry, evaluating max flow rate (Qmax), voiding time and voided volume values. Results. A total of 83 patients (43 males, mean age of 59.8613.3 years) were enrolled. Our study showed 28 males and 10 females with a significant reduction of Qmax (P<0.001) while 21 females reported a significant increase of Qmax (P<0.001) with a prevalence of 49.5% of functional urological disease. Moreover, we showed a significant association between Qmax and creatinine (P¼0.013), estimated glomerular filtration rate (P¼0.029) and voiding volume (P¼0.05). We have not shown significant associations with age (P¼0.215), body mass index (P¼0.793), systolic blood pressure (P¼0.642) or diastolic blood pressure (P¼0.305). Moreover, Pearson’s chi-squared test showed a significant association between Qmax altered with CKD (v2 ¼1.885, P¼0.170) and recurrent infection (v2¼8.886, P¼0.012), while we have not shown an association with proteinuria (v2¼0.484, P¼0.785), diabetes (v2¼0.334, P¼0.563) or hypertension (v2¼1.885, P¼0.170).Conclusions. We showed an elevated prevalence of urological diseases in nephropathic patients; therefore, we suggest to include uroflowmetry in CKD patient assessment, considering the non-invasiveness, repeatability and low cost of examination. Uroflowmetry could be used to identify previously unrecognized urological diseases, which may prevent the onset of CKD or progression to end-stage renal disease and reduce the costs of management

    The Complex Systems and Biomedical Sciences group at the ESRF: current status and new opportunities after Extremely Brilliant Source upgrade

    The Complex System and Biomedical Sciences (CBS) group at the European Synchrotron Radiation Facility (ESRF) in Grenoble is dedicated to the study of a broad family of materials and systems, including soft and hard condensed matter, nanomaterials, and biological materials. The main experimental methods used for this purpose are X-ray diffraction, reflectivity, scattering, photon correlation spectroscopy, and time-resolved X-ray scattering/diffraction. After a recent and successful Extremely Brilliant Source (EBS) upgrade, the Grenoble synchrotron has become the first of the 4th generation high energy facilities, which offers unprecedented beam parameters for its user community, bringing new experimental opportunities for the exploration of the nanoscale structure, kinetics, and dynamics of a myriad of systems. In this contribution, we present the impact of the recent upgrade on the selected beamlines in the CBS group and a summary of recent scientific activities after the facility reopening

    Validación y detección automática del transporte dispersivo del emisario submarino de Mar del Plata, Argentina

    The submarine outfall of Mar del Plata city at Camet was projected considering the mean and maximum of forecasted sewage discharges, the inner-shelf depth, coliform concentration and its decay (T90) mainly induced by sunlight effect and costal salinity. In 2016 the outfall was operating with a length of 3,810 m and diffusers in the last 526 m. An economical method to monitor its performance in relation to the surroundings, is remote-sensing techniques, applying either visible or radar images. Tidal currents parallel to the coast are responsible for the transport of the sedimentary plume in the far field, after a primary dilution from a depth of 11 m. Visible images (1.5 to 6 m spatial resolution) are effective in monitoring the plume entrained in the upper portion of the water column. These analyses led to study the interaction between waves and coastal currents. Radar images (30 m resolution X and C bands) permit to survey the slick-alike plume that differs from the environment water by the surface roughness. Comparing both techniques visible images can distinguish the different colours of the plume; instead, the radar images are showing the surface roughness from the slick-alike plume. The main advantage of active sensors is that they can map the plume during a cloudy weather and even during night time.El emisario submarino de Mar del Plata en Camet fue proyectado considerando las descargas cloacales promedio y máximas previstas, la profundidad de la plataforma vecina, la concentración de coliformes y el decaimiento (T90) inducido por la luz solar y la salinidad. En 2016 el emisario operaba con una longitud de 3.810 m con difusores en los últimos 526 m. Un método poco oneroso para analizar su comportamiento en relación a su entorno es la aplicación de técnicas de teledetección tanto en el espectro visible como mediante imágenes de radar. Las corrientes de marea paralelas a la costa son responsables de una pluma sedimentaria en el campo lejano, luego de una dilución primaria desde una profundidad de 11 m. Las imágenes visibles (resolución espacial de 1,5 a 6 m) son efectivas para monitorear la pluma extendida en la capa superior del mar. Estos análisis permiten el estudio de la interacción entre olas y corrientes costeras. Las imágenes de radar (resolución de 30 m en las bandas X y C) permiten relevar plumas superficiales semejantes a derrames de aceites por su rugosidad. Comparando ambas técnicas las imágenes visibles pueden distinguir plumas de diferentes colores del agua; por el contrario, las imágenes de radar están mostrando diferencias en la tensión superficial. La principal ventaja de los sensores activos es que permiten monitorear la pluma durante tiempo nuboso incluso sin luz solar

    Focus on the Use of Resveratrol in Bladder Cancer

    Bladder cancer is the most common tumor of the urinary system, with a high incidence in the male population. Surgery and intravesical instillations can eradicate it, although recurrences are very common, with possible progression. For this reason, adjuvant therapy should be considered in all patients. Resveratrol displays a biphasic dose response both in vitro and in vivo (intravesical application) with an antiproliferative effect at high concentrations and antiangiogenic action in vivo (intraperitoneal application) at a low concentration, suggesting a potential role for it in clinical management as an adjuvant to conventional therapy. In this review, we examine the standard therapeutical approach to bladder cancer and the preclinical studies that have investigated resveratrol in xenotransplantation models of bladder cancer.     Recombinant peptide reverses cryo-capacitation in ram sperm and improves in vitro fertilization

    Semen cryopreservation is a very important technique for assisted reproduction; however, the cryopreservation process is harmful because it results in a reduction in sperm motility and viability, and leads to premature signals of capacitation, resulting in lesser than desirable fertility rates after artificial insemination. A fraction of seminal plasma, enriched in proteins that contain type II fibronectin domains (FNII) can reverse molecular indicators of cryo-capacitation. The beneficial effects of these proteins, however, depend on the relative abundance in seminal plasma. To create a safe additive for improving frozen sperm functionality, in the present study there was cloning and expression of a recombinant peptide containing four FNII domains (named TrxA-FNIIx4-His6) and evaluation of its effect after addition to frozen/thawed ram sperm. The cDNA for this protein was expressed in E. coli and after denaturation and re-naturalization of the protein, toxicity and binding capacity were assessed. By fluorescent labelling assessment, there was binding of the protein to the thawed sperm. At the two doses used (0.15 and 0.3 μM), TrxA-FNIIx4-His6 had the capacity to reverse the molecular indicators of cryo-capacitation as indicated by the reduction on phosphorylated substrates of PKA. Furthermore, the supplementation with this protein resulted in a normal capacitation process as evidenced by the increase in the in vitro fertilization rate when the greatest concentration of the protein was evaluated (73.25 ± 2.95; 40.13 ± 11.82 for 0.3 μM and control, respectively). There was no effect of protein supplementation on sperm objective motility compared to untreated sperm. In conclusion, the use of TrxA-FNIIx4-His6 is a promising biotechnological approach for cryopreserving ram sperm and maintaining sperm viability.Fil: Ledesma, Alba.
