    Islands of conformational stability for Filopodia

    Filopodia are long, thin protrusions formed when bundles of fibers grow outwardly from a cell surface while remaining closed in a membrane tube. We study the subtle issue of the mechanical stability of such filopodia and how this depends on the deformation of the membrane that arises when the fiber bundle adopts a helical configuration. We calculate the ground state conformation of such filopodia, taking into account the steric interaction between the membrane and the enclosed semiflexible fiber bundle. For typical filopodia we find that a minimum number of fibers is required for filopodium stability. Our calculation elucidates how experimentally observed filopodia can obviate the classical Euler buckling condition and remain stable up to several tens of . We briefly discuss how experimental observation of the results obtained in this work for the helical-like deformations of enclosing membrane tubes in filopodia could possibly be observed in the acrosomal reactions of the sea cucumber Thyone, and the horseshoe crab Limulus. Any realistic future theories for filopodium stability are likely to rely on an accurate treatment of such steric effects, as analysed in this work

    A large-scale study of a poultry trading network in Bangladesh: implications for control and surveillance of avian influenza viruses

    Since its first report in 2007, avian influenza (AI) has been endemic in Bangladesh. While live poultry marketing is widespread throughout the country and known to influence AI dissemination and persistence, trading patterns have not been described. The aim of this study is to assess poultry trading practices and features of the poultry trading networks which could promote AI spread, and their potential implications for disease control and surveillance. Data on poultry trading practices was collected from 849 poultry traders during a cross-sectional survey in 138 live bird markets (LBMs) across 17 different districts of Bangladesh. The quantity and origins of traded poultry were assessed for each poultry type in surveyed LBMs. The network of contacts between farms and LBMs resulting from commercial movements of live poultry was constructed to assess its connectivity and to identify the key premises influencing it

    Antifouling paint particles in intertidal estuarine sediments from southwest England and their ingestion by the harbour ragworm, Hediste diversicolor

    © 2019 Antifouling paint particles (APPs) of between 500 μm and >2 mm in diameter have been identified in silty, intertidal estuarine sediments through a combination of microscopy and x-ray fluorescence spectrometry. APPs were heterogeneously distributed, with maximal concentrations of 430 particles L −1 (0.2 g L −1 ) near to a facility where boats are regularly maintained and 400 particles L −1 (4.2 g L −1 ) at a location where old boats had been abandoned, with the majority of particles encountered in the finest size fraction retrieved. APPs contained variable concentrations of Cu, Zn, Sn and Pb, with respective maxima of 562,000, 269,000, 9,970 and 126,000 mg kg −1 . These characteristics are attributed to a multitude of contemporary and historic sources of an assortment of formulations and result in significant but heterogeneous metal contamination of local sediments. APPs were also identified in the guts of the deposit-feeding ragworm, Hediste diversicolor, that inhabited sediments impacted by abandoned boats or boating activities. The tissue of H. diversicolor was particularly enriched in Cu where ingested APPs were observed, with a significant correlation between dry weight Cu concentrations in the two media (r = 0.734) presumably reflecting the inability of the animal to regulate this metal. While the toxicity of APPs requires further investigation, there is clearly a need for stricter regulations on antifouling wastes in boatyards and marinas and a requirement to better manage abandoned boats.     Antifouling paint particles (APPs) and associated metals have been identified in sediments around boatyards and marinas globally, but the effects of APPs on benthic organisms are largely unknown. Sub-lethal endpoints were measured following laboratory exposures of the harbour ragworm (Hediste diversicolor) and the common cockle (Cerastoderma edule) to environmentally relevant concentrations of biocidal (‘modern’ and ‘historic’) and biocide-free (‘silicone’) APPs added to clean estuarine sediment. Further, the 5-day median lethal concentrations (LC50) and effects concentrations (EC50) for modern biocidal APPs were calculated. For ragworms, significant decreases in weight (15.7%; p < 0.01) and feeding rate (10.2%; p < 0.05) were observed in the modern biocidal treatment; burrowing behaviour was also reduced by 29% in this treatment, but was not significant. For cockles, the modern biocidal treatment led to 100% mortality of all replicates before endpoints were measured. In cockles, there was elevated levels of metallothionein-like protein (MTLP) in response to both modern and historic biocidal treatments. Ragworms had a higher tolerance to modern APPs (5-day LC50:19.9 APP g L−1; EC50: 14.6 g L−1) compared to cockles (5-day LC50: 2.3 g L−1 and EC50: 1.4 g L−1). The results of this study indicate that modern biocidal APPs, containing high Cu concentrations, have the potential to adversely affect the health of benthic organisms at environmentally relevant concentrations. The findings highlight the need for stricter regulations on the disposal of APP waste originating from boatyards, marinas and abandoned boats

