    Smart Environments design: the SPLASH project case. Journal of e-Learning and Knowledge Society

    This paper describes the process of collaborative design of a Smart Environment developed for the school context. The Living Lab project entitled “SPLASH” (“Smart platform for learning and active social habitat”) will be presented in the following pages. We will describe the whole process of the platform design: from the implementation of the layout and the functionalities of the digital environment, up to the first steps of prototype use. A research team composed mainly by an ICT company, teachers and students of two secondary school, and academic researchers, has been involved in a complex process of collaborative design, in order to create the prototype of a smart, social, innovative learning environment. Finally, assessment tools provided within the platform will be described. They have been co-designed with teachers and students of the schools involved in the project

    Toxic Tau Oligomers Modulated by Novel Curcumin Derivatives

    The pathological aggregation and accumulation of tau, a microtubule-associated protein, is a common feature amongst more than 18 different neurodegenerative diseases that are collectively known as tauopathies. Recently, it has been demonstrated that the soluble and hydrophobic tau oligomers are highly toxic in vitro due to their capacity towards seeding tau misfolding, thereby propagating the tau pathology seen across different neurodegenerative diseases. Modulating the aggregation state of tau oligomers through the use of small molecules could be a useful therapeutic strategy to target their toxicity, regardless of other factors involved in their formation. In this study, we screened and tested a small library of newly synthesized curcumin derivatives against preformed recombinant tau oligomers. Our results show that the curcumin derivatives affect and modulate the tau oligomer aggregation pathways, converting to a more aggregated non-toxic state as assessed in the human neuroblastoma SH-SY5Y cell line and primary cortical neuron cultures. These results provide insight into tau aggregation and may become a basis for the discovery of new therapeutic agents, as well as advance the diagnostic field for the detection of toxic tau oligomers

    On the complexity of the boundary layer structure and aerosol vertical distribution in the coastal Mediterranean regions: A case study

    The planetary boundary layer structure in the coastal areas, and particularly in complex orography regions such as the Mediterranean, is extremely intricate. In this study, we show the evolution of the planetary boundary layer based on in situ airborne measurements and ground-based remote sensing observations carried out during the MORE (Marine Ozone and Radiation Experiment) campaign in June 2010. The campaign was held in a rural coastal Mediterranean region in Southern Italy. The study focuses on the observations made on 17 June. Vertical profiles of meteorological parameters and aerosol size distribution were measured during two flights: in the morning and in the afternoon. Airborne observations were combined with ground-based LIDAR, SODAR, microwave and visible radiometer measurements, allowing a detailed description of the atmospheric vertical structure. The analysis was complemented with data from a regional atmospheric model run with horizontal resolutions of 12, 4 and 1 km, respectively; back-trajectories were calculated at these spatial resolutions. The observations show the simultaneous occurrence of dust transport, descent of mid-tropospheric air and sea breeze circulation on 17 June. Local pollution effects on the aerosol distribution, and a possible event of new particles formation were also observed. A large variability in the thermodynamical structure and aerosol distribution in the flight region, extending by approximately 30km along the coast, was found. Within this complex, environment-relevant differences in the back-trajectories calculated at different spatial resolutions are found, suggesting that the description of several dynamical processes, and in particular the sea breeze circulation, requires high-resolution meteorological analyses. The study also shows that the integration of different observational techniques is needed to describe these complex conditions; in particular, the availability of flights and their timing with respect to the occurring phenomena are crucial

    Natives’ and immigrants’ fertility intentions in Europe: the role of employment

    In several European countries, births to immigrant women represent more than 10% of all births; however, there is evidence showing that in the aftermath of the Great Recession migrants’ fertility might have fallen more pronouncedly among migrants than natives. With this study, we aim to provide further evidence about the employment/fertility link among migrants by focusing on the relationship between employment and fertility intentions - instead of the behaviours. In fact, most research on migrant fertility has focused on childbearing behaviours, disregarding the ideational dimensions represented by attitudes and intentions, which are of primary importance to complete the picture through a better understanding of the normative side of fertility. We do this relying on data from the European Social Survey and focusing on two years, 2004 and 2010, which allows us focussing on whether and how the financial crisis started in 2008 (the so-called Great Recession) may have modified the link between employment and fertility intentions.     Is the Combination of the Nationalities of the Couple Affecting Women’s Fertility Intentions? Insights from Italy

    Recently, there has been a rising interest on migrants’ fertility intentions (FIs). However, to the best of our knowledge, none of these studies have investigated the role played by the combination of the nationalities of the couple on women’s short-term intentions of having a(nother) child. This article is aimed at contributing to the existing literature on the subject by filling this gap, testing whether and how women’s FIs differ according to the type of couple (native, mixed and migrant couples) while disentangling the influence of the selection and socialization or adaptation hypotheses. Data drawn from two ISTAT surveys were harmonized and merged for multivariate analyses. Results shown that both individual and couples’ characteristics matter in shaping differences in FIs between foreign women partnered to Italian men and foreign women partnered to foreign men. Simultaneously, the FIs gap between foreign women partnered to Italian men and Italian women partnered to Italian men also decreased, giving support to the adaptation theory. Finally, findings point out to the presence of gradients in the explanatory power of the adaptation hypothesis to differences in FIs across groups of migrant women according to their area of nationality

