    Telomerase is an enzyme that adds repetitive DNA sequences to the ends of chromosomes and consists of two main subunits: the telomerase reverse transcriptase (TERT) protein and an associated telomerase RNA (TER). The telomerase essential N-terminal (TEN) domain is a conserved region of TERT proposed to mediate DNA substrate interactions. Here, we have employed single molecule telomerase binding assays to investigate the function of the TEN domain. Our results reveal telomeric DNA substrates bound to telomerase exhibit a dynamic equilibrium between two states: a docked conformation and an alternative conformation. The relative stabilities of the docked and alternative states correlate with the number of basepairs that can be formed between the DNA substrate and the RNA template, with more basepairing favoring the docked state. The docked state is further buttressed by the TEN domain and mutations within the TEN domain substantially alter the DNA substrate structural equilibrium. We propose a model in which the TEN domain stabilizes short RNA-DNA duplexes in the active site of the enzyme, promoting the docked state to augment telomerase processivity

    The allocation of attention, including the initial orienting and the subsequent dwell time, is affected by several bottom-up and top-down factors. How item memory affects these processes, however, remains unclear. Here, we investigated whether item memory affects attentional dwell time by using a modified version of the attentional blink (AB) paradigm. Across four experiments, our results revealed that the AB was significantly affected by memory status (novel vs. old), but critically, this effect depended on the ongoing memory context. Specifically, items that were unique in terms of memory status demanded more resources, as measured by a protracted AB. The present findings suggest that a more comprehensive understanding of memory’s effects on attention can be obtained by accounting for an item’s memorial context, as well as its individual item memory strength. Our results provide new evidence that item memory and memory context play a significant role in the temporal allocation of attention

We have reported here a novel method that can smooth these features whilst avoiding edge down-turn. This process can be scaled up to E-ELT segment fabrication time-scale. It has been experimentally demonstrated that the surface quality is good enough for subsequent Zeeko form correction technology to achieve form specifications

    Automated Inspection Device for Explosive Charge in Shells - AIDECS

    Certain defects in the explosive charge of an artillery shell can cause the projectile to explode prematurely in the barrel of the launcher from which it is fired. The sensitivity of the radiographic technique presently used is limited by the large influence of the steel shell casing on the transmitted radiation. A filmless radiometric technique utilizing the basic radiation principle of Compton scattering, which will detect cavities in the explosive filler with minimal interference from the steel casing, has been identified and tested. By scanning the shell with a beam of radiation and observing the Compton scattering through a unique collimating system, it has been possible to detect voids as small as 1/16 inch in cross section. The hardware consists of the source, beam collimator, detector collimator, and a large plastic scintillator detector system. The projectile is inserted into the beam path and moved through a fixed scanning pattern by a mechanical handling system. The scanning sequence is computer contra ll ed and results in a threedimensional data matrix giving a direct representation of density within the projectile. Voids are identified and classified by computer analysis, and shell acceptability decisions are automatically generated. An engineering prototype system is currently being assembled and tested. (A production prototype conceptual design is concurrently under development.) This new technique will replace an existing film radiography inspection procedure and eliminate the need for human interpretation of the defects, while providing more consistent and reliable inspections at lower costs

    Using Innovation-Corps (I-Corps™) methods to adapt a mobile health (mHealth) obesity treatment for community mental health settings

    BACKGROUND: We employed Innovation Corps (I-Corps™) methods to adaptation of a mobile health (mHealth) short-message-system (SMS) -based interactive obesity treatment approach (iOTA) for adults with severe mentall illness receiving care in community settings. METHODS: We hypothesized jobs to be done in three broad stakeholder groups: decision makers (DM = state and community clinic administrators), clinician consumers (CC = case managers, peer supports, nurses, prescribers) and service consumers (SC = patients, peers and family members). Semistructured interviews ( RESULTS: Four themes emerged across groups: education, inertia, resources and ownership. Sub-themes in education and ownership differed between DM and CC groups on implementation ownership, intersecting with professional development, suggesting the importance of training and supervision in scalability. Sub-themes in resources and intertia differed between CC and SC groups, suggesting illness severity and access to healthy food as major barriers to engagement, whereas the SC group identified the need for enhanced emotional support, in addition to pragmatic skills like menu planning and cooking, to promote health behavior change. Although SMS was percieved as a viable education and support tool, CC and DM groups had limited familiarity with use in clinical care delivery. CONCLUSIONS: Based on customer discovery, the characteristics of a minimum viable iOTA for implementation, scalability and sustainability include population- and context-specific adaptations to treatment content, interventionist training and delivery mechanism. Successful implementation of an SMS-based intervention will likely require micro-adaptations to fit specific clinical settings
