    Experimental demonstration of the supersonic-subsonic bifurcation in the circular jump: A hydrodynamic white hole

    We provide an experimental demonstration that the circular hydraulic jump represents a hydrodynamic white hole or gravitational fountain (the time-reverse of a black hole) by measuring the angle of the Mach cone created by an object in the "supersonic" inner flow region. We emphasise the general character of this gravitational analogy by showing theoretically that the white hole horizon constitutes a stationary and spatial saddle-node bifurcation within dynamical-systems theory. We also demonstrate that the inner region has a "superluminal" dispersion relation, i.e., that the group velocity of the surface waves increases with frequency, and discuss some possible consequences with respect to the robustness of Hawking radiation. Finally, we point out that our experiment shows a concrete example of a possible "transplanckian distortion" of black/white holes.Comment: 5 pages, 5 figures. New "transplanckian effect" described.     Myelopathy is an increasingly recognized presentation of many antibody-mediated neuroinflammatory disorders. While specific features of certain autoimmune myelopathies such as aquaporin-4 antibody associated neuromyelitis optica spectrum disorder (NMOSD) and myelin oligodendrocyte glycoprotein associated disorder (MOGAD) are well-characterized, other less commonly seen antibody-associated myelopathies are not as well-defined. These include but are not limited to, Hu/ANNA1, anti-glial fibrillary acidic protein (GFAP), anti-CV2/collapsin response mediator protein (CRMP5), and amphiphysin. Here, we review the mentioned more common antibody mediated myelopathies as well those that as less common, followed by a review of differentials that may mimic these disorders

    Breeding for carrot resistance to Alternaria dauci without compromising taste

    Developing carrot varieties highly resistant to Alternaria dauci is a top priority for breeders. Meanwhile, consumers are increasingly demanding as regards final product quality, particularly taste. Bitterness is one of the five common taste sensations, but it is rejected by most carrot consumers. Therefore, providing tools for efficient plant breeding of resistant, low bitter carrots would be helpful. While resistance QTLs (rQTLs) have already been identified for carrot resistance to A. dauci, the genetic control and mapping of the metabolites involved in bitterness perception have not been addressed so far. We identified the metabolites most involved in bitterness by combining chemical and sensory analyses of a set of resistant and susceptible carrot genotypes grown in different environments. We evaluated their genetic control and heritability in a segregating F2:3 population over 2 years of field trials and searched for colocalizations between rQTLs and metabolite QTLs (mQTLs) to evaluate the link between bitterness and resistance traits. Our results suggest that it is possible to increase resistance while favoring low bitter varieties by selecting genomic regions involved in the expression of one or the other trait and counter-selecting others when r- and mQTL colocalization is unfavorable

    ENMTools 1.0: an R package for comparative ecological biogeography

    The ENMTools software package was introduced in 2008 as a platform for making measurements on environmental niche models (ENMs, frequently referred to as species distribution models or SDMs), and for using those measurements in the context of newly developed Monte Carlo tests to evaluate hypotheses regarding niche evolution. Additional functionality was later added for model selection and simulation from ENMs, and the software package has been quite widely used. ENMTools was initially implemented as a Perl script, which was also compiled into an executable file for various platforms. However, the package had a number of significant limitations; it was only designed to fit models using Maxent, it relied on a specific Perl distribution to function, and its internal structure made it difficult to maintain and expand. Subsequently, the R programming language became the platform of choice for most ENM studies, making ENMTools less usable for many practitioners. Here we introduce a new R version of ENMTools that implements much of the functionality of its predecessor as well as numerous additions that simplify the construction, comparison and evaluation of niche models. These additions include new metrics for model fit, methods of measuring ENM overlap, and methods for testing evolutionary hypotheses. The new version of ENMTools is also designed to work within the expanding universe of R tools for ecological biogeography, and as such includes greatly simplified interfaces for analyses from several other R packages

    Innovation and optimization in autoimmune encephalitis trials: the design and rationale for the Phase 3, randomized study of satralizumab in patients with NMDAR-IgG-antibody-positive or LGI1-IgG-antibody-positive autoimmune encephalitis (CIELO)

    Background: Autoimmune encephalitis (AIE) encompasses a spectrum of rare autoimmune-mediated neurological disorders, which are characterized by brain inflammation and dysfunction. Autoantibodies targeting the N-methyl-d-aspartic acid receptor (NMDAR) and leucine-rich glioma-inactivated 1 (LGI1) are the most common subtypes of antibody-positive AIE. Currently, there are no approved therapies for AIE. Interleukin-6 (IL-6) signaling plays a role in the pathophysiology of AIE. Satralizumab, a humanized, monoclonal recycling antibody that specifically targets the IL-6 receptor and inhibits IL-6 signaling, has demonstrated efficacy and safety in another autoantibody-mediated neuroinflammatory disease, aquaporin-4 immunoglobulin G antibody-positive neuromyelitis optica spectrum disorder, and has the potential to be an evidence-based disease modifying treatment in AIE. Objectives: CIELO will evaluate the efficacy, safety, pharmacodynamics, and pharmacokinetics of satralizumab compared with placebo in patients with NMDAR-immunoglobulin G antibody-positive (IgG+) or LGI1-IgG+ AIE. Study design: CIELO (NCT05503264) is a prospective, Phase 3, randomized, double-blind, multicenter, basket study that will enroll approximately 152 participants with NMDAR-IgG+ or LGI1-IgG+ AIE. Prior to enrollment, participants will have received acute first-line therapy. Part 1 of the study will consist of a 52-week primary treatment period, where participants will receive subcutaneous placebo or satralizumab at Weeks 0, 2, 4, and every 4 weeks thereafter. Participants may continue to receive background immunosuppressive therapy, symptomatic treatment, and rescue therapy throughout the study. Following Part 1, participants can enter an optional extension period (Part 2) to continue the randomized, double-blind study drug, start open-label satralizumab, or stop study treatment and continue with follow-up assessments. Endpoints: The primary efficacy endpoint is the proportion of participants with a ≥1-point improvement in the modified Rankin Scale (mRS) score from study baseline and no use of rescue therapy at Week 24. Secondary efficacy assessments include mRS, Clinical Assessment Scale of Autoimmune Encephalitis (CASE), time to rescue therapy, sustained seizure cessation and no rescue therapy, Montreal Cognitive Assessment, and Rey Auditory Verbal Learning Test (RAVLT) measures. Safety, pharmacokinetics, pharmacodynamics, exploratory efficacy, and biomarker endpoints will be captured. Conclusion: The innovative basket study design of CIELO offers the opportunity to yield prospective, robust evidence, which may contribute to the development of evidence-based treatment recommendations for satralizumab in AIE

    The known unknowns of hydraulic engineering

    Hydraulic engineers and researchers deal with scientific challenges involving turbulent flow motion and its interactions with the surroundings. Turbulent flows are characterised by unpredictable behaviour, and little systematic research has yet been conducted in natural systems. This paper discusses the implications of recent developments in affordable instrumentation previously characterised by intrinsic weaknesses that adversely affect the quality of the signal outputs. A challenging application is the unsteady turbulence field in tidal bores. The interactions between open channel flows and movable boundaries and atmosphere illustrate another aspect of our limited knowledge. Rapid siltation of reservoirs and air entrainment in turbulent free-surface flows are discussed. In both applications, hydraulic engineers require some broad-based expertise.
