The application of naked DNA containing type I interferon (IFN) transgenes is a promising potential therapeutic approach for controlling chronic viral infections. Herein, we detail the application of this approach that has been extensively used to restrain ocular HSV-1 infection, for antagonizing vaginal HSV-2 infection. We show that application of IFN-α1, -α5, and –β transgenes to vaginal mouse lumen 24 hours prior to HSV-2 infection reduces HSV-2 mediated mortality by 2.5 to 3-fold. However, other type I IFN transgenes (IFN- α4, -α5, -α6, and –α9) are non effectual against HSV-2. We further show that the efficacy of IFN-α1 transgene treatment is independent of CD4+ T lymphocytes. However, in mice depleted of CD8+ T lymphocytes, the ability of IFN-α1 transgene treatment to antagonize HSV-2 was lost

Universal emission intermittency in quantum dots, nanorods, and nanowires

Virtually all known fluorophores, including semiconductor nanoparticles, nanorods and nanowires exhibit unexplainable episodes of intermittent emission blinking. A most remarkable feature of the fluorescence intermittency is a universal power law distribution of on- and off-times. For nanoparticles the resulting power law extends over an extraordinarily wide dynamic range: nine orders of magnitude in probability density and five to six orders of magnitude in time. The exponents hover about the ubiquitous value of -3/2. Dark states routinely last for tens of seconds, which are practically forever on quantum mechanical time scales. Despite such infinite states of darkness, the dots miraculously recover and start emitting again. Although the underlying mechanism responsible for this phenomenon remains an enduring mystery and many questions remain, we argue that substantial theoretical progress has been made.Comment: 9 pages, 2 figures, Accepted versio

Re-examining the Unified Theory of Acceptance and Use of Technology (UTAUT): Towards a Revised Theoretical Model

YesBased on a critical review of the Unified Theory of Acceptance and Use of Technology (UTAUT), this study first formalized an alternative theoretical model for explaining the acceptance and use of information system (IS) and information technology (IT) innovations. The revised theoretical model was then empirically examined using a combination of meta-analysis and structural equation modelling (MASEM) techniques. The meta-analysis was based on 1600 observations on 21 relationships coded from 162 prior studies on IS/IT acceptance and use. The SEM analysis showed that attitude: was central to behavioural intentions and usage behaviours, partially mediated the effects of exogenous constructs on behavioural intentions, and had a direct influence on usage behaviours. A number of implications for theory and practice are derived based on the findings

A Fundamental Study on the Effect of Visual Guidance to Inspectors Using Visual Indicator on Defect Detection in Visual Inspection Utilizing Peripheral Vision

Part 7: Industry 4.0 – Collaborative Cyber-physical Production and Human SystemsInternational audienceIn recent years, it is reported that a number of inspectors in the visual inspection process of industrial products utilized not only the central vision but also the peripheral vision. The visual inspection has been realized with both efficiency and accuracy. However, for the inspectors of many manufacturing industries to adopt this method, visual guidance is necessary for visual attention. Therefore, to obtain the fundamental knowledge for training of the visual inspection method, this study considers the effect of visual guidance on the defect detection performed by inspectors using visual indicators. Specifically, the presence or absence of visual guidance, defect location, and defect characteristics (luminance contrast between defect and inspection model and size) are designed as experimental factors. Further, the effect on inspection accuracy is evaluated with six subjects. As a result, it is clarified that the presence or absence of visual guidance does not affect the defect detection regardless of the defect location and defect characteristics. That is, the visual guidance is not an impediment to inspection accuracy; it is suggested that the visual guidance is an effective tool for the education and training of the visual inspection method utilizing peripheral vision
