    Readiness to change drinking behaviour among heavy-drinking university students in England

    There is growing literature on possible ways of reducing alcohol consumption and alcohol-related harm among university students (Larimer and Cronce, 2002; Siegers and Carey, 2010). However, interventions with this aim might be made more effective by information on students’ readiness to change their drinking behaviour (Carey et al., 2007a), where an assessment of readiness to change might influence the kind of approach that is thought most likely to be successful. For example, it has been found that readiness to change moderated the effects of a brief intervention among heavy-drinking students (either brief motivational intervention or alcohol expectancy challenge) such that high readiness to change made an expectancy challenge relatively more effective in reducing drinking (Capone and Wood, 2009). This study also reported an association between higher readiness to change and greater reductions in alcohol consumption in the overall sample, thus supporting previous findings (Fromme and Corbin, 2004; Carey et al., 2007b). Although high readiness to change may increase the chances of successful brief intervention among heavy-drinking students, it has been found that, even among individuals referred to a university-based alcohol intervention programme, there was limited acknowledgement of a drinking problem or interest in changing behaviour (Caldwell, 2002; Vik et al., 2000). Such research has been conducted mainly in the USA and, with the exception of one study (Hosier, 2001), it is unknown whether a comparable lack of concern about heavy drinking is true of students in England. Moreover, there is limited understanding of the different factors associated with, and predictive of, readiness to change in heavy-drinking students. The aims of this paper are therefore (i) to assess levels of readiness to change among heavy-drinking students at universities in England, (ii) to identify variables predictive of readiness to change among heavy-drinking students and (iii) to generate hypotheses that could be tested in further research

    Autistic traits and mental health in women with the fragile-X premutation: maternal status versus genetic risk

    BACKGROUND: Research on women with the fragile-X premutation (FX-p) has been underrepresented within the field of behavioural phenotypes. AIMS: To understand whether the FX-p confers risk for autistic traits, depression and anxiety, independent of maternal status. METHOD: In study 1, mothers of children with fragile-X syndrome (M-FXp; n = 51, mean age 43 years (s.d. = 5.80)) were compared with mothers of autistic children (M-ASD; n = 59, mean age 42 (s.d. = 5.80)), mothers of children with Smith-Magenis syndrome (M-SMS; n = 27, mean age 39 (s.d. = 7.20)) and mothers of typically developing children (M-TD; n = 44, mean age 40 (s.d. = 4.90)). In study 2, the M-FXp group were compared with non-mothers with the FX-p (NM-FXp; n = 17, mean age 32 (s.d. = 9.20)), typically developed non-mothers (NM-TD; n = 28, mean age 31 (s.d. = 6.80)) and the M-TD group. All participants completed an online survey, including measures of IQ, autistic traits, anxiety, depression and positive affect. RESULTS: In study 1: the M-FXp group reported more autistic traits than the M-TD group (P < 0.05, η2 = 0.046). Anxiety and parental stress were elevated in the M-FXp, M-SMS and M-ASD groups relative to the M-TD group (all P ≤ 0.003, η2 = 0.079-0.322). In study 2: a main effect of premutation status indicated that women with the FX-p report elevated autistic traits and anxiety (P ≤ 0.007, η2 = 0.055-0.060); this did not interact with maternal status. CONCLUSIONS: The findings indicate that women with the FX-p show an increased risk for autistic traits and anxiety. This risk is specific to the presence of the FX-p and is not fully accounted for by maternal status or the stress of caring for children with neurodevelopmental disorders

    Reactions to the National Living Wage in Hospitality

Purpose: The purpose of this paper is to explore reactions to the introduction by the UK Government of the National Living Wage (NLW) in the UK hospitality sector and consider implications for the status of employee relations. Design/methodology/approach: In-depth interviews were conducted with senior industry representatives of the hospitality sector in the UK. Findings: Concerns surrounding an increase in the wage bill, in maintaining pay differentials and in shifting employment to youth were confirmed. Managers expressed ambiguity in face of the legislation, offering agreement at a personal level with the rationale underpinning the NLW, but also expressing concern about impacts on their businesses. Research limitations/implications: This exploratory study offers the basis for further research in understanding the foundation of employee relations in hospitality. Social implications: A reconsideration of the nature of the employment relationship is key at a time of growing concerns about the business-society relationship. Originality/value: Uses reactions to the UK Government’s stipulation of a NLW to explore the basis of employee relations in the hospitality sector. This is timely where work to date in hospitality has largely focussed on symptoms but not causes of poor working conditions

