    Micropropagation Of Agapanthus Umbellatus Var. Minor By Using Two Systems Of Multiplication

    For conventional micropropagation methods, semisolidified medium (SM) is used; the use of this medium requires intense manipulation of the cultures and skilled labor. Systems that use liquid medium show equal or better efficiency of the multiplication process, besides reducing the cost for the elimination of agar. In this study, we evaluated the mass propagation of Agapanthus umbellatus var. minor two in vitro multiplication systems (SM system and temporary immersion system [SIT]). The plant material was grown in MS medium supplemented with 6-benzylaminopurine (6-BA; 0.0, 8.9, 17.8, and 35.6 Ī¼M). The data obtained in this study demonstrate that the two systems used were efficient for the multiplication phase of this species. However, we recommend SIT in view of its reuse in the process of multiplication and rooting. Moreover, simple construction, low cost of the culture medium, and low cost of the bioreactors and the fact that agar is not required qualify this system as an efficient alternative for large-scale micropropagation of Agapanthus umbellatus var. minor. We recommend 17.8 Ī¼M 6-BA for the SM system and 8.9 Ī¼M 6-BA for SIT.3752923293

    A utilizaĆ§Ć£o de porta-enxertos anƵes, tolerantes a problemas fitossanitĆ”rios da cultura da macieira, como a podridĆ£o do colo (Phytophthora cactorum), pode aumentar o rendimento e reduzir os custos de produĆ§Ć£o. Com o presente trabalho, objetivou-se estabelecer um protocolo de micropropagaĆ§Ć£o do porta-enxerto 'SeleĆ§Ć£o 69', tolerante Ć  podridĆ£o do colo. Ɓpices meristemĆ”ticos foram cultivados in vitro em meio de cultura MS, suplementado com 4,4mM de 6-Benzilaminopurina (BAP), 2,5mM de Ɓcido indolbutĆ­rico (AIB), 0,3mM de Ɓcido giberĆ©lico (GA3), 3% de sacarose e 0,6% de Ć”gar. Foram testados o efeito da concentraĆ§Ć£o de BAP (2,2 e 4,4mM) e o efeito dos meios de cultura (MS, Cheng e Quoirin & Lepoivre) na multiplicaĆ§Ć£o das brotaƧƵes. No alongamento das brotaƧƵes, foram testados os efeitos do GA3 (0; 0,5; 1,0 e 1,5mM) e AIB (0; 0,5 e 1,0mM). Para a fase de enraizamento ex vitro e aclimatizaĆ§Ć£o, foram testadas diferentes concentraƧƵes de AIB (0; 1 e 2g L-1). A taxa de desenvolvimento dos Ć”pices meristemĆ”ticos cultivados in vitro foi de 30%. Para estimular a brotaĆ§Ć£o, a melhor concentraĆ§Ć£o de BAP foi 2,2mM, a qual proporcionou 2,6 brotaƧƵes por explante. O meio MS proporcionou o maior nĆŗmero de brotaƧƵes por explante (2,5), porĆ©m nĆ£o diferiu estatisticamente do meio Cheng (2,0 brotaƧƵes por explante). Para a fase de alongamento, o meio MS suplementado com 2,2mM de BAP e 1,0mM de AIB, proporcionou a maior altura das brotaƧƵes (23mm). Na fase de enraizamento ex vitro e aclimatizaĆ§Ć£o, as concentraƧƵes de 0, 1 e 2g L-1 de AIB possibilitaram em mĆ©dia, 77,3% de enraizamento das microestacas.<br>The use of tolerant apple rootstocks to phytossanitary problems as the rotting (Phytophthora cactorum) is able to increase the production with cost reduction. The objective of the present study is to establish in vitro micropropagation protocol of the '69 Selection' rootstocks tolerant to the rotting. Meristems was cultivated in vitro on MS medium supplemented with 4.4mM of 6-Benzilaminopurine (BAP), 2.5mM of Indol Butiric Acid (IBA), 0.3mM of Giberilic Acid (GA3), 3% of sucrose and 0.6% of agar. The effect of BAP concentration (2.2 and 4.4mM) and the effect of culture media (MS, Cheng and Quoirin & Lepoivre) were tested in the shoot multiplication. In the shoot elongation phase, the effects of GA3 (0; 0.5; 1.0 and 1.5mM) and IBA (0; 0.5 and 1.0mM) were tested. During the rooting ex vitro and acclimatization phase, different IBA concentrations (0; 1 and 2g L-1) were tested. The rate of acme meristems development was 30% of the meristems cultivated in vitro. For the shoot number, the best BAP concentration was 2.2mM that provided 2.6 shoots/explant. The MS medium provided the largest shoot number for explant (2.5), however it was not differing of Cheng medium (2.0 shoots/explant). For the shoot elongation phase, the MS medium supplemented 1.0mM of IBA provided the largest height of shoots (23mm). In the ex vitro rooting and acclimatization phase, the 0, 1.0 and 2.0g L-1 of IBA concentrations, were able to achieve on average of 77,3% of microcuttings rooting

