    From moths to caterpillars: Ideal conditions for Galleria mellonella rearing for in vivo microbiological studies.

    ABSTRACT Galleria mellonella is a well-accepted insect model for the study of pathogen-host interactions and antimicrobial compounds. The main advantages of this model include the low cost of maintenance, the fast life cycle, the possibility of using a large number of caterpillars and the innate immune system, which is evolutionarily conserved relative to mammals. Because of these advantages, different research groups have been working to implement the rearing of G. mellonella in laboratory conditions. This protocol describes our experience in the rearing of G. mellonella caterpillars for experimental infection models and the influence of different artificial diets on developmental and physiological parameters. Here, we suggest a diet composition that benefits the life cycle of G. mellonella by accelerating the larval phase length and increasing the caterpillar weight. This diet also stimulated the immune system of G. mellonella by increasing the hemolymph volume and hemocyte concentration. In addition, our rearing protocol generated caterpillars that are more resistant to infection by Staphylococcus aureus, Escherichia coli and Candida albicans. A standard G. mellonella rearing protocol is fundamental to minimize external influences on the results, and this simple and easy protocol can support researchers starting to rear G. mellonella

    Aiming at radiopharmaceutical application, 111In3+ complexes of the polyaminocarboxylates TTHA, TTHA-bis(butylamide) and TTHA-bis(glucamide) were investigated. The in vitro stability of 111In(TTHA)3− and 111In(TTHA-bis(butylamide)- was evaluated by measuring the exchange of 111In3+ from the complexes to transferrin and the results were compared with those for 111In(DTPA)2−. We also performed biodistribution studies of the three 111In3+ complexes by gamma-imaging in Wistar rats and by measuring the radioactivity in their organs. TTHA and its derivatives seem to have similar in vivo biodistribution with prevailing renal excretion

    WARP: a WIMP double phase Argon detector

    The WARP programme for dark matter search with a double phase argon detector is presented. In such a detector both excitation and ionization produced by an impinging particle are evaluated by the contemporary measurement of primary scintillation and secondary (proportional) light signal, this latter being produced by extracting and accelerating ionization electrons in the gas phase. The proposed technique, verified on a 2.3 liters prototype, could be used to efficiently discriminate nuclear recoils, induced by WIMP's interactions, and measure their energy spectrum. O conhecimento sobre os principais produtos químicos, as formas de utilização com os devidos cuidados na aplicação e na escolha correta do produto constituem parte das estratégias disponíveis para minimizar os prejuízos causados pelos carrapatos. A situação em termos de disponibilidade de produtos carrapaticidas eficazes para a maioria das populações de carrapatos no Brasil é bastante preocupante, especialmente na região Sudeste. A transferência do conhecimento existente, de forma organizada, pode aglutinar os setores público e privado em torno de uma causa comum: melhorar o controle existente do carrapato no país

    A infestação por carrapatos traz vários prejuízos a pecuária brasileira. Dessa forma, objetivou desenvolver nanossistemas imunomoduladores através de nanopartículas lipossomais para reprogramação metabólica de células específicas do sistema imune bovino combater infestação por carrapatos. Para isso, foi feita a extração de fosfolipídios de macrófagos. Durante o cultivo celular dos macrófagos ocorreram sucessivas contaminações das células não sendo possível ter uma eficiente extração de fosfolipídios. Mais estudos precisam ser realizados para amplificação da obtenção dos fosfolipídios e criação de nanossistemas para imunização de uma quantidade maior de animais

    Expressed sequenced tags profiling of resistant and susceptible Gyr x Holstein cattle infested with the tick Rhipicephalus (Boophilus) microplus.

    Tick resistance in cattle is mainly found in zebu (Bos indicus) animals, although it is also present in some taurine (B. taurus) breeds. In order to characterize functional genes involved in tick resistance/susceptibility in cattle, two cDNA libraries were generated using skin tissues of selected Holstein x Gyr animals. A total of 2700 high-quality reads from both resistant and susceptible cDNA were assembled into 458 sequences (contigs) and 834 singletons, with a mean size of 447.7 nucleotides. Assignment of homologous proteins by BLASTX revealed 790 (61.1%) and 300 (23.2%) hits in resistant and susceptible cDNA, respectively; 121 of these hits matched bovine proteins. A total of 502 (38.9%) unique sequences were found to have no significant homology with known sequences and were classified as novel sequences. In general, the most abundant sequences consisted of those coding for hypothetical proteins whose function had not yet been determined, in addition to ribosomal proteins, binding proteins and structural proteins, such as keratin and collagen. The most abundant protein found was collagen type III alpha, although ribosomal proteins accounted for half of the 40 most frequent hits. In addition, five matches within the top 40 best hits corresponded to immune response proteins. These sequences could be used for future studies on functional genomics of cattle tick resistance as well as for genomic sequencing projects.Projeto/Plano de Ação: 02.10.60.300-01
