    Expression of The αβ T-Cell Receptor Is Necessary for The Generation of The Thymic Medulla

    The architecture of the thymus of mice that congenitally fail to express the αβ T-cell receptor (TCRαβ) has been examined by immunohistology. In these mice, a defined mutation was introduced into the TCRc gene by homologous recombination. By using antibodies specific for cortical or medullary epithelium and for major histocompatibility complex antigens, the network of cortical epithelium in these mice was shown to be essentially unaltered in comparison with that of normal mice. In contrast, the thymic medulla was considerably reduced in size. This analysis shows that expression of the αβ TCR but not the γδ TCR is obligatory for establishing the thymic medulla and suggests that the growth of medullary epithelial cells may require contact with TCRαβ-expressing cells

    Storms modify baroclinic energy fluxes in a seasonally stratified shelf sea: inertial-tidal interaction

    Observations made near the Celtic Sea shelf edge are used to investigate the interaction between wind-generated near-inertial oscillations and the semidiurnal internal tide. Linear, baroclinic energy fluxes within the near-inertial (f) and semidiurnal (M2) wave bands are calculated from measurements of velocity and density structure at two moorings located 40 km from the internal tidal generation zone. Over the 2 week deployment period, the semidiurnal tide drove 28–48 W m−1 of energy directly on-shelf. Little spring-neap variability could be detected. Horizontal near-inertial energy fluxes were an order of magnitude weaker, but nonlinear interaction between the vertical shear of inertial oscillations and the vertical velocity associated with the semidiurnal internal tide led to a 25–43% increase in positive on-shelf energy flux. The phase relationship between f and M2 determines whether this nonlinear interaction enhances or dampens the linear tidal component of the flux, and introduces a 2 day counter-clockwise beating to the energy transport.     Baroclinic energy flux at the continental shelf edge modified by wind-mixing

    Temperature and current measurements from two moorings onshore of the Celtic Sea shelf break, a well-known hot spot for tidal energy conversion, show the impact of passing summer storms on the baroclinic wavefield. Wind-driven vertical mixing changed stratification to permit an increased on-shelf energy transport, and baroclinic energy in the semidiurnal band appeared at the moorings 1–4 days after the storm mixed the upper 50 m of the water column. The timing of the maximum in the baroclinic energy flux is consistent with the propagation of the semidiurnal internal tide from generation sites at the shelf break to the moorings 40 km away. Also, the ∼3 day duration of the peak in M2 baroclinic energy flux at the moorings corresponds to the restratification time scale following the first storm

    Expression of p16 Within Myenteric Neurons of the Aged Colon: A Potential Marker of Declining Function.

    Human colonic neuromuscular functions decline among the elderly. The aim was to explore the involvement of senescence. A preliminary PCR study looked for age-dependent differences in expression of CDKN1A (encoding the senescence-related p21 protein) and CDKN2A (encoding p16 and p14) in human ascending and descending colon (without mucosa) from 39 (approximately 50: 50 male: female) adult (aged 27-60 years) and elderly donors (70-89 years). Other genes from different aging pathways (e.g., inflammation, oxidative stress, autophagy) and cell-types (e.g., neurons, neuron axonal transport) were also examined. Unlike CDKN1A, CDKN2A (using primers for p16 and p14 but not when using p14-specific primers) was upregulated in both regions of colon. Compared with the number of genes appearing to upregulate in association with temporal age, more genes positively associated with increased CDKN2A expression (respectively, 16 and five of 44 genes studied for ascending and descending colon). Confirmation of increased expression of CDKN2A was sought by immunostaining for p16 in the myenteric plexus of colon from 52 patients, using a semi-automated software protocol. The results showed increased staining not within the glial cells (S100 stained), but in the cytoplasm of myenteric nerve cell bodies (MAP2 stained, with identified nucleus) of ascending, but not descending colon of the elderly, and not in the cell nucleus of either region or age group (5,710 neurons analyzed: n = 12-14 for each group). It was concluded that increased p16 staining within the cytoplasm of myenteric nerve cell bodies of elderly ascending (but not descending) colon, suggests a region-dependent, post-mitotic cellular senescence-like activity, perhaps involved with aging of enteric neurons within the colon

