    Challenging the Paradigms on the Origin, Specification and Development of the Female Germ Line in Placental Mammals

    Most of our understanding on the origin, specification and development of the female germ line in placental mammals comes from studies in the laboratory mouse. The molecular pathway leading to the development and establishment of the female germ line in mouse has erected as the paradigm for placental mammals. It remains, however, largely unexplored whether the well-established mouse regulatory pathway is a common mechanism to other or all placental mammals. Discrete differences in mammals other than mouse reveal the existence of alternative mechanisms that challenge the currently accepted tenets on the origin and establishment of the mammalian female germinal reserve. Here, we will discuss the mouse framework in the light of emerging discrepancies seen in other placental mammals

    Interaction of spin and intrinsic orbital angular momentum of light is observed, as evidenced by length-dependent rotations of both spatial patterns and optical polarization in a cylindrically-symmetric isotropic optical fiber.     Tunable SNAP Microresonators via Internal Ohmic Heating

    We demonstrate a thermally tunable Surface Nanoscale Axial Photonics (SNAP) platform. Stable tuning is achieved by heating a SNAP structure fabricated on the surface of a silica capillary with a metal wire positioned inside. Heating a SNAP microresonator with a uniform wire introduces uniform variation of its effective radius which results in constant shift of its resonance wavelengths. Heating with a nonuniform wire allows local nanoscale variation of the capillary effective radius, which enables differential tuning of the spectrum of SNAP structures as well as creation of temporary SNAP microresonators that exist only when current is applied. As an example, we fabricate two bottle microresonators coupled to each other and demonstrate differential tuning of their resonance wavelengths into and out of degeneracy with precision better than 0.2 pm.     Simple gene transfer technique based on I-SceI meganuclease and cytoplasmic injection in IVF bovine embryos

    Although transgenic methods in mammals are inefficient, an easy and highly efficient transgenesis systemusing I-SceI meganuclease (intron-encoded endonuclease fromS. cerevisiae)was recently described in Xenopus. The method consisted of injection into fertilized eggs of an I-SceI reaction mixture with a plasmid DNA carrying the transgene, flanked by the meganuclease recognition sites (pIS). In the present study, the effects of I-SceI on gene transfer were tested apparently for the first time in mammals, in particular, in cattle. Various conditions were evaluated, including three concentrations of the plasmid pIS Pax6egfp, carrying I-SceI recognition sites flanking egfp under Pax6 promoter and two injection times (before IVM and after IVF) of pIS CAGegfp, carrying I-SceI sites fanking egfp under CAG promoter. In addition, the quantity of transgenewasmeasured using quantitative polymerase chain reaction, and presence of transgene signals was evaluated using fluorescence in situ hybridization analysis. Transgene expression rateswere higher (P< 0.05) for groups treated after IVF (79.1%, 91/115 and 63.0%, 75/ 119) than before IVM (32.6%, 31/95 and 34.7%, 33/95), with and without I-SceI, respectively. Interestingly, injectionwith pIS plus I-SceI after IVF increased frequency (P<0.05) of nonmosaic transgene-expressing embryos (58.3%, 42/72 vs. 29.7%, 25/84) for pIS plus I-SceI and pIS alone. Based on fluorescence in situ hybridization analysis, injectionwith I-SceI increased (P<0.05) the proportion of embryos with transgene signals in all blastomeres compared with pIS alone (44.0%,11/25 vs. 6.9%, 2/29) for pIS plus I-SceI and pIS alone. In addition, transgene copy number was numerically higher for the group treated with pIS plus I-SceI compared with pIS alone.     [Reactive arthritis: advances in diagnosis and treatment]

    Reactive Arthritis (ReA) is an aseptic synovitis developing after a primary infection distant from the joint, mainly localized in the gastrointestinal (Enteroarthritis) or genitourinary tract (Uroarthritis). Because of either the asymmetric joint involvement, the possibility of involvement of the spine and enthesis, and the HLA-B27 association ReA is considered one of the spondylarthropathies. Recently, bacterial components or viable bacteria were found in joints during ReA. For this reason, the limits between ReA itself and infectious arthritis are now less definite. Generally accepted diagnostic and classification criteria are still lacking but the improvement in techniques for detection of bacteria increase the possibility to identify the triggering agents. Several studies have examined the role of antimicrobial drugs in ameliorating the natural course of ReA, with some positive results for Uroarthritis only. However, more conventional treatments based on NSAIDs, sulfasalazine and steroids are effective in many cases

    Entanglement swapping for generation of heralded time-frequency-entangled photon pairs

    Photonic time-frequency entanglement is a promising resource for quantum information processing technologies. We investigate swapping of continuous-variable entanglement in the time-frequency degree of freedom using three-wave mixing in the low-gain regime with the aim of producing heralded biphoton states with high purity and low multi-pair probability. Heralding is achieved by combining one photon from each of two biphoton sources via sum-frequency generation to create a herald photon. We present a realistic model with pulsed pumps, investigate the effects of resolving the frequency of the herald photon, and find that frequency-resolving measurement of the herald photon is necessary to produce high-purity biphotons. We also find a trade-off between the rate of successful entanglement swapping and both the purity and quantified entanglement resource (negativity) of the heralded biphoton state.Comment: 17 pages and 9 figures.
