    303-306The larvicidal potential of two marine organisms was investigated by testing their non-polar to polar organic extracts against mosquito Culex pipens fatigans. In the present investigation the non-polar petroleum ether extract of prawn Nematopalaemon. tenuipes, Hendersen and polar methanol extract of sea cucumber Holothuria scabra, Jaeger body wall were found to be effective against mosquito larvae. The preliminary chemical analysis showed the presence of steroids in the active extract of prawns, whereas the presence of saponins in the active extract of sea cucumber body wall. In this context, the observed mosquito larvicidal activity could be attributed to the presence of steroids and saponins. In summary, this investigation explores the importance of marine organisms as a valuable resource for the discovery of novel insecticidal molecules

    Effect of Bombay high crude oil and its water-soluble fraction on growth and metabolism of diatom Thalassiosira sp.

    251-255Effect of Bombay high crude oil (BHC) and its water-soluble fraction (WSF) on growth and metabolism of the phytoplankton, Thalassiosira sp. was assessed. The study revealed the signs of acute toxicity at higher concentrations of crude oil (0.5%) and WSF (40%), while stimulatory effect was observed at lower concentrations (0.01 and 0.1% of BHC and 5, 10% of WSF). WSF at higher concentrations (20 and 40%) caused reduction in DNA and RNA of the diatom. At lower concentrations it caused increase in protein and RNA content indicating increased metabolism. High concentrations of oil and its fraction had inhibitory effect on growth, protein content and nucleic acid content. This indicates that biosynthesis of these molecules may be probable targets for toxicity of oil

Bioresource Technology. 2024: 130513.Demonstrating outdoor cultivation of engineered microalgae at considerable scales is essential for their prospective large-scale deployment. Hence, this study focuses on the outdoor cultivation of an engineered Chlamydomonas reinhardtii strain, 3XAgBs-SQs, for bisabolene production under natural dynamic conditions of light and temperature. Our preliminary outdoor experiments showed improved growth, but frequent culture collapses in conventional Tris-acetate-phosphate medium. In contrast, modified high-salt medium (HSM) supported prolonged cell survival, outdoor. However, their subsequent outdoor scale-up from 250 mL to 5 L in HSM was effective with 10 g/L bicarbonate supplementation. Pulse amplitude modulation fluorometry and metabolomic analysis further validated their improved photosynthesis and uncompromised metabolic fluxes towards the biomass and the products (natural carotenoids and engineered bisabolene). These strains could produce 906 mg/L bisabolene and 54 mg/L carotenoids, demonstrating the first successful outdoor photoautotrophic cultivation of engineered C. reinhardtii, establishing it as a one-cell two-wells biorefinery

    Clinical Characterization and Genomic Analysis of Samples from COVID-19 Breakthrough Infections during the Second Wave among the Various States of India

    From March to June 2021, India experienced a deadly second wave of COVID-19, with an increased number of post-vaccination breakthrough infections reported across the country. To understand the possible reason for these breakthroughs, we collected 677 clinical samples (throat swab/nasal swabs) of individuals from 17 states/Union Territories of the country who had received two doses (n = 592) and one dose (n = 85) of vaccines and tested positive for COVID-19. These cases were telephonically interviewed and clinical data were analyzed. A total of 511 SARS-CoV-2 genomes were recovered with genome coverage of higher than 98% from both groups. Analysis of both groups determined that 86.69% (n = 443) of them belonged to the Delta variant, along with Alpha, Kappa, Delta AY.1, and Delta AY.2. The Delta variant clustered into four distinct sub-lineages. Sub-lineage I had mutations in ORF1ab A1306S, P2046L, P2287S, V2930L, T3255I, T3446A, G5063S, P5401L, and A6319V, and in N G215C; Sub-lineage II had mutations in ORF1ab P309L, A3209V, V3718A, G5063S, P5401L, and ORF7a L116F; Sub-lineage III had mutations in ORF1ab A3209V, V3718A, T3750I, G5063S, and P5401L and in spike A222V; Sub-lineage IV had mutations in ORF1ab P309L, D2980N, and F3138S and spike K77T. This study indicates that majority of the breakthrough COVID-19 clinical cases were infected with the Delta variant, and only 9.8% cases required hospitalization, while fatality was observed in only 0.4% cases. This clearly suggests that the vaccination does provide reduction in hospital admission and mortality