    We assessed predominantly pediatric patients in Vietnam with dengue and other febrile illness 3 months after acute illness. Among dengue patients, 47% reported >1 postacute symptom. Most resolved by 3 months, but alopecia and vision problems often persisted. Our findings provide additional evidence on postacute dengue burden and confirm children are affected

    Adverse childhood experiences and mental health in young adults: a longitudinal survey

    BACKGROUND: Adverse childhood experiences (ACEs) have been consistently linked to psychiatric difficulties in children and adults. However, the long-term effects of ACEs on mental health during the early adult years have been understudied. In addition, many studies are methodologically limited by use of non-representative samples, and few studies have investigated gender and racial differences. The current study relates self-reported lifetime exposure to a range of ACEs in a community sample of high school seniors to three mental health outcomes–depressive symptoms, drug abuse, and antisocial behavior–two years later during the transition to adulthood. METHODS: The study has a two-wave, prospective design. A systematic probability sample of high school seniors (N = 1093) was taken from communities of diverse socioeconomic status. They were interviewed in person in 1998 and over the telephone two years later. Gender and racial differences in ACE prevalence were tested with chi-square tests. Each mental health outcome was regressed on one ACE, controlling for gender, race/ethnicity, and SES to obtain partially standardized regression coefficients. RESULTS: Most ACEs were strongly associated with all three outcomes. The cumulative effect of ACEs was significant and of similar magnitude for all three outcomes. Except for sex abuse/assault, significant gender differences in the effects of single ACEs on depression and drug use were not observed. However, boys who experienced ACEs were more likely to engage in antisocial behavior early in young adulthood than girls who experienced similar ACEs. Where racial/ethnic differences existed, the adverse mental health impact of ACEs on Whites was consistently greater than on Blacks and Hispanics. CONCLUSION: Our sample of young adults from urban, socio-economically disadvantaged communities reported high rates of adverse childhood experiences. The public health impact of childhood adversity is evident in the very strong association between childhood adversity and depressive symptoms, antisocial behavior, and drug use during the early transition to adulthood. These findings, coupled with evidence that the impact of major childhood adversities persists well into adulthood, indicate the critical need for prevention and intervention strategies targeting early adverse experiences and their mental health consequences

    Traumatic fracture of central venous catheter resulting in potential migration of distal fragment: a case report

    We report a surgical retrieval of an indwelling portion of a traumatic rupture of the Central venous catheter following hair cutting by a confused patient secondary to Postoperative cognitive dysfunction. He had a dynamic compression screw for fixation of fractured neck of femur after previously failed surgical procedure. The second procedure was complicated with major blood loss, which required central venous and arterial line insertion for intra-operative and post-operative management. The patient was discharged to the ward following an uneventful stay on intensive care. While on the ward, he decided to trim his hair and in the process he inadvertently cut through the right internal jugular catheter. Complications and management resulting from embolisation of central line are reviewed

    An approach for particle sinking velocity measurements in the 3–400 μm size range and considerations on the effect of temperature on sinking rates

    The flux of organic particles below the mixed layer is one major pathway of carbon from the surface into the deep ocean. The magnitude of this export flux depends on two major processes—remineralization rates and sinking velocities. Here, we present an efficient method to measure sinking velocities of particles in the size range from approximately 3–400 μm by means of video microscopy (FlowCAM®). The method allows rapid measurement and automated analysis of mixed samples and was tested with polystyrene beads, different phytoplankton species, and sediment trap material. Sinking velocities of polystyrene beads were close to theoretical values calculated from Stokes’ Law. Sinking velocities of the investigated phytoplankton species were in reasonable agreement with published literature values and sinking velocities of material collected in sediment trap increased with particle size. Temperature had a strong effect on sinking velocities due to its influence on seawater viscosity and density. An increase in 9 °C led to a measured increase in sinking velocities of ~40 %. According to this temperature effect, an average temperature increase in 2 °C as projected for the sea surface by the end of this century could increase sinking velocities by about 6 % which might have feedbacks on carbon export into the deep ocean