    Specific tagging of the egress-related osmiophilic bodies in the gametocytes of Plasmodium falciparum

    <p>Abstract</p> <p>Background</p> <p>Gametocytes, the blood stages responsible for <it>Plasmodium falciparum </it>transmission, contain electron dense organelles, traditionally named osmiophilic bodies, that are believed to be involved in gamete egress from the host cell. In order to provide novel tools in the cellular and molecular studies of osmiophilic body biology, a <it>P. falciparum </it>transgenic line in which these organelles are specifically marked by a reporter protein was produced and characterized.</p> <p>Methodology</p> <p>A <it>P. falciparum </it>transgenic line expressing an 80-residue N-terminal fragment of the osmiophilic body protein Pfg377 fused to the reporter protein DsRed, under the control of <it>pfg377 </it>upstream and downstream regulatory regions, was produced.</p> <p>Results</p> <p>The transgenic fusion protein is expressed at the appropriate time and stage of sexual differentiation and is trafficked to osmiophilic bodies as the endogenous Pfg377 protein. These results indicate that a relatively small N-terminal portion of Pfg377 is sufficient to target the DsRed reporter to the gametocyte osmiophilic bodies.</p> <p>Conclusions</p> <p>This is the first identification of a <it>P. falciparum </it>aminoacid sequence able to mediate trafficking to such organelles. To fluorescently tag such poorly characterized organelles opens novel avenues in cellular and imaging studies on their biogenesis and on their role in gamete egress.</p

    Evaluation of esophageal motility using multichannel intraluminal impedance in healthy children and children with gastroesophageal reflux: comments

    Abstract OBJECTIVE: : Multichannel intraluminal impedance (MII) directly evaluates esophageal bolus transport. There is a good correlation between MII and manometry in healthy adults, but there are no reports concerning children.The aim of the present study was to determine normal values of esophageal motility using only impedance measurements in healthy children and in a pediatric population with gastroesophageal reflux (GER). PATIENTS AND METHODS: : We described in the present study 60 children submitted to pH-MII for 24 hours for suspected GER. Patients were divided into 2 different groups on the basis of their pH-MII report. Group 1 patients showed acid GER, whereas group 2 patients had negative pH-MII analysis for GER despite symptoms. We described impedance reflux and motility parameters on 10 standardized swallows: number of reflux, mean acid clearing time, median bolus clearing time, bolus presence time, total bolus transit time, segmental transit time, and total propagation velocity. RESULTS: : In group 1, the median mean acid clearing time was 151 seconds, whereas the median mean bolus clearing time was 25 seconds. In group 2 patients, all of the reflux parameters were normal. In group 1 the median bolus presence time at each measuring site, the median total bolus transit time, and the median segmental transit time were significantly greater and total propagation velocity lower than values reported in group 2 (P < 0.001), if compared with those described for adult patients. CONCLUSIONS: : The pH-MII is an ideal test in children because it studies GER with its characteristics and motility pattern. Our report summarizes for the first time impedance motility parameters in healthy children

    Quantifying the effects of commercial clam aquaculture on C and N cycling : an integrated ecosystem approach

The definitive version was published in Estuaries and Coasts 39 (2016): 1746–1761, doi: 10.1007/s12237-016-0106-0.Increased interest in using bivalve cultivation to mitigate eutrophication requires a comprehensive understanding of the net carbon (C) and nitrogen (N) budgets associated with cultivation on an ecosystem scale. This study quantified C and N processes related to clam (Mercenaria mercenaria) aquaculture in a shallow coastal environment (Cherrystone Inlet, VA) where the industry has rapidly increased. Clam physiological rates were compared with basin-wide ecosystem fluxes including primary production, benthic nutrient regeneration, and respiration. Although clam beds occupy only 3% of the ecosystem’s surface area, clams filtered 7-44% of the system’s volume daily, consumed an annual average of 103% of the phytoplankton production, creating a large flux of particulate C and N to the sediments. Annually, N regenerated and C respired by clam and microbial metabolism in clam beds were ~3-fold and ~1.5-fold higher, respectively, than N and C removed through harvest. Due to the short water residence time, the low watershed load, and the close vicinity of clam beds to the mouth of Cherrystone Inlet, cultivated clams are likely subsidized by phytoplankton from the Chesapeake Bay. Consequently, much of the N released by mineralization associated with clam cultivation is ‘new’ N as it would not be present in the system without bivalve facilitation. Macroalgae that are fueled by the enhanced N regeneration from clams represents a eutrophying process resulting from aquaculture.
