    Effect of storage conditions on seed germination of eigTyrrhenian endemic vascular plant species of conservation interest

    The conservation of endemic and endangered plant species is of great interest to the scientific and research community. In this frame, seed banks play a crucial role when biodiversity preservation and climate change are considered. The study of seed viability and germination during storage conditions provides basic and useful information to ensure successful ex situ conservation. The aim of this study was to evaluate whether storage time and conditions (i.e., base collection at -25°C and active collection at +5°C) affect seed germination in the long term. For these purposes, eight Tyrrhenian endemic vascular plant species (mostly endangered) with orthodox seeds were studied: Brassica insularis, Centranthus amazonum, Dianthus morisianus, Digitalis purpurea var. gyspergerae, Ferula arrigonii, Helicodiceros muscivorus, Iberis integerrima and Verbascum plantagineum. These species were stored in the Sardinian Germplasm Bank (BG-SAR) at -25°C and at +5°C for a time ranging from 2 to 12 years. Germination tests were carried out following the optimal conditions reported in the literature for each species. The results showed, in general terms, the high seed germination capacity of all species stored at both conditions; regarding the time of seed storage, germination in some tested species (such as B. insularis and C. amazonum) slightly decreased over time. We argued that seed dehydration, low seed moisture content during storage and the use of hermetic glass containers can be considered key factors for long-term conservation of these orthodox seeds. In conclusion, this study showed that the conservation of these endemic species is ensured by seed bank storage, according to the general assumption that seed longevity depends on seed lot quality, on well-sealed storage containers and conditions before and during storage

    Rubinstein-Taybi Syndrome: spectrum of CREBBP mutations in Italian patients

    BACKGROUND: Rubinstein-Taybi Syndrome (RSTS, MIM 180849) is a rare congenital disorder characterized by mental and growth retardation, broad and duplicated distal phalanges of thumbs and halluces, facial dysmorphisms and increased risk of tumors. RSTS is caused by chromosomal rearrangements and point mutations in one copy of the CREB-binding protein gene (CREBBP or CBP) in 16p13.3. To date mutations in CREBBP have been reported in 56.6% of RSTS patients and an average figure of 10% has ascribed to deletions. METHODS: Our study is based on the mutation analysis of CREBBP in 31 Italian RSTS patients using segregation analysis of intragenic microsatellites, BAC FISH and direct sequencing of PCR and RT-PCR fragments. RESULTS: We identified a total of five deletions, two of the entire gene and three, all in a mosaic condition, involving either the 5' or the 3' region. By direct sequencing a total of 14 de novo mutations were identified: 10 truncating (5 frameshift and 5 nonsense), one splice site, and three novel missense mutations. Two of the latter affect the HAT domain, while one maps within the conserved nuclear receptor binding of (aa 1–170) and will probably destroy a Nuclear Localization Signal. Identification of the p.Asn1978Ser in the healthy mother of a patient also carrying a de novo frameshift mutation, questions the pathogenetic significance of the missense change reported as recurrent mutation. Thirteen additional polymorphisms, three as of yet unreported, were also detected. CONCLUSION: A high detection rate (61.3%) of mutations is confirmed by this Italian study which also attests one of the highest microdeletion rate (16%) documented so far

    A diet enriched with Mugil cephalus processed roes modulates tissue lipid profile in healthy rats: a biochemical and chemometric assessment

    The effect of a diet enriched with mullet bottarga on the lipid profile (total lipids, total cholesterol, unsaturated fatty acids, α-tocopherol, and hydroperoxides) of plasma, liver, kidney, brain, and perirenal adipose tissues of healthy rats was investigated. Rats fed a 10% bottarga enriched-diet for 5 days showed body weights and tissue total lipid and cholesterol levels similar to those of animals fed control diet. Univariate and multivariate results showed that bottarga enriched-diet modified the fatty acid profile in all tissues, except brain. Significant increases of n-3 PUFA, particularly EPA, were observed together with a 20:4 n-6 decrease in plasma, liver, and kidney. Perirenal adipose tissue showed a fat accumulation that reflected the diet composition. The overall data suggest that mullet bottarga may be considered as a natural bioavailable source of n-3 PUFA and qualify it as a traditional food product with functional properties and a potential functional ingredient for preparation of n-3 PUFA enriched foods

    The salted and semidried mullet (Mugil cephalus) ovary product (bottarga) is proposed as an important source of the long-chain n−3 polyunsaturated fatty acids (PUFA), eicosapentaenoic acid, and docosahexaenoic acid. In this work, we investigated the extent of lipid oxidation and browning of grated bottarga samples during 7 mo of storage at −20 ◦C, 2 to 3 ◦C in the absence of light, and at room temperature in the presence or absence of light. Modifications of the levels of total choline (as index of phospholipid breakdown), total sugars, and free amino acids such as lysine, methionine, and tryptophan (involved in nonenzymatic browning) were also studied at different storage conditions. Storage of bottarga did not significantly affect the n−3 PUFA and cholesterol levels with respect to the control; nevertheless, a significant hydroperoxide increase was observed during 7 mo in bottarga samples at all the storage conditions, while low malondialdeyde levels were measured. Samples placed at room temperature in the absence and in the presence of light showed over time a marked browning process, lipid breakdown, a sensible decrease in the levels of total sugars, tryptophan, and methionine with respect to control and samples stored at −20 ◦C and 2 to 3 ◦C. The resistance against the oxidation of the isolated bottarga lipids was also assessed in dry state at several temperatures (37, 75, and 140 ◦C)
