    Role of solvent for globular proteins in solution

    The properties of the solvent affect the behavior of the solution. We propose a model that accounts for the contribution of the solvent free energy to the free energy of globular proteins in solution. For the case of an attractive square well potential, we obtain an exact mapping of the phase diagram of this model without solvent to the model that includes the solute-solvent contribution. In particular we find for appropriate choices of parameters upper critical points, lower critical points and even closed loops with both upper and lower critical points, similar to one found before [Macromolecules, 36, 5845 (2003)]. In the general case of systems whose interactions are not attractive square wells, this mapping procedure can be a first approximation to understand the phase diagram in the presence of solvent.     Water loss in horticultural products. Modelling, data analysis and theoretical considerations

    The water loss of individual fruit (melon, plum and mandarin) was analysed using the traditional diffusion based approach and a kinetic approach. Applying simple non linear regression, both approaches are the same, resulting in a quite acceptable analysis. However, by applying mixed effects non linear regression analysis, explicitly including the variation over the individuals, the kinetic approach was found to reflect the processes occurring during mass loss better than the diffusion approach. All the variation between the individuals in a batch could be attributed to the initial mass or size of the individuals. The fraction of the fruit mass that is available for transpiration is the key item in the water loss process, rather than the skin resistance and fruit area.     Optimization and transformation of Arundo donax L. using particle bombardment

    An optimized particle bombardment protocol to introduce DNA into Arundo donax L. (giant reed) embryogenic callus cells was developed. The physical and biological parameters tested for optimal transient expression of ß-glucuronidase (GUS) and green fluorescent protein (GFP) genes were: helium pressure, distance from stopping screen to target tissue and vacuum pressure together with other factors such as gold microparticle size, DNA concentration and the number of bombardments. The highest transient GUS and GFP expression was obtained when cells were bombarded twice at 1100 psi, with 9 cm target distance, 24 mm Hg vacuum pressure, 1 mm gold particle size, 1.5 mg DNA per bombardment, three days pre-culture prior to bombardment and six days post bombardment culture. This is the first report of optimization of particle bombardment parameters for high-efficiency DNA delivery combined with minimum damage to target giant reed tissues.Key words: Arundo donax, particle bombardment, ß-glucuronidase (GUS), green fluorescent protein (GFP), transient gene expression, genetic transformation

    PULMONARY DIROFILARIASIS IN MAN - A NEW ITALIAN CASE, REVIEW OF THE EUROPEAN LITERATURE

    In June 1991 a 62-year-old retired mon, from Udine (northern Italy), was suddenly affected by dyspnoea. X-ray and CT control detected a coin lesion in the lung. In May 1992 this lesion was removed surgically. Histological examination revealed the presence of a nematode inside an arteriole which had provoked a small infarct in the pulmonary tissue. The parasite presented marked regressive phenomena that made on accurate morphological analysis impossible. However, in the light of certain details oi the cuticle, and by analogy with lour similar cases occurring in northern Italy, as well as 10 others (nine subcutaneous and one sub-mucosal) reported in man from the same region (Venetia) over the last 15 years, the aetiologic agent was thought io be Dirofilaria (N.) repens. Over all 10 cases of human pulmonary dirofilariasis were reported in Europe : five in Italy, probably by D.(N.) repens, two in Germany, in patients coming from Corsica and two in Spain (only by serology), attributed io D. immitis. In addition one case was reported in U.S.A. in a man who previously visited Italy

    Venice risks to be submerged as a consequence of two problems: local land subsidence and sea level rise due to global warming. They both contribute to what is referred as Apparent Sea Level Rise (ASLR). Flooding Tides (locally: Acqua Alta) submerge Venice with an exponentially increasing frequency. The Acqua Alta is generated by a number of factors, the main of them being the Sirocco wind blowing over the Adriatic Sea, that ultimately displaces waters towards Venice. These extreme events have been investigated by using the documentary description of past floods, accurately reported over the last millennium, and tide gauge records for the recent period. A fundamental problem is to know the trend of the ASLR, possibly distinguishing between land subsidence and sea level components. Instrumental data go back to 1872 and a key point is to extend ourkno wledge back in time. Long-term ASLR has been investigated with the help of a biological indicator, i.e. the height of the green belt of the algae that live in the tidal range and whose upper front shows the average high tide level. Fortunately, in the first half of the 18th century, this indicator was accurately drawn by the famous painter Antonio Canaletto (1697-1768) and his pupils, mainly Bernardo Bellotto (1722-1780), in their photographic paintings made with an optical camera obscura. It has been possible to compare the tidal level, as it was in the 1700s and today. After careful spot investigation and minor corrections for some changes to the hydrological system occurred in the meantime, the bulk submersion of Venice estimated from the paintings is 61 ± 11 cm with average yearly trend 1.9 mm y−1

    Mechanism of Bacterial Inactivation by (+)-Limonene and Its Potential Use in Food Preservation Combined Processes

    This work explores the bactericidal effect of (+)-limonene, the major constituent of citrus fruits' essential oils, against E. coli. The degree of E. coli BJ4 inactivation achieved by (+)-limonene was influenced by the pH of the treatment medium, being more bactericidal at pH 4.0 than at pH 7.0. Deletion of rpoS and exposure to a sub-lethal heat or an acid shock did not modify E. coli BJ4 resistance to (+)-limonene. However, exposure to a sub-lethal cold shock decreased its resistance to (+)-limonene. Although no sub-lethal injury was detected in the cell envelopes after exposure to (+)-limonene by the selective-plating technique, the uptake of propidium iodide by inactivated E. coli BJ4 cells pointed out these structures as important targets in the mechanism of action. Attenuated Total Reflectance Infrared Microspectroscopy (ATR-IRMS) allowed identification of altered E. coli BJ4 structures after (+)-limonene treatments as a function of the treatment pH: ß-sheet proteins at pH 4.0 and phosphodiester bonds at pH 7.0. The increased sensitivity to (+)-limonene observed at pH 4.0 in an E. coli MC4100 lptD4213 mutant with an increased outer membrane permeability along with the identification of altered ß-sheet proteins by ATR-IRMS indicated the importance of this structure in the mechanism of action of (+)-limonene. The study of mechanism of inactivation by (+)-limonene led to the design of a synergistic combined process with heat for the inactivation of the pathogen E. coli O157:H7 in fruit juices. These results show the potential of (+)-limonene in food preservation, either acting alone or in combination with lethal heat treatments

    Focusing of timelike worldsheets in a theory of strings

    An analysis of the generalised Raychaudhuri equations for string world sheets is shown to lead to the notion of focusing of timelike worldsheets in the classical Nambu-Goto theory of strings. The conditions under which such effects can occur are obtained . Explicit solutions as well as the Cauchy initial value problem are discussed. The results closely resemble their counterparts in the theory of point particles which were obtained in the context of the analysis of spacetime singularities in General Relativity many years ago.Comment: 14 pages, RevTex, no figures, extended, to appear in Phys Rev