    Development of a yeast model to study the contribution of vacuolar polyphosphate metabolism to lysine polyphosphorylation

    A recently discovered protein post-translational modification, lysine polyphosphorylation (K-PPn), consists of the covalent attachment of inorganic polyphosphate (polyP) to lysine residues. The non-enzymatic nature of K-PPn means that the degree of this modification depends on both polyP abundance and the amino acids surrounding the modified lysine. K-PPn was originally discovered in budding yeast (Saccharomyces cerevisiae), in which polyP anabolism and catabolism are well characterized. However, yeast vacuoles accumulate large amounts of polyP, and upon cell lysis, the release of the vacuolar polyP could non-physiologically cause K-PPn of nuclear and cytosolic targets. Moreover, yeast vacuoles possess two very active endopolyphosphatases, Ppn1 and Ppn2, that could have opposing effects on the extent of K-PPn. Here, we characterized the contribution of vacuolar polyP metabolism to K-PPn of two yeast proteins, Top1 (DNA topoisomerase 1) and Nsr1 (nuclear signal recognition 1). We discovered that whereas Top1-targeting K-PPn is only marginally affected by vacuolar polyP metabolism, Nsr1-targeting K-PPn is highly sensitive to the release of polyP and of endopolyphosphatases from the vacuole. Therefore, to better study K-PPn of cytosolic and nuclear targets, we constructed a yeast strain devoid of vacuolar polyP by targeting the exopolyphosphatase Ppx1 to the vacuole and concomitantly depleting the two endopolyphosphatases (ppn1Δppn2Δ, vt-Ppx1). This strain enabled us to study K-PPn of cytosolic and nuclear targets without the interfering effects of cell lysis on vacuole polyP and of endopolyphosphatases.     Electrochemical measurement of antibody­‐antigen recognition biophysics: thermodynamics and kinetics of human chorionic gonadotropin (hCG) binding to redox-­tagged antibodies

    The thermodynamics and kinetics of antigen binding under diffusive conditions to an electrode surface modified with ferrocene-tagged antibodies is studied within this work, and realised experimentally for the case of human chorionic gonadotropin (hGC) as the antigen with monoclonal anti-hCG antibodies immobilised on an electrode surface via a molecular wire platform. The formation of the antigen-antibody complex is monitored through the blocking of the ferrocene voltammetry, thereby enabling the fractional coverage of the electrode binding sites to be unravelled as a function of time. It is found that, at low antigen concentrations, a Frumkin adsorption isotherm fits the data, with repulsive interactions between bound antigens playing a significant rôle, with an affinity constant that is an order of magnitude larger than in the case of an untagged antibody, suggesting that the chemical hydrophobicity of the redox tag may encourage stronger binding. Comparison of the experimental temporal data with relevant diffusion-adsorption models under activation control allows for the extraction of the kinetic parameters; at zero coverage, the rate constants for adsorption and desorption are, respectively, larger and smaller than the untagged antibody. The kinetic study enables the confirmation that this type of platform may be utilised for rapid (15 min) and quantitative electroimmunoassay. This is validated through proof-of-concept analytical measurements, yielding a limit of detection around 25 mIU mL−1 (corresponding to 2.7 ng mL−1) – a value used clinically for urine hCG measurements corresponding to around four weeks of gestational age

    Signal and System Approximation from General Measurements

    In this paper we analyze the behavior of system approximation processes for stable linear time-invariant (LTI) systems and signals in the Paley-Wiener space PW_\pi^1. We consider approximation processes, where the input signal is not directly used to generate the system output, but instead a sequence of numbers is used that is generated from the input signal by measurement functionals. We consider classical sampling which corresponds to a pointwise evaluation of the signal, as well as several more general measurement functionals. We show that a stable system approximation is not possible for pointwise sampling, because there exist signals and systems such that the approximation process diverges. This remains true even with oversampling. However, if more general measurement functionals are considered, a stable approximation is possible if oversampling is used.