    Control of a phytoplankton bloom by wind‐driven vertical mixing and light availability

    The balance of physical and biological processes governing phytoplankton growth rates and the accumulation of biomass is widely debated in the literature, notably during the winter–spring transition. Here we show, in a temperate shelf sea that variability in the depth of the actively mixing surface layer is the leading order control. During a 2-week period preceding the peak of the spring bloom we observe two distinct regimes; first, growth within the euphotic zone during the day and re-distribution of new biomass to the seasonal pycnocline at night by convective mixing; then, more rapid biomass accumulation trapped within a shallower, wind-driven actively mixing layer that was decoupled from the pycnocline below. Our observations of the bloom in the Celtic Sea, Northwest European Shelf, were made using ocean gliders and include measurements of the dissipation of turbulent kinetic energy. A 1-D phytoplankton growth model driven by our measurements of dissipation and incident irradiance replicates the observed bloom and reinforces the conclusion that physical processes that mediate light availability were key. Day-to-day variability in cloud cover and the ability of phytoplankton to acclimate to their light environment were also important factors in determining growth rates, and the timing of the biomass peak. Our results emphasize the need for accurate turbulent mixing parameterizations in coupled hydrodynamic-ecosystem models. Our findings are applicable to any region where wind-driven mixing can modify nutrient and light availability, especially across subpolar shelves in the northern hemisphere where light rather than nutrients is typically the limiting factor on phytoplankton growth

    Feasibility study of the National Autistic Society EarlyBird parent support programme

    The EarlyBird programme is a group-based psychoeducation intervention for parents of young children with autism. Although it is widely used in the United Kingdom, the evidence base for the programme is very limited. Using a mixed method, non-randomised research design, we aimed to test (1) the acceptability of the research procedures (recruitment, retention, suitability of measures), (2) the parental acceptability of EarlyBird (attendance, views of the programme, perceived changes) and (3) the facilitator acceptability of EarlyBird (fidelity, views of the programme, perceived changes). Seventeen families with a 2- to 5-year-old autistic child and 10 EarlyBird facilitators took part. Pre- and post-intervention assessment included measures of the child’s autism characteristics, cognitive ability, adaptive behaviour, emotional and behavioural problems and parent-reported autism knowledge, parenting competence, stress and wellbeing. Semi-structured interviews were completed at post-intervention with parents and facilitators. For those involved in the study, the research procedures were generally acceptable, retention rates were high and the research protocol was administered as planned. Generally, positive views of the intervention were expressed by parents and facilitators. Although the uncontrolled, within-participant design does not allow us to test for efficacy, change in several outcome measures from pre- to post-intervention was in the expected direction. Difficulties were encountered with recruitment (opt-in to the groups was ~56% and opt-in to the research was 63%), and strategies to enhance recruitment need to be built into any future trial. These findings should be used to inform protocols for pragmatic, controlled trials of EarlyBird and other group-based interventions for parents with young autistic children

    Child and parent outcomes following parent interventions for child emotional and behavioral problems in autism spectrum disorders: A systematic review and meta-analysis

    There is growing interest in the development of behavioral parent interventions targeting emotional and behavioral problems in children with autism spectrum disorders. Such interventions have potential to improve a number of child and parental well-being outcomes beyond disruptive child behavior. This systematic review and meta-analysis assesses evidence for the efficacy of behavioral parent interventions for disruptive and hyperactive child behavior in autism spectrum disorders, as well as parenting efficacy and stress. A total of 11 articles from nine randomized controlled trials were included. Sufficient data were available to calculate standardized mean difference and show favorable effects of behavioral parent interventions on parent-reported measures of child disruptive behavior (standardized mean difference = 0.67), hyperactivity (standardized mean difference = 0.31) and parent stress (standardized mean difference = 0.37); effects on parent efficacy are less clear (standardized mean difference = 0.39, p = 0.17). There were insufficient data to explore intervention effects on internalizing behavior in autism spectrum disorders, parenting behaviors, or observational and teacher-reported outcomes, providing important avenues for future research. This review adds to growing evidence of the efficacy of behavioral parent interventions for child behavior and parental well-being in autism spectrum disorders (Prospero: CRD42016033979)