    Avaliou-se, sob duas densidades de plantio, um variante somaclonal de porte baixo de bananeira, comparando-o com a variedade NanicĆ£o Jangada que lhe deu origem. Os materiais genĆ©ticos 'NanicĆ£o Jangada'(controle) e o variante somaclonal representado pelas seleƧƵes 224 e 225 de um ensaio anterior, foram avaliados nos espaƧamentos 2,0m X 2,0m (densidade 2500 plantas ha-1) e 3,0m X 2,0m (1666plantas ha-1), na Fazenda de Ensino e Pesquisa da Faculdade de Engenharia - UNESP, Campus de Ilha Solteira-SP. O ensaio foi conduzido em blocos ao acaso com cinco repetiƧƵes, com utilizaĆ§Ć£o de mudas micropropagadas, sob irrigaĆ§Ć£o por gotejamento, no perĆ­odo de dezembro de 1998 a marƧo de 2001, com avaliaĆ§Ć£o dos dois primeiros ciclos de produĆ§Ć£o. Constatou-se efeito da densidade e do ciclo sobre a produĆ§Ć£o estimada de frutos sendo que no cultivo mais denso, a mĆ©dia foi de 81,25 t.ha-1 no primeiro ciclo de produĆ§Ć£o e 67,93 t.ha-1 no segundo ciclo. No cultivo de menor densidade a produĆ§Ć£o estimada no primeiro ciclo foi de 51,35 t.ha-1 e 44,08 t.ha-1 no segundo. As seleƧƵes do variante de porte baixo apresentaram menor altura da planta e mostraram-se relativamente mais precoces e com produĆ§Ć£o semelhante a cv. NanicĆ£o Jangada no primeiro ciclo. No segundo ciclo houve uma queda na produĆ§Ć£o do bananal, com maior intensidade para a seleĆ§Ć£o 225 do variante.<br>A dwarf somaclonal variant of banana was evaluated under two planting densities. It was compared with the cultivar "NanicĆ£o Jangada", the original clone from which the variant derived. The genotypes 'NanicĆ£o Jangada'(control) and the somaclone, represented by selections 224 and 225 from a previous experiment, were evaluated at spaced 2.0 m X 2.0 m (density of 2500 plants ha-1) and 3.0 m X 2.0 m (1666 plants ha-1) at the "Fazenda de Ensino e Pesquisa da Faculdade de Engenharia - UNESP", Campus de Ilha Solteira, SP. The experiment was conducted in randomized complete blocks with 5 replications, using micropropagated plants under irrigation from December 1998 to March 2001, with evaluation of the first two production cycles. Planting density and production cycle significantly affected estimated fruit yield, with the average in the densest cultivation being 81.25 t.ha-1 in the 1st cycle and 67.93 t.ha-1 in the second. In the lowest planting density cultivation, yield was estimated to be 51.35 t.ha-1 in the 1st cycle and 44.08 t.ha-1 in the 2nd. The dwarf somaclone selections were shorter and produced earlier with yield similar to cv. "NanicĆ£o Jangada" in the first cycle. In the second cycle, there was a decrease in yield, especially for the somaclone variant selection
